About 100 S-fatty acyl thiol compounds designed as substrates for pancreatic lipase [EC 3.1.1.3] were synthesized and tested for susceptibility to hydrolysis by hog pancreatic lipase and hog hepatic carboxylic esterase [EC 3.1.1.1] using 5,5'-dithiobis(2-nitrobenzoic acid) as a chromogenic reagent to determine the hydrolytic rates of their S-acyl bonds. In general, the hydrolytic rates of S-acyl bonds by the lipase were fast with thioglycerol type thiol moieties, slow with dithioethyleneglycol type or monothiol type, and negligible with thiopolyol type. As for the acyl moieties, the hydrolysis of the S-acyl bonds was fast with C3-5 acyl groups, followed by C6-8 acyl groups, and the rate decreased as the acyl chain length deviated from these values or branched. On the other hand, the hydrolysis of S-acyl bonds by the esterase occurred with all types of S-acyl esters except for esters of long S-acyl chains. Of all the compounds tested with the lipase, the rate of hydrolysis of S-acyl bond was maximum with 2,3-dimercaptopropan-1-ol tributyroate [I], high with 3-mercaptopropane-1,2-diol tributyroate [II], but negligible with the analogous compound, 1,3-dimercaptopropan-2-ol tributyroate. Compounds [I] and [II] may be practically useful as substrates for lipase assay in human serum samples pretreated with phenylmethylsulfonyl fluoride, a potent inhibitor against both serum arylesterase [EC 3.1.1.2] and hepatic esterase, which attack [I] and [II].  相似文献

A spherical aberration (Cs)-corrected 200 kV TEM was newly developed. The column of the microscope was extended by 25 cm and the inner yoke of the objective lens was modified to insert some parts of the corrector elements. The corrector has two hexapole elements that play a main role in Cs correction and they are placed at a position equivalent to the coma-free point of the objective lens by using two transfer doublet lenses. The Cs correction was successfully carried out by means of the third-order aberration that was generated in the two extended hexapoles. The Cs can be corrected to the desired value and also can be overcompensated in order to produce a negative Cs, as with the corrected Cs of -23 microm shown in this work. The optical system of the corrector does not produce second- and fourth-order aberrations, and can correct residual aberrations up to the third order. All of the corrector elements are computer-controlled and the third-order aberrations are quite stable after they are properly corrected. The resolution of 0.135 nm was experimentally confirmed by the Young's fringe method. Image simulations of a silicon [110] single crystal were made with various Cs and defocus values to demonstrate the effectiveness of arbitral control of Cs.  相似文献

This paper presents a new robot-vision system architecture for real-time moving object localization. The 6-DOF (3 translation and 3 rotation) motion of the objects is detected and tracked accurately in clutter using a model-based approach without information of the objects’ initial positions. An object identification task and an object tracking task are combined under this architecture. The computational time-lag between the two tasks is absorbed by a large amount of frame memory. The tasks are implemented as independent software modules using stereo-vision-based methods which can deal with objects of various shapes with edges, including planar to smooth-curved objects, in cluttered environments. This architecture also leads to failure-recoverable object tracking, because the tracking processes can be automatically recovered, even if the moving objects are lost while tracking. Experimental results obtained with prototype systems demonstrate the effectiveness of the proposed architecture.  相似文献

The problem of reducing the pulsating torque in current source inverter induction motor systems is investigated. Two typical approaches presented in recent publications are analyzed by making clear 1) the correlation among the current harmonics and the torque harmonics, and 2) the conceptual contradiction existing between these two approaches. By synthesizing the analyzed results, fundamental principles for treating the current harmonics and torque harmonics are established. In addition, the method for evaluating the degree of torque ripple is described, along with a proposed criterion for torque ripple content. By applying this criterion to these two approaches, objective estimates of degree of torque pulsations, which were unfeasible so far, are now made possible.  相似文献

