To protect bone marrow cells from the toxicity of chemotherapy, a multidrug resistant gene or a dihydrofolate reductase gene has been introduced into stem cells. These genes, however, are not capable of conferring refractoriness to alkylating agents (AA), which are some of the most commonly used agents in chemotherapy regimens. In the present study, an attempt was made to endow human stem cell (CD34+ cells) with resistance to cyclophosphamide, a well-known AA, and adriamycin (ADM) by transducing the glutathione-S-transferase pi (GST-pi) gene whose product is thought to detoxify AA by conjugating them with glutathione and to remove a toxic peroxide formed by ADM. The gene transduction was carried out retrovirally with a virus titer of 1 x 10(5) FFU/ml, employing a recombinant fibronectin fragment; transduction efficiency was extremely low without the fragment. Incubation with interleukin-6 and stem cell factor enhanced the expression of fibronectin ligands VLA4 and VLA5 on CD34+ cells. This enhanced expression of VLA4 and VLA5 was considered to facilitate a close contact of the CD34+ cell to the retroviral vector via fibronectin fragments and the subsequent transduction process. The GST-pi gene-transduced CD34+ cells formed almost 3- and 2.5-fold more CFU-GM than neo gene-transduced CD34+ cells in the presence of 2.5 microg/ml of 4-hydroperoxycyclophosphamide (4-HC), an active form of cyclophosphamide, and 30 ng/ml ADM, respectively. The transfectants formed an appreciable number of colonies, even at higher concentrations of these drugs (5.0 microg/ml of 4-HC, 50 ng/ml of ADM) whereas neo gene-transduced or nontransduced CD34+ cells formed no colonies at all, indicating the possibility of selecting out the transfectants by exposing them to these anticancer drugs. Thus, we were able to demonstrate that transduction of the GST-pi gene confers resistance to cyclophosphamide as well as to ADM, and therefore this approach can be applied clinically for high-dose chemotherapy.  相似文献

GTP cyclohydrolase I feedback regulatory protein (GFRP) mediates feedback inhibition of GTP cyclohydrolase I activity by tetrahydrobiopterin and also mediates the stimulatory effect of phenylalanine on the enzyme activity. To characterize the molecular structure of GFRP, we have purified it from rat liver using an efficient step of affinity chromatography and isolated cDNA clones, based on partial amino acid sequences of peptides derived from purified GFRP. Comparison between the amino acid sequence deduced from the cDNA and the N-terminal amino acid sequence of purified GFRP showed that the mature form of GFRP consists of 83 amino acid residues with a calculated Mr of 9,542. The isolated GFRP cDNA was expressed in Escherichia coli as a fusion protein with six consecutive histidine residues at its N terminus. The fusion protein was affinity-purified and digested with thrombin to remove the histidine tag. The resulting recombinant GFRP showed kinetic properties similar to those of GFRP purified from rat liver. Cross-linking experiments using dimethyl suberimidate indicated that GFRP was a pentamer of 52 kDa. Sedimentation equilibrium measurements confirmed the pentameric structure of GFRP by giving an average Mr of 49,734, which is 5 times the calculated molecular weight of the recombinant GFRP polypeptide. Based on the pentameric structure of GFRP, we have proposed a model for the quaternary structure of GFRP and GTP cyclohydrolase I complexes.  相似文献

Photocurrents in Langmuir multimolecular layers of cadmium arachidate mixed with cyanine dyes between metal electrodes were studied. The photocurrent efficiency was found to depend on the applied voltage, the temperature and the distance between the dye molecules and the electrode. All samples exhibited non- ohmic behavior of the photocurrent. It was concluded that the photocurrent was initiated by the thermionic emission of electrons from the excited dyes. The activation energy necessary for the electron emission from the excited dye in the mixed layers was determined to be 0.52 eV. Absorption spectra were measured for layers deposited on an aluminum film and on a glass slide.  相似文献

We report herein the rare case of a 65-year-old man found to have esophageal intramural metastasis from cancer of the gastric cardia. Endoscopic examination initially revealed an infiltrating ulcerative tumor of the gastric cardia involving the esophagogastric junction, as well as a submucosal tumor of the lower esophagus. A total thoracic esophagogastrectomy with lower mediastinal lymphadenectomy was performed, and the resected specimen demonstrated that both the cardia and esophageal tumors were adenocarcinomas with the same cellular differentiation. As lymphatic invasion and metastases to the paracardial and mediastinal lymph nodes were observed, the esophageal submucosal tumor was considered to be an intramural metastasis from the carcinoma of the gastric cardia resulting from extensive lymphatic spread. The patient died of recurrent disease 9 months after the resection. This case report serves to demonstrate that intramural metastasis may be a local indicator of the systemic spread of disease in patients with gastric carcinoma, as it is in esophageal carcinoma.  相似文献

