Change blindness--our inability to detect large changes in natural scenes when saccades, blinks and other transients interrupt visual input--seems to contradict psychophysical evidence for our exquisite sensitivity to contrast changes. Can the type of effects described as 'change blindness' be observed with simple, multi-element stimuli, amenable to psychophysical analysis? Such stimuli, composed of five mixed contrast elements, elicited a striking increase in contrast increment thresholds compared to those for an isolated element. Cue presentation prior to the stimulus substantially reduced thresholds, as for change blindness with natural scenes. On one hand, explanations for change blindness based on abstract and sketchy representations in short-term visual memory seem inappropriate for this low-level image property of contrast where there is ample evidence for exquisite performance on memory tasks. On the other hand, the highly increased thresholds for mixed contrast elements, and the decreased thresholds when a cue is present, argue against any simple early attentional or sensory explanation for change blindness. Thus, psychophysical results for very simple patterns cannot straightforwardly predict results even for the slightly more complicated patterns studied here.  相似文献

We consider whether the continuum model of hydration optimized to reproduce vacuum-to-water transfer free energies simultaneously describes the hydration free energy contributions to conformational equilibria of the same solutes in water. To this end, transfer and conformational free energies of idealized hydrophobic and amphiphilic solutes in water are calculated from explicit water simulations and compared to continuum model predictions. As benchmark hydrophobic solutes, we examine the hydration of linear alkanes from methane through hexane. Amphiphilic solutes were created by adding a charge of +/-1e to a terminal methyl group of butane. We find that phenomenological continuum parameters fit to transfer free energies are significantly different from those fit to conformational free energies of our model solutes. This difference is attributed to continuum model parameters that depend on solute conformation in water, and leads to effective values for the free energy/surface area coefficient and Born radii that best describe conformational equilibrium. In light of these results, we believe that continuum models of hydration optimized to fit transfer free energies do not accurately capture the balance between hydrophobic and electrostatic contributions that determines the solute conformational state in aqueous solution.  相似文献

Autocrine EGF-receptor (EGFR) ligands are normally made as membrane-anchored precursors that are proteolytically processed to yield mature, soluble peptides. To explore the function of the membrane-anchoring domain of EGF, we expressed artificial EGF genes either with or without this structure in human mammary epithelial cells (HMEC). These cells require activation of the EGFR for cell proliferation. We found that HMEC expressing high levels of membrane- anchored EGF grew at a maximal rate that was not increased by exogenous EGF, but could be inhibited by anti-EGFR antibodies. In contrast, when cells expressed EGF lacking the membrane-anchoring domain (sEGF), their proliferation rate, growth at clonal densities, and receptor substrate phosphorylation were not affected by anti-EGFR antibodies. The sEGF was found to be colocalized with the EGFR within small cytoplasmic vesicles. It thus appears that removal of the membrane-anchoring domain converts autocrine to intracrine signaling. Significantly, sEGF inhibited the organization of HMEC on Matrigel, suggesting that spatial restriction of EGF access to its receptor is necessary for organization. Our results indicate that an important role of the membrane-anchoring domain of EGFR ligands is to restrict the cellular compartments in which the receptor is activated.  相似文献

Recent data indicate that the cell surface glycoprotein CD5 functions as a negative regulator of T cell receptor (TCR)-mediated signaling. In this study, we examined the regulation of CD5 surface expression during normal thymocyte ontogeny and in mice with developmental and/or signal transduction defects. The results demonstrate that low level expression of CD5 on CD4(-)CD8(-) (double negative, DN) thymocytes is independent of TCR gene rearrangement; however, induction of CD5 surface expression on DN thymocytes requires engagement of the pre-TCR and is dependent upon the activity of p56(lck). At the CD4(+)CD8(+) (double positive, DP) stage, intermediate CD5 levels are maintained by low affinity TCR-major histocompatibility complex (MHC) interactions, and CD5 surface expression is proportional to both the surface level and signaling capacity of the TCR. High-level expression of CD5 on DP and CD4(+) or CD8(+) (single positive, SP) thymocytes is induced by engagement of the alpha/beta-TCR by (positively or negatively) selecting ligands. Significantly, CD5 surface expression on mature SP thymocytes and T cells was found to directly parallel the avidity or signaling intensity of the positively selecting TCR-MHC-ligand interaction. Taken together, these observations suggest that the developmental regulation of CD5 in response to TCR signaling and TCR avidity represents a mechanism for fine tuning of the TCR signaling response.  相似文献

