Glioblastoma (GBM) is the leading malignant intracranial tumor and is associated with a poor prognosis. Highly purified, activated natural killer (NK) cells, designated as genuine induced NK cells (GiNKs), represent a promising immunotherapy for GBM. We evaluated the anti-tumor effect of GiNKs in association with the programmed death 1(PD-1)/PD-ligand 1 (PD-L1) immune checkpoint pathway. We determined the level of PD-1 expression, a receptor known to down-regulate the immune response against malignancy, on GiNKs. PD-L1 expression on glioma cell lines (GBM-like cell line U87MG, and GBM cell line T98G) was also determined. To evaluate the anti-tumor activity of GiNKs in vivo, we used a xenograft model of subcutaneously implanted U87MG cells in immunocompromised NOG mice. The GiNKs expressed very low levels of PD-1. Although PD-L1 was expressed on U87MG and T98G cells, the expression levels were highly variable. Our xenograft model revealed that the retro-orbital administration of GiNKs and interleukin-2 (IL-2) prolonged the survival of NOG mice bearing subcutaneous U87MG-derived tumors. PD-1 blocking antibodies did not have an additive effect with GiNKs for prolonging survival. GiNKs may represent a promising cell-based immunotherapy for patients with GBM and are minimally affected by the PD-1/PD-L1 immune evasion axis in GBM.  相似文献

The <110>-oriented BaTiO3 ceramics were fabricated using BaCO3 matrix and H1.08Ti1.73O4.nH2O (HTO) template particles, and the mechanism of BaTiO3 phase formation was investigated. The dielectric, ferroelectric, and piezoelectric properties were also investigated. The transformation of the HTO phase into the TiO2 bronze or TiO2 (B) phase was observed at 600°C, where the BaTiO3 nucleation was accompanied by the formation of a Ba2TiO4 phase. The TiO2 phase reacted with the Ba2TiO4 phase at 800°C to give a BaTiO3 phase, whereas its reaction with the BaTiO3 resulted in the formation of BaTi2O5 phase that got decomposed into BaTiO3 and Ba6Ti17O40 phase at sintering temperature ≥1300°C. Sintering with samples’ embedding in BaTiO3 powders prevented the formation of the Ba6Ti17O40 secondary phase. The crystallographic orientation along the <110> direction (F110) was developed by the epitaxial grain growth mechanism. In addition to the contribution of the grain-size increment for enhancing the F110, the preservation of the platelike structure was also found to have a significant impact. The ceramics prepared by the embedded sintering (grain size ≈12.4 µm and F110 = 83%) exhibited the room-temperature dielectric constant of 1708 and piezoelectric strain constant of 445 pm/V, which are higher than those of the BaTiO3 ceramics with randomly oriented grains.  相似文献

0.24Pb(In1/2Nb1/2)O3-0.42Pb(Mg1/3Nb2/3)O3-0.34PbTiO3 transparent ceramics were fabricated by a conventional sintering technique. Through optimization of sintering conditions of calcination and sintering temperatures and time, the obtained ceramics showed high optical transmittance of 53% and 71% at light wavelengths of 1300 and 2000 nm, respectively. The ceramics showed a rhombohedral to tetragonal phase transition at ~120°C and a tetragonal to cubic phase transition at 222°C. These transition temperatures were higher than those of 0.67Pb(Mg1/3Nb2/3)-0.33PbTiO3 ceramics. In addition, the ceramics had a ferroelectric hysteresis loop, a large piezoelectric constant d33 of 407 pC/N, and a planar electromechanical coupling factor kp of 52%. These results suggest that the transparent ceramics may be used as a temperature-stable, linear electro-optic material.  相似文献

AuCl4 ions in sol-gel-derived silica gels (Au/silica gels) were photoreduced by ultraviolet light irradiation at room temperature. In the presence of ethylene glycol in the silica gels, small Au particles were effectively formed by the irradiation. When the storage temperature of the gels was increased in the dark after the irradiation, the surface plas-mon resonance absorption peak of the Au particles was shifted to longer wavelengths, indicating that the Au particles became large. At a storage temperature below 9°C, Au particles could not be observed in the silica gels. The apparent activation energy of Au particle formation was estimated to be 17. 3 kcal·tmol−1.  相似文献

We developed a fluorescence‐quenching‐based assay system to determine the hydrolysis activity of endo‐β‐N‐acetylglucosaminidases (ENGases). The pentasaccharide derivative 1 was labeled with an N‐methylanthraniloyl group as a reporter dye at the non‐reducing end and with a 2,4‐dinitrophenyl group as a quencher molecule at the reducing end. This derivative is hydrolyzed by ENGase, resulting in an increase in fluorescence intensity. Thus, the fluorescence signal is directly proportional to the amount of the tetrasaccharide derivative, hence allowing ENGase activity to be evaluated easily and quantitatively. Using this system, we succeeded in measuring the hydrolysis activities of ENGases and thus the inhibitory activities of known inhibitors. We confirmed that this assay system is suitable for high‐throughput screening for potential inhibitors of human ENGase that might serve as therapeutic agents for the treatment of N‐glycanase 1 (NGLY1) deficiency.  相似文献

Hybridization comprised of an algebraic turbulence model based on the Reynolds average Navier-Stokes (RANS) equations with a monotonically integrated large eddy simulation (MILES) is proposed to simulate transient fluid motion during separation and vortex shedding at high Reynolds numbers. The proposed hybridization utilizes the Baldwin-Lomax model with the Degani-Schiff modification as the RANS model in the near-wall region and a MILES far from the wall. Although the hybridization is assumed to be a MILES with wall modeling, the transition line between the RANS and the MILES modes is determined by the turbulent intensity that is dominated by the large eddies in the grid-scale. This hybrid model is applied to the flows past three different types of airfoils (NACA633-018, NACA631-012 and NACA64A-006) near stall, at a chord Reynolds number of Re = 5.8 × 106. These airfoils are classified as trailing-edge-stall, leading-edge-stall and thin-airfoil-stall airfoils, respectively. The computed results are compared with wind tunnel experiments. The hybrid model successfully demonstrates accurate stall angle and surface pressure distribution predictions near the stall for each type of airfoil. The airfoil simulation results confirmed that the hybrid model provides a better prediction than the RANS model for unsteady turbulent flows with separation and vortex shedding simulations.  相似文献

To accurately recognize human actions in less computational time is one important aspect for practical usage. This paper presents an efficient framework for recognizing actions by a RGB-D camera. The novel action patterns in the framework are extracted via computing position offset of 3D skeletal body joints locally in the temporal extent of video. Action recognition is then performed by assembling these offset vectors using a bag-of-words framework and also by considering the spatial independence of body joints. We conducted extensive experiments on two benchmarking datasets: UCF dataset and MSRC-12 dataset, to demonstrate the effectiveness of the proposed framework. Experimental results suggest that the proposed framework 1) is very fast to extract action patterns and very simple in implementation; and 2) can achieve a comparable or a better performance in recognition accuracy compared with the state-of-the-art approaches.  相似文献

We developed a low cost, user-friendly multimedia delivery system, to provide medical lectures saved as multimedia contents to persons engaged in medicine. This system was created using the RealSystem package with the TCP/IP network. Users can review lectures and medical meeting presentations with video and audio through the Internet, whenever convenient. Each medical source of video and slide has been clearly displayed on a screen. Members of medical associations or medical students can easily review the most interesting parts of these files. This system is being used efficiently in distance learning and aids the diffusion of the latest information and technology to busy physicians and medical students.  相似文献
