Klebsiella pneumoniae 40bXX, a mutant strain that constitutively produces D-arabinose isomerase (D-AI), was isolated through a series of repeated subcultures from the parent strain on a mineral salt medium supplemented with L-Xylose as the sole carbon source. D-AI could be efficiently immobilized on chitopearl beads. The optimum temperature for the activity of the immobilized enzyme was 40 degrees C and the enzyme was stable up to 50 degrees C. The D-Al was active at pH 10.0 and was stable in the range of pH 6.0-11.0. The enzyme required manganese ions for maximum activity. Three immobilized enzymes, D-xylose isomerase (D-XI), D-tagatose 3-epimerase (D-TE and D-AI were used for the preparation of D-arabinose from D-xylose in a coupling reaction. After completion of the reaction, degradation of D-xylulose was carried out by Saccharomyces cerevisiae. The reaction mixture containing D-Xylose, D-ribulose and the product was then separated by ion exchange column chromatography. After crystallization, the product was checked by HPLC, IR spectroscopy, NMR spectroscopy and optical rotation measurements. Pomegranate peel was used in juicing to find out its effects on the juice products’ (storable juice and wine) sensory property, polyphenols composition, and antioxidant ability. Macroporous resin was used to purify the polyphenols, and 6 different in vitro assays were used to comprehensively determine the antioxidant activity of each. The results showed that juicing with peel made the juice bitter and astringent, but contributed better sensory quality to wine. Peel contributed higher total polyphenols and flavonoids, but lower anthocyanins to the juice products, and caused the phenolics content to fluctuate more dramatically during making wine than the storable juice. Polyphenols purified from the juice products containing peel showed higher total reducing ability and 2,2‐diphenyl‐1‐picrylhydrazyl and 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid radical elimination abilities, but their clearance activity of hydroxyl radicals was not positive, and their superoxide anion radical elimination ability showed no significant difference when compared to polyphenols purified from juice products without peel. 相似文献

Bioactive polyphenols, cartenoids, and anthocyanins present in fruits and vegetables are receiving much attention because of their potential antioxidant activity. This study was conducted to determine antioxidant activity of leaves, peels, stem bark, and kernel of mango varieties langra and chonsa. Total phenolic (TPC) and total flavonoid contents (TFCs) in segments of langra ranged from 63.89 to 116.80 mg GAE/g DW and 45.56 to 90.89 mg CE/g DW, respectively, and that of chonsa were 69.24 to 122.60 mg GAE/g DW and 48.43 to 92.55 mg CE/g DW, respectively. The 2, 2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and linoleic inhibition capacity in segments of langra ranged from 53.30% to 61.10% and 40.0% to 47.20%, respectively, whereas for chonsa; 56.40% to 66.0% and 48.1% to 49.0%, respectively. The reducing potentials of different segments of langra and chonsa at concentration of 10 mg/mL were 0.512 to 0.850 and 0.595 to 0.665 mV, respectively. Comparison between both varieties showed chonsa exhibited better antioxidant activity. An active catalyst, MnO(2)/Al(2)O(3), was prepared by a novel technique as well as wet impregnation method. Total oxidation of chlorinated aromatics on supported manganese oxide catalysts was investigated. It is seen that MnO(2) supported on Al(2)O(3) is an effective catalyst for the oxidative destruction of o-DCB in the concentration range of 1611-2263ppm at temperatures over 350 degrees C. It is also seen that for the catalyst with 0.029% MnO(2), the activity per unit amount of MnO(2) is much higher than that for the other catalysts and the activity per unit amount of MnO(2) decreases sharply for MnO(2)-loading above 0.5%. This high activity may be attributed to a high degree of dispersion of MnO(2) achieved by deposition from colloidal solution. No chlorinated byproducts are detected in the effluent during the oxidative destruction of o-DCB. It is proposed that catalyst with MnO(2)-loading less than 0.5% MnO(2) can be effective for the oxidative degradation of o-DCB. Such catalysts will be less difficult to dispose of after use in view of their low heavy-metal content, and consequently, less harmful to the environment. The possibility of supporting MnO(2) on clay materials may be explored and very cheap catalysts are likely to be obtained by depositing MnO(2) from colloidal solutions. 相似文献