We report a 51-year-old man with severe ischemic cardiomyopathy and heart failure in whom incessant bigeminal ventricular ectopy failed to generate a detectable arterial pressure. This created a mechanical bradycardia despite an adequate electrical heart rate. Dual chamber pacing increased the effective heart rate and allowed discontinuation of an intraaortic balloon pump from which the patient could not otherwise be weaned.  相似文献

The production of organic precursors to life depends critically on the form of the reactants. In particular, an environment dominated by N2 is far less efficient in synthesizing nitrogen-bearing organics than a reducing environment rich in ammonia. Relatively reducing lithospheric conditions on the early Earth have been presumed to favour the generation of an ammonia-rich atmosphere, but this hypothesis has not been studied experimentally. Here we demonstrate mineral-catalysed reduction of N2, NO2- and NO3- to ammonia at temperatures between 300 and 800 degrees C and pressures of 0.1-0.4 GPa-conditions typical of crustal and oceanic hydrothermal systems. We also show that only N2 is stable above 800 degrees C, thus precluding significant atmospheric ammonia formation during hot accretion. We conclude that mineral-catalysed N2 reduction might have provided a significant source of ammonia to the Hadean ocean. These results also suggest that, whereas nitrogen in the Earth's early atmosphere was present predominantly as N2, exchange with oceanic, hydrothermally derived ammonia could have provided a significant amount of the atmospheric ammonia necessary to resolve the early-faint-Sun paradox.  相似文献

A case of very rare uterine clear cell epithelioid leiomyomatosis is reported. The patient presented with a recurrent pelvic mass after hysterectomy 2 years earlier. The recurrent tumor was located mainly in the vessels of the broad ligament as worm-like plugs. The reviewed hysterectomy specimen showed a typical multi-lobulated intramural mass together with worm-like plugs within the myometrial vessels. Computer tomography exhibited metastatic nodules in both lungs. According to an histological examination, all the tumor cells were of epithelioid type and contained abundant clear cytoplasm. Characteristic large, thick-walled blood vessels were observed. Immunohistochemical staining and ultrastructural examination supported smooth-muscle origin in this case. Electron microscopic study revealed that the clear cytoplasm was attributed to the presence of numerous dilated mitochondria that had lost their cristae. The patient is still alive with the disease 28 months after surgery.  相似文献

The influence of a solution treatment on the evolution of through‐thickness texture gradients in a low carbon steel was studied. In order to intensify through‐thickness variations of the strain state, the samples were cold rolled dry, i.e. without lubrication. The evolution of the texture and the microstructure during rolling and recrystallization was tracked by X‐ray texture analysis and TEM microstructure investigations. The strain distribution during rolling was simulated for several layers of the rolled sheets by the finite element method. After the rolling deformation, a specimen taken directly from the hot band showed pronounced through‐thickness texture gradients, whereas the solution treated specimen showed a rather uniform rolling texture in all layers analyzed. Analysis of the friction behaviour between the different specimens and the roll material points to a much higher friction between the softer hot band and the rolls. This leads to stronger through‐thickness variations in the strain state and, thus, to the pronounced texture gradients observed. The texture gradients in the recrystallized samples reflected those of the as‐rolled sheets. In the center layer of the hot band specimen and in all layers of the solution treated specimen characteristic though notably weakened typical plane strain recrystallization textures formed; the textures in outer layers of the hot band sample were even almost random. These weak recrystallization textures could be attributed to the highly inhomogeneous as‐deformed microstructure.  相似文献

运用非线性有限元程序LS-DYNA对轴压下纵向开槽方形管的耗能能力进行研究。通过冲压作用开槽,在随后的碰撞试验中考虑了冲压作用中产生的有效塑性应变和厚度变化的分布。从模拟结果中可知:当在边墙中开槽时,一般钢管的耗能率将升高82·7%,钢管最大受力减少22·3%。对部分参数的影响进行了分析,包括钢管厚度、开槽长度和开槽数量,并且描述了开槽钢管变形模式的特征。研究发现:开槽可以有效提高薄壁结构的防撞性。  相似文献