A spot-size converter integrated semiconductor optical amplifier (SSC-SOA) was developed as a gate element in an optical switch matrix for photonic switching applications. By a selective metal-organic vapor phase epitaxy technique, a bulk InGaAsP stripe, including an active region and thickness tapered SSCs, was grown. Wavelength composition of the stripe, strain on the stripe, and electrode coverage above the SSC region were optimized. As a result, gate operation for fiber-to-fiber gain of 0 dB was achieved at a low injection current of 30 mA, and the polarization dependence of the gain was eliminated as well  相似文献

In recent years, magnetic resonance imaging (MRI) systems have been in demand to diagnose moving parts in the human body such as the heart or the blood in angiography, where images should be taken in milliseconds instead of minutes. Gradient power amplifiers, which are small but vital components for advanced MRI, require higher output power capacity as well as faster rise/fall dynamic response characteristics under a variety of specified current reference signals. This paper presents a novel switch‐mode gradient power amplifier using IGBTs which are connected in parallel to a conventional four‐switch full‐bridge power conversion circuit at their inputs/outputs in order to realize a higher power density. To satisfy the design specifications, which require minimized ripple and improved rise/fall dynamic response characteristics of the current in the gradient coil, a unique digital control scheme based on an optimal type 1 servo system is proposed and described in detail. The effectiveness of the above is discussed and evaluated through computer‐aided analysis. This paper describes a transient cooling technology for electronic equipments using phase-change material (PCM). The module is made of low-cost materials, yet it is designed to achieve a reasonably high level of heat transfer performance. Paraffin is used as the PCM. In previous our report, we can estimate the cooling performance of PCM by using a thermal network method, which cannot calculate melted PCM flow. In this paper, we consider the heat transfer phenomena of PCM module more deeply by using computational fluid dynamics (CFD) analysis with an enthalpy porosity method. By using this method, we can calculate phase-change phenomena and flow phenomena of melted PCM with CFD analysis. First, we briefly explain the results of the experiment and the thermal network analysis. Then we describe the details of CFD analysis with the enthalpy porosity method. In this calculation, melted PCM flow and heat absorption of latent heat can be analyzed. Therefore, we can discuss the reason why the thermal network analysis can estimate cooling performance of PCM module without dealing with melted PCM flow. The calculation results showed that natural convective flow of melted PCM affects the cooling performance of the PCM module. In the case where the PCM module is set vertically, high temperature and low temperature locations exist on the substrate. If several devices are cooled with the PCM module, device consuming the most power must be set in the lower part of the PCM module. From these results, we can conclude that no natural convective flow occurs in our experiment due to the shape of the PCM module.  相似文献

A method for simultaneous determination of multiple pesticide residues in agricultural products was developed by using a pretreatment with ultrafiltration, followed by liquid chromatography-tandem mass spectrometry (LC/MS/MS). The pretreatment process (extraction of pesticides from agricultural products with methanol, dilution of the extract with water, and ultrafiltration) gave recoveries in the range of 50-150% for 63 of 83 pesticides spiked at 0.25 microg/ g into 6 agricultural products. The detection limits of pesticides by LC/MS/MS were below 0.0005-0.05 micro/g. This method is useful for screening purposes and for multiresidue analysis of pesticides in agricultural products. Pesticide residues in 50 domestic crops were investigated by this method, and residues of 14 pesticides were detected in 30 crops.  相似文献

Relatively high purity of nitrogen atom encapsulated fullerene (N@C60) has been synthesized by an electron beam superimposed radio frequency (RF) discharge plasma method. Nitrogen species are characterized by an optical emission spectroscopy (OES); and a relationship between optical emission spectra and the purity of N@C60 has been examined. It is observed that the increased amount of nitrogen molecule ions impinging on the sublimated fullerenes enhance the synthesis of N@C60. Here, it is cleared that the efficient synthesis of N@C60 is possible by controlling the parameters of electron beam superimposed RF plasma. As a consequence, comparatively high purity of about 0.08% of N@C60 has been obtained.  相似文献