The present study relates to enhancing the dosing efficiency of pharmaceutical dry powder formulations administered by pulmonary inhalation. In particular, the study relates to the provision of dry powder inhalers (DPI) by forming nanosized particles of salbutamol sulfate (SBM) in order to augment the drug penetrability and deposition in the lungs. SBM, an antiasthmatic was selected to be developed into a nanosized formulation by different techniques like solvation, high-pressure homogenization, and spray drying, which were then compared on the basis of particle shape, particle size, and particle size distribution. In case of solvation method the nanosuspension was prepared by dispersing SBM into a nonsolvent and adding Tween-80 as a surfactant to prevent the agglomeration, the particles obtained therein were in the range of 2-10 mu. The second attempt was made by passing the suspension of SBM through high-pressure homogenizer at 10,000-15,000 psi. A treatment of six cycles of homogenization in presence of a Tween-80 as surfactant was found to give a nanosuspension within a size range 50-100 nm. The only drawback seemed with this technique was the low-product yield and high-processing time (3-4 h). In order to overcome this drawback spray-drying technique was further explored; the solution of SBM containing Tween-80 was stirred on magnetic stirrer at 1,200 rpm and finally dried by using spray dryer at an inlet and outlet temperature of 75 degrees C and 56 degrees C, respectively. The feed rate for spray dryer was kept to be 91 ml/h. The sample was collected and analyzed for particle size distribution which was found to be in the range of 50-100 nm. Keeping in view the positive outcomes in terms of higher yield and lower processing time, the spray-drying technique was taken to give the optimized formulation. Nanosized particles, thus obtained were evaluated for particle size, surface topology and particles size distribution, by Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM), and Quasi-elastic light scattering (QELS) technique, respectively. The nanosized particles were subjected to investigate changes on the physical stability of the powder, for this different analytical method was used as: Fourier transform infrared spectroscopy (FT-IR), Differential Scanning Calorimetry (DSC) and X-ray diffraction (XRD) analysis and thus the result indicates that there was no physical disparity when compared with the commercial SBM sample. 相似文献

The quality assurance and authentication of crude herbal drugs play important role in the effective therapeutic effect of herbal drug and their products. There are many reported problems in quality assurance of herbal crude drugs concerning to their correct identification. The present study was designed with the aim to document the authentication and quality assurance of the herbal crude drugs (Argyrolobium roseum and Viola stocksii) thorough light microscopy (LM) and scanning electron microscopy (SEM). The detailed foliar anatomical studies showed polygonal epidermal cells having anticlinal walls in Argyrolobium roseum while rounded epidermal cells were observed in Viola stocksii. The anomocytic stomata type was observed in Argrolobium roseum while actinocytic was noticed in Viola stocksii. The pollen of studied species appeared as tricolporate showing reticulate exine sculpturing in Argrolobium roseum while fine perforations were recorded in Viola stocksii. Furthermore, quantitative analysis of phenols, flavonoids, and antioxidant activity showed high flavonoid and phenol content in Argyrolobium roseum as compared with Viola stocksii. It was observed that Argyrolobium roseum was discriminated from the Viola stocksii based on the leaf and pollen micromorphological traits by using LM and SEM techniques. It was concluded that LM and SEM techniques were found useful for the quality assurance of botanicals and their authentication. 相似文献

