A method for industrial-scale preparation of carboxypeptidase Y (CPY), which was secreted by Saccharomyces cerevisiae KS58-2D/pCY303 into the culture broth, was developed. Because the purification process consists of a few simple unit operations including only one chromatography step, a higher CPY recovery was achieved than that in the process using disrupted baker's yeast. Approximately 100 g of purified CPY powder was constantly obtained using the final culture broth from a 500-l fermentor.  相似文献

This report describes a method of determining irradiation effectiveness of different neutron spectra in causing radiation effects to fuel cladding and reactor structural materials. It involves the definition of a semi-empirical damage function or cross section using measured data from specimens irradiated in thermal and fast test reactor spectra. The damage function is applied to design problems involving irradiation effects to reactor structural and fuel cladding materials to predict the fluence which would produce a specific change in material properties.  相似文献

The uncertainty analyses have been considered as a relevant topic since WASH-1400 and analysis was performed for identifying the risk measure, e.g. plant- and core-damage frequency or the frequency of a large early release of radioactivity in the probabilistic safety assessment (PSA) or probabilistic risk assessment. There are two main sources of uncertainty such as aleatory uncertainty and epistemic uncertainty (parameter uncertainty, model uncertainty and completeness uncertainty) for risk analysis in PSA or risk-monitor system. A sensitivity analysis is related field to uncertainty, which can provide information of the most effective on those inputs of PSA, which are mostly contributed to the uncertainty.

In this paper, uncertainty analysis (epistemic) has been conducted in the evaluation of dynamic reliability of safety-related subsystem for risk analysis. GO-FLOW methodology has been employed for the procedure of uncertainty analysis alternatively to Fault Tree Analysis and Even Tree because it is success-oriented system-analysis technique and comparatively easy to conduct the reliability analysis of the complex system. The method used sample data from Monte Carlo simulation to quantify uncertainty in terms of appropriate estimates for analysis results. Pressurized water reactor containment spray system has been taken as an example of safety-related subsystem. The results of this paper show that the uncertainty analysis is an important part for the practical evaluation of the system dynamic reliability and makes the reliability prediction more accurate compared with the result without the uncertainty analysis. The GO-FLOW methodology can be employed easily for uncertainty analysis with its advance functions.  相似文献

BACKGROUND: Little is known about human herpesvirus (HHV)-6 infection after liver transplantation. We present our experiences with four cases of HHV-6 infection after liver transplantation from living related donors. METHODS: Peripheral blood was collected from four donor and recipient pairs at the time of transplantation and biweekly from the recipients after transplantation. We attempted to isolate HHV-6 and measure antibody titers to HHV-6 and HHV-7. RESULTS: HHV-6 was isolated from four recipients approximately 2 weeks after transplantation. A significant rise in HHV-6 antibody titers was observed in four recipients at some point in their course, whereas HHV-7 antibody titers were increased in one recipient. Four isolates were variant B. When HHV-6 was isolated, all recipients had an unexplained fever. CONCLUSIONS: HHV-6 variant B infection after pediatric liver transplantation was confirmed. HHV-6 infection occurred approximately 2 weeks after transplantation. Moreover, there appears to be an association between HHV-6 infection and unexplained fever.  相似文献

beta-Fructofuranosidase I (FFase I) formed by Aureobasidium pullulans DSM 2404 was purified. The enzyme had a molecular weight of about 430 kDa, was not affected by various metal ions and showed high transfructosylating activity. BACKGROUND: The purpose of this study was to evaluate the effects of leaf order or crop season on anthocyanins and other chemicals in the anthocyanin‐rich tea cultivar ‘Sunrouge’ (Camellia sinensis x C. taliensis) by using high‐performance liquid chromatography, and to study the effect of ‘Sunrouge’ extract on acetylcholinesterase (AChE) activity in human neuroblastoma SK‐N‐SH cells. RESULTS: The total anthocyanin content was higher in the third (3.09 mg g?1) than in the second (2.24 mg g?1) or first crop season (1.79 mg g?1). The amount of anthocyanins contained in the stem was high (1.61 mg g?1). In the third crop season, the concentrations of delphinidin‐3‐O‐β‐D ‐(6‐(E)‐p‐coumaroyl)galactopyranoside (DCGa), cyanidin‐3‐O‐β‐D ‐(6‐(E)‐p‐coumaroyl)galactopyranoside, delphinidin‐3‐O‐β‐D ‐galactopyranoside, delphinidin‐3‐O‐β‐D ‐(6‐O‐(Z)‐p‐coumaroyl)galactopyranoside, cyanidin‐3‐O‐β‐D ‐galactoside, and delphinidin‐3‐O‐β‐D ‐glucoside were 1.57 mg g?1, 0.52 mg g?1, 0.40 mg g?1, 0.22 mg g?1, 0.14 mg g?1, and 0.11 mg g?1, respectively. DCGa accounted for about 50% of the anthocyanins present. The suppressive effect of ‘Sunrouge’ water extract on AChE activity in human neuroblastoma SK‐N‐SH cells was the strongest among the three tea cultivars (‘Sunrouge’, ‘Yabukita’ and ‘Benifuuki’). CONCLUSION: These results suggested that ‘Sunrouge’ might protect humans from humans from AChE‐related diseases by suppressing AChE activity. To obtain sufficient amounts of anthocyanins, catechins and/or caffeine for a functional food material, ‘Sunrouge’ from the third crop season should be used. Transparent lead lanthanum zirconate titanate (PLZT) ceramics were fabricated from fine powders using an inexpensive two-stage sintering technique. The powders were prepared by hydrolysis from low-cost inorganic precursors. In the two-stage sintering method, uniaxially pressed green pellets were densified to nearly theoretical values in an oxygen gas atmosphere during the first-stage sintering, at 1000°C for 1 h, and then residual, free lead oxide in the pellets was removed by second-stage sintering at 1100°C for 12 h. Transparent ceramic with an average grain size of 1.6 μm and a porosity of 1.3% was obtained. The transparency and dielectric characteristics of the present samples were compared with those of hot-pressed samples: The study of the polarization–field hysteresis loops of the present samples yielded a remanent polarization of 6.8 μC/m2 and a coercive field of 1.6 kV/cm. The low coercive field of PLZT ceramics could potentially reduce the driving voltage of electrooptic devices in many applications.  相似文献
