We present a beam-shaping technique for two-photon excitation (TPE) fluorescence microscopy. We show that by inserting a properly designed three-ring pupil filter in the illumination beam of the microscope, the effective optical sectioning capacity of such a system improves so that the point spread function gets a quasi-spherical shape. Such an improvement, which allows the acquisition of 3D images with isotropic quality, is obtained at the expense of only a small increase of the overall energy in the axial sidelobes. The performance of this technique is illustrated with a scanning TPE microscopy experiment in which the image of small beads is obtained. We demonstrate an effective narrowing of 12.5% in the axial extent of the point spread function, while keeping the 82% of the spot-fluorescence efficiency.  相似文献

The effect of selection for growth rate on the sensory characteristics of rabbit meat was assessed by comparing a selected and a control population of rabbits. Embryos belonging to generation 7th were frozen, thawed and implanted in does in order to produce the control group. The control group was formed from the offspring of the embryos belonging to the 7th generation. Selected animals belonging to 21st generation (S) were compared with animals of the control group (C), both were contemporary. Forty animals per group were slaughtered at 9-weeks-old. The sensory analysis was carried out on samples of the Longissimus dorsi muscle. The parameters evaluated were: intensity of rabbit flavour (IRF), aniseed odour (AO), aniseed flavour (AF), liver flavour (LF), tenderness (T), juiciness (J), fibrousness (F). A Bayesian analysis was performed. The ratio of the selection and control effects was analysed. There was a difference between the selected and control groups for IRF, AO, AF and LF. Conversely, no differences were found in T, J and F between groups. Selected group had 3% and 23% higher values of IRF and LF, respectively, than the control group. A relevant effect of selection on AO and AF appeared (probability of relevance P(r)=1), with lower values for selected animals. There was a difference between male and female groups for IRF, but this was not relevant. No differences between sexes were found for the rest of the characteristics evaluated. Selection for growth rate did not affect the main sensory characteristics of meat, like T and J but, it had a negative effect on some flavour characteristics.  相似文献

7 young, healthy, male subjects performed exercise on bicycle ergometers in two 20 min periods with an interval of 1 h. The first 10 min of each 20 min period consisted of arm exercise (38--62% of Vo2 max for arm exercise) or leg exercise (58--78% of Vo2 max for leg exercise). During the last 10 min the subjects performed combined arm and leg exercise (71--83% of Vo2 max for this type of exercise). The following variables were measured during each type of exercise: oxygen uptake, heart rate, mean arterial blood pressure, cardiac output, leg blood flow (only during leg exercise and combined exercise), arterio-venous concentration differences for O2 and lactate at the levels of the axillary and the external iliac vessels. Superimposing a sufficiently strenuous arm exercise (oxygen uptake for arm exercise greater than 40% of oxygen uptake for combined exercise) on leg exercise caused a reduction in blood flow and oxygen uptake in the exercising legs with unchanged mean arterial blood pressure. Superimposing leg exercise on arm exercise caused a decrease in mean arterial blood pressure and an increased axillary arterio-venous oxygen difference. These findings indicate that the oxygen supply to one large group of exercising muscles may be limited by vasoconstriction or by a fall in arterial pressure, when another large group of muscles is exercising simultaneously.  相似文献

The fatty acid pattern of muscle and fat tissue of the arctic seal, birds and mammals are presented and compared with the fatty acid pattern of the corresponding tissues of domestic animals normally used as meat sources in western countries. The triglyceride content of muscle samples were also assayed in these animals. A gas chromatography-mass spectrometry system was used for the localization of the position of double bonds in the unsaturated acids using their pyrrolidides. The data indicated that the fat tissue from the seal is the major source of polyenoic acids i. e., tri- and pentaenoic acids in the diet of the arctic hunter. These acids are derived metabolically from linolenic acid. In contrast polyenoic acids i. e. linoleic acid and its derivatives in the non-arctic diet are mainly supplied from muscle tissue of non-ruminant animals as well from the sources of vegetable origin. The fat content of muscle tissue in arctic animals was not lower than that of domestic meat. The data presented are related to recent studies of the apparent low incidence of coronary heart diseases in arctic populations.  相似文献

To evaluate mode of control of prosthetic terminal devices in terms of prehension force, 20 unilateral below-elbow amputees (10 regular voluntary opening hook wearers and 10 regular voluntary closing hook wearers) performed simple tasks requiring grasping and transporting light objects. Both types can be controlled precisely enough to avoid crushing objects such as straws and paper cups. The objective of this research was to evaluate the antimicrobial activity of carbon dioxide extracts of the unicellular biflagellated green alga Dunaliella salina against Escherichia coli, Staphylococcus aureus, Candida albicans, and Aspergillus niger. The effects of different extraction pressures ranging from 185 to 442 bar and extraction temperatures ranging from 9.8 to 45.2 degrees C on the extracts' composition and consequently on their antimicrobial activities were investigated. The extracts were analyzed by gas chromatography-mass spectrometry in order to identify the compounds responsible for the antimicrobial activity detected. Fourteen different volatile compounds and several fatty acids were identified. The highest antimicrobial activity was obtained using 314 bar and 9.8 degrees C. Under these conditions, the presence of an indolic derivative that had never been reported in D. salina was detected in the extract, together with polyunsaturated fatty acids and compounds related to carotene metabolism, such as beta-ionone and neophytadiene, with known antimicrobial activity.  相似文献