Two 20-membered penta- and hexadentate macrocycles containing one or two pyridine subunits, namely [20]aneNO2S2 (L1) and [20]aneN2O2S2 (L2), have been synthesized. Reaction of L1 with silver(I) nitrate afforded a unique 2:2 (M:L) disilver(I) complex [μ2-Ag2(L1)2](NO3)2 (1) in which two four-coordinate Ag atoms bridge two ligands to form a cyclic dimer. In this study, plants belonging to family Solanaceae growing in Western Himalaya region have been observed palynologically under Light Microscope and Scanning electron microscope. Present investigation comprises of 10 genera and 23 species, namely, Atropa acuminata, Capsicum decoraticus, Capsicum frutescens, Cestrum aurantiacum, Cestrum diurnum, Cestrum nocturnum, Datura alba, Datura innoxia, Datura stramonium, Hyoscymus niger, Lycopersicon esculentum, Nicotiana rustica, Nicotiana tabacum, Petunia alba, Petunia hybrida, Solanum erianthum, Solanum melongena, Solanum miniatum, Solanum pseudocapsicum, Solanum surratense, Solanum tuberosum, Withania coagulans, Withania somnifera. Solanaceae is a eurypalynous family. Grains are usually Tricolporate and Tetracolporate, radially symmetrical, isopolar, prolate‐spheroidal to oblate‐spheroidal to oblate‐spheroidal to subprolate to per prolate or suboblate to oblate, size range: 8.55–72 μm, amb circular, semi‐angular or subangular, aperture drop‐type, labrum common‐type, exine usually 2 μm thick, nexine 1–1.5 μm thick. Tectum usually psilate, sexine reticulate, granulate or striato‐reticulate, with obscure pattern, sexine 1–2 μm thick, nexine 1–1.5 μm thick, and intine 0.5–1 μm thick. Most striking variation has been found in the shape class, aperture‐type, and tectal surface. Based on these characters, taxonomic keys have been made for correct identification of members in Solanaceae. However, the grains of this family are usually tricolporate and have direct relationship with certain members of the family Scrophulariaceae. Palyno‐morphological characters of family Solanaceae have been studied for the first time in Western Himalayan region of Pakistan. The medicinal plants are utilized globally considering the cheap and chemical free source, but their correct identification and authentication is prerequisite for safety and efficacy of plant‐based medicines. The present study encompassed traded medicinal plants (16) with high therapeutic value from diverse families like Brassicaceae, Berberidaceae, Malvaceae, Salicaceae, Myrtaceae, Papilionaceae, Ascelpiadaceae, Colchicaceae, Violaceae, and Vitaceae for detailed microscopic study of characters that is, morphology, pollen shape and sizes, P/E ratio, pore length and width, spine length, colpi dimensions, and exine sculpture pattern. The plants showed noteworthy differences in microscopy of Wattakaka volubilis having pollinia, translator and corpusculum like structures while pores were visible in Colchicum luteum, Alcea rosea, and Hibiscus syriacus. The spines were observed in Centipeda minima, A. rosea, and H. syriacus being dimorphic spines in A. rosea and monomorphic in H. syriacus. The exine sculpturing pattern was reticulate in mostly studied plants however distinctive exine pattern was noted in Berberis aristata and Berberis lyceum. The highest polar diameter, equatorial diameter and exine thickness among studied plants were observed in H. syriacus (161 μm), C. luteum (50 μm) and Vitis jacquemontii (1.10), respectively. Thus, microscopy of medicinal plants in addition to other taxonomic evidence offers a supportive skill in authentication, consequently utilization by local consumers and pharmaceutical industries. 相似文献

The research was performed to investigate pollen morphology of endangered species of Capparidaceae in subtropical regions of Pakistan. The distinguishing characters were investigated by using light microscope and scanning electron microscope. Palynological study is comprised of pollen shape, pollen type, exine sculpturing, polar and equatorial diameter, length and width of colpi, mesocolpium, and exine thickness. In polar view, Cleome viscosa exhibited the highest pollen size 26.4 (32.7–24.5 μm) ±0.776 whereas Capparis spinosa appeared to be the lowest 12.6 (14.5?10.7 μm) ±0.400. In equatorial view, Cleome viscosa had the largest pollen size 17.1 (20.0–15.0 μm) ±0.606 and Capparis spinosa had the smallest pollen size 9.7 (12.50–8.00 μm) ±0.394. The maximum fertility percentage has been observed in Capparis spinosa, that is, 98.96% and minimum in Cleome viscosa, that is, 82.93%. Diagnostic key has been constructed to state the essential diagnostic features by means of which the taxa can be identified. Remarkable variations have been observed in pollen size, shape, and exine sculpturing. All the selected species were tricolporate. Prolate to subprolate pollen were observed. There is a great variation existed in exine sculpturing such as in Capparis decidua and C. sp. nova sculpturing is reticulate, in Capparis himalayensis sculpturing is Scabrate granulate, in Capparis spinosa sculpturing is Psilate, in Cleome viscosa sculpturing is regulate‐reticulate, in Dipterygium glaucum sculpturing is regulate and in Gynandropsis gynandra sculpturing is striate‐regulate. On the basis of overall characteristics of pollen it seems that palynology of this family is helpful at the generic and specific level. 相似文献
