Alkaline phosphatase from human placenta has been chemically immobilized on a hydrophilic cross-flow microfiltration membrane made from poly(vinylidene difluoride) (PVDF) derivatized with 1,1′-carbonyldiimidazole. The physicochemical characterization of the immobilized biocatalyst paid special attention to the irreversibility of the bonding of the enzyme to the support, the effects of pH, temperature and ionic strength on this activity, the existence of limitations of internal and external diffusion for H+, substrate and/or products, and the kinetic behavior (intrinsic and/or effective) of the immobilized enzyme. With respect to enzyme stability, patterns of hysteresis or memory are proposed, to account for a catalytic activity affected by previous experimental events and situations. The intrinsic kinetic behaviour, rate versus substrate concentration in the absence of diffusional restrictions, was analysed graphically and numerically (by non-linear regression and by utilizing the F statistical test for model discrimination), postulating a minimum rational rate equation of 2:2 degree in substrate concentration. In concordance, a mechanistic kinetic scheme for the catalytic enzyme action has been postulated.  相似文献

This article presents an effective analytical model for determining the moisture diffusivities and moisture transfer coefficients for solid objects (namely, infinite slab, infinite cylinder, sphere; and also for irregularly shaped objects, by using a shape factor) subject to drying applications in a medium. The unsteady-state moisture diffusion analysis is used on the basis of two important criteria: 0·1 <Bi < 100 and Bi > 100. The drying coefficients and lag factors were employed. The analytical models are then verified using available experimental data taken from the literature. The results show that the method presented here can be used to determine the moisture diffusion coefficients and moisture transfer coefficients for such solid objects in a simple and accurate manner for a variety of drying applications.  相似文献

Digital cellular systems will have profound implications for the integration of mobile computing and communications allowing true networking on the move. The European-developed GSM system is rapidly becoming the de-facto world standard for mobile communications. Other emerging standards include IS-54 in North America and PDC in Japan, in addition to the CDMA standard (IS-95) in the USA. 2-Aroyl-3-methyl-1 H-1,4-benzothiazine ylids ( 2a – k ) were prepared by alkylation of the corresponding 4H-benzothiazines 1 . The ylids 2 are labile at room temperature; proper analytical and 1H-n.m.r. data were obtained for their picrates. Fine flour from corn and the isolated starch were dextrinized in the presence of acid and irradiated with y-rays. Starch irradiated with high doses of y-rays was highly modified while fine flour was less affected. The irradiated starch was reduced with sodium borohydride and then hydrolysed. Erythritol and other compounds were identified in the hydrolysate. It was concluded that the C2-C3 bonds in the intermediate glucose units are probably broken by irradiation. Such hypothesis was confirmed by periodate oxidation, hydrolysis with α-amylase and ferricyanide number determination of the irradiated starch.  相似文献

In the clinical management of solid tumors, the possibility to successfully couple the regeneration of injured tissues with the elimination of residual tumor cells left after surgery could open doors to new therapeutic strategies. In this work, we present a composite hydrogel–electrospun nanofiber scaffold, showing a modular architecture for the delivery of two pharmaceutics with distinct release profiles, that is potentially suitable for local therapy and post-surgical treatment of solid soft tumors. The composite was obtained by coupling gelatin hydrogels to poly(ethylene oxide)/poly(butylene terephthalate) block copolymer nanofibers. Results of the scaffolds’ characterization, together with the analysis of gelatin and drug release kinetics, displayed the possibility to modulate the device architecture to control the release kinetics of the drugs, also providing evidence of their activity. In vitro analyses were also performed using a human epithelioid sarcoma cell line. Furthermore, publicly available expression datasets were interrogated. Confocal imaging showcased the nontoxicity of these devices in vitro. ELISA assays confirmed a modulation of IL-10 inflammation-related cytokine supporting the role of this device in tissue repair. In silico analysis confirmed the role of IL-10 in solid tumors including 262 patients affected by sarcoma as a negative prognostic marker for overall survival. In conclusion, the developed modular composite device may provide a key-enabling technology for the treatment of soft tissue sarcoma.  相似文献
