The recently described synthesis of ferric adsorbent paper has made possible the modification of protein kinase assays. The adsorbent contains ferric chelate groups, which are responsible for the binding of phosphopeptide via phosphate group. The selective adsorption of phosphopeptide contra nonphosphorylated peptide allows the use of tritium-labeled peptides and unlabeled ATP as substrates. The binding of the reaction product to the adsorbent was complete and was not affected by the amino acid sequence of the peptide. The conditions required for the separation of the produced phosphopeptide from the initial peptide have been worked out as well. The firmly bound phosphopeptide should be released from the ferric adsorbent paper prior to liquid scintillation counting. Using 0.1 m NH4HCO3 solution (pH 8.0), the elution of phosphopeptides was almost complete. The modified protein kinase assay proposed herein is rapid and allows handling of multiple samples simultaneously. In addition, the ferric paper method avoids the use of 32P-isotope, replacing it with 3H which has lower radiation energy and a much longer half-life.  相似文献

Two types of miniature gamma-ray endoscopic probes developed for insertion into a body cavity to detect invisibly small or deeply located tumors are described. One is a single detector system using a small BGO (Bi4Ge3O12) crystal connected with a fiber optic light guide, and the other is a dual detector system using a pair of single detector probes of the same size combined with a random coincidence technique. The performance of these detectors was tested by using a point source and a hollow cylindrical water phantom that simulated a body cavity. Measured and calculated results indicated that a dual detector system can be used for realistic tumor localization in a body cavity, and a single detector system may be applied only for the detection of tumors in which radiopharmaceuticals that emit gamma rays of energy below about 200 keV are accumulated  相似文献

Construction of 275-kV and 500-kV substations in densely populated areas is thought to be necessary in the future to maintain a stable supply of electricity. The use of gas-insulated transformers is now considered for substations because such transformers are non-flammable and nonexplosive. Since the completion of the 77-kV gas-insulated transformer in 1979, we have continued research to develop gas-insulated transformers applicable to a higher voltage level up to 500 kV. As the results of the research, 275-kV, 300-MVA a liquid cooling-type gas-insulated transformer has been developed.  相似文献

The plane stress problem of forced flexural vibrations of an elastic column supported by an elastic half-space and subjected to a sinusoidally varying force at the free end is investigated analytically, by applying the Timoshenko theory to the column. Numerical results obtained from the Timoshenko theory are compared with those from the Bernoulli-Euler theory, and the effects of column slenderness and foundation stiffness on the column response curves and the resonant frequencies of the system are clarified.  相似文献

Calluses were induced from the stem and the root tissues of horse radish plantlets. Cell aggregate selections were carried out on them to obtain a cell line with higher peroxidase activity, and the isozyme patterns of the peroxidases from the cultured cells and their utility values as a clinical diagnostic reagent were investigated. The repetition of the cell aggregate selection resulted in increases in peroxidase activities in each generation of selection; about eleven fold in the 13th generation of selection from the stem tissues and about nine fold in the 13th generation from the root tissues. One of the cell lines (N5K6-S), originally derived from the stem tissues, after selectin showed a tenfold increase in peroxidase activity within two weeks of culture in a liquid medium. The value as a clinical diagnostic reagent of peroxidases extracted from the N5K6-S cells was estimated to be equivalent to those already commercially available for the same purpose. There were significant differences in the isoenzyme patterns of the peroxidases from root tissues and those from cultured cells. From these results, the possibility of commercial production of peroxidase utilizing plant tissue and cell culture techniques was discussed.  相似文献

OBJECTIVE: Cross-reactivity of anti-Sm autoantibodies with a certain ribosomal protein has been reported previously. The present study was undertaken to identify the anti-Sm-reactive ribosomal protein, and to characterize the cross-reactive epitope. METHODS: Two-dimensional gel electrophoresis followed by immunoblotting was used to identify the ribosomal protein (S10) which was reactive with the Y12 anti-Sm monoclonal antibody (MAb). Human anti-Sm antibodies were also tested for cross-reactivity with the Sm-B/B', Sm-D, and isolated S10 proteins by immunoblotting. Epitope analysis was performed by immunoprecipitation of in vitro-translated products of the recombinant S10 and its various mutants. RESULTS: The Y12 MAb and the affinity-purified human anti-Sm autoantibodies cross-reacted with ribosomal S10 protein. Reactivity of the Y12 MAb with S10 protein was abolished by deletion of 19 amino acids at the carboxyl-terminus of S10, containing the Gly-Arg-Gly sequence motif shared by Sm-B/B' and Sm-D (D1 and D3). Replacements of Arg-158 with Gly and of Arg-158/Arg-160 with Gly/Gly at the carboxyl-terminal 157-Gly-Arg-Gly-Arg-Gly region disrupted the Y12 MAb recognition. CONCLUSION: At least a part of human anti-Sm antibodies and Y12 MAb show cross-reactivity among Sm-B/B', Sm-D, and ribosomal protein S10. The carboxyl-terminal Gly-Arg-Gly region of S10 protein is involved in constructing the cross-reactive epitope. This demonstrates that a common structural feature is shared by the ribosomal protein and the small nuclear RNP proteins.  相似文献

To investigate the behavior of metallic particles in 3-phase gas insulated buses (3-phase GIB), we conducted a test by using a 3-phase 275kV, ac, 60Hz full-scale model. The test results revealed that in a 3-phase electric field, unlike in a single-phase field, metallic particles were trapped on and around the high-voltage conductors and revolved circumferentially. Computer analysis shows that it was closely related to locational and periodic changes in the electric field provided by the 3-phase GIB, and that there were points on the high-voltage conductors in which metallic particles were readily trapped. It also was proved that phenomena peculiar to the 3-phase GIB could be reproduced very well by computer simulation.  相似文献