To quantify the sampling error of wind speed (W) and the surface air specific humidity (Q a) resulting from Sun-synchronous polar-orbit satellite sampling and the effect of single- and multi-satellite sampling, we compared satellite-simulated data with true daily mean data using buoy data. True daily mean data were obtained by averaging buoy data at all available times over 24 h, while satellite-simulated data were the averages of buoy data sampled at satellite passing times (once or twice each day). The difference between true and satellite-simulated data was defined as the sampling error. The sampling error of the daily mean data of W and Q a depends considerably on the satellite observation time and location. Although the sampling error is fairly reduced if multi-satellite sampling is employed, a noticeable sampling error remains in some cases if a wrong sampling combination is employed. Therefore, multi-satellite data should be carefully used to obtain more accurate global data.  相似文献

Summary Organocobalt poly(arylene-vinylene)s composed of 1,2- or 1,3- linkage at their cyclobutadiene moieties (3 and 4, respectively) were prepared by the Heck reaction of p-divinylbenzene with (cyclobutadiene)cobalt-containing monomers having two aryl bromide moieties (1 and 2, respectively). The number average molecular weights (M n) of the obtained organocobalt poly(arylene-vinylene)s (3 and 4) were 4,100 and 4,000, respectively (GPC, on the basis of polystyrene). UV-vis spectra of 3 and 4 exhibited main π-π* absorption peaks at 362 and 397 nm, respectively, which were ca. 60 nm bathochromically shifted from those of the corresponding monomeric units. Both polymers were found to show thermotropic liquid crystallinity in the range of room temperature to ca. 80 °C by employing differential scanning calorimetry (DSC) and the optical measurement using crossed polarizers. A modified heat treatment has been suggested whereby lower temperature plane-strain fracture toughness (K IC) of 4340 ultrahigh strength steel is dramatically improved in developed strength and Charpy impact energy levels. The modified heat-treated 4340 steel (MHT-4340 steel) consists of a mixed structure of martensite and about 25 vol pct lower bainite which appears in acicular form and partitions prior austenite grains. This is produced through isothermal transformation at 593 K for a short time followed by an oil quench (after austenitizing at 1133 K and subsequent interrupted quenching in a lead bath at 823 K). The mechanical properties obtained at room temperature (293 K) and 193 K have been compared with those achieved using various heat treatments. Significant conclusions are as follows: the MHT-4340 steel compared to the 1133 K directly oil-quenched 4340 steel increased theK IC values by 15 to 20 MPa • m1/2 at increased strength and Charpy impact energy levels regardless of the test temperature examined. At 193 K,K IC values of the MHT-4340 steel were not less than those of the 1473 K directly oil-quenched 4340 steel, in whichK IC values are significantly enhanced at markedly increased strength, ductility, and Charpy impact energy levels. The MHT-4340 steels compared to austempered 4340 steels at 593 K, which have excellent Charpy impact properties, showed superiorK IC values at significant increased strength levels irrespective of test temperatures. The lower temperature improvement inK IC can be attributed to not only the crack-arrest effect by acicular lower bainite but also to the stress-relief effect by the lower bainite just ahead of the current crack.  相似文献

The molecular chain network model for elastic deformation behavior and the reptation theory for viscoelastic deformation behavior are used to derive a constitutive equation for rubber. The new eight-chain-like model contains eight standard models consisting of Langevin springs and dashpot to account for the interaction of chains with their surroundings. Monotonic and cyclic deformation behavior of rubber with relaxation under different strain rates have been examined. The results reveal the roles of the individual springs and dashpot, and the strain rate dependence of materials in the monotonic and cyclic deformation behaviors, particularly softening and hysteresis loss, that is, the Mullins effect, occurring in stress-stretch curves under cyclic deformation processes. The validity of the results is checked through comparison with experimental results. The deformation behaviors of a plane strain rubber unit cell containing carbon-black (CB) under monotonic and cyclic straining are investigated by computational simulation using the proposed constitutive equation and homogenization method. The results reveal the substantial enhancement of the resistance of CB-filled rubber to macroscopic deformation, which is caused by the marked orientation hardening due to the highly localized deformation of rubber. The role of strain rate sensitivity on such characteristic deformation behaviors as increases in the resistance to deformation, hysteresis loss, and the effects of the distribution morphology and the volume fraction of CB on the deformation behavior is clarified. The increases in the volume fraction and in the aggregation of the distribution of CB substantially raise the resistance to deformation and hysteresis loss.  相似文献
