Using polymerase chain reaction (PCR) a complementary DNA (cDNA) encoding a 5-hydroxytryptamine (5-HT) receptor was isolated from rat forebrain. The amplified cDNA specifies an open reading frame of 374 amino acids comprising seven putative transmembrane regions. Expression of the cloned cDNA in human embryonic kidney cells (HEK 293) was used to establish the pharmacological profile of the encoded receptor polypeptide. Membranes containing the cloned receptor showed high affinity binding of [3H]-5-HT. Competition binding experiments with a variety of serotonin receptor ligands displayed a rank order of affinities corresponding to a 5-HT1D subtype: 5-CT > 5-HT = metergoline > CGS 12066 > methysergide > sumatriptan > mianserin = (-)alpha-Me-5-HT = yohimbine > 8-OH-DPAT > or = rauwolscine > spiperone > DOI > propranolol > or = 2-Me-5-HT > or = ICS 205930. Ketanserin and ritanserin displaced [3H]-5-HT-binding in a biphasic manner. In situ hybridization revealed highest expression of the corresponding mRNA in the pyramidal layer of the olfactory tubercle and the nucleus caudatus and accumbens.  相似文献

We recently reported that CYP2D16, a xenobiotic-metabolizing P450 isozyme, was expressed at higher levels in adrenal microsomes from inbred Strain 13 guinea pigs than in those from outbred English Short Hair (ESH) animals. Studies were done to determine if there also were strain differences in adrenal microsomal steroid metabolism. In both inner (zona reticularis) and outer (zona fasciculata plus zona glomerulosa) zone preparations of the adrenal cortex, 21-hydroxylase activities were greater in microsomes from ESH than from Strain 13 guinea pigs. By contrast, 17alpha-hydroxylase activities were similar in the two strains. In both strains, 21-hydroxylase activities were greater in inner than outer zone microsomes, but the opposite was found for 17alpha-hydroxylase activities (outer>inner). Northern and Western analyses revealed higher levels of CYP21 mRNA and protein in adrenals from ESH than Strain 13 guinea pigs, but there were no strain differences in CYP17 mRNA or protein concentrations. Despite the zonal differences in adrenal 17alpha-hydroxylase and 21-hydroxylase activities, CYP17 and CYP21 mRNA and protein levels were similar in the inner and outer zones within each strain of guinea pig. The results demonstrate strain differences in microsomal steroid metabolism that are explained by differences in CYP21 expression. By contrast, the zonal differences in steroid hydroxylase activities may be attributable to post-translational mechanisms.  相似文献

The Potential Behaviour of the Interface of Metallic Materials at Mild and Strong Vibratory Cavitation I. Experimental technics In case of moving liquids in metallic structural materials or, vice versa such materials in liquids there can occur very different surface stressing of the materials. These surface stressings can be either of mechanic or of corrosive nature or of both of them. This will be dependent on the force being made effective as well as on the nature of the liquid. The equivalent for these forces is the acceleration with a range from zero up to 100 000 g at sinoidal oscillations of cylindrical specimens in liquids which is taken as basis for the vibratory cavitation, as it is well known. Therewith, the expenditure of equipments, above all with the vibratory equipments has turned out to be more difficult and more expensive than it first has been anticipated. Therefore it has been anticipated. Therefore it has been necessary to provide a number of experimental technical conditions on which we shall report here first. This report will be followed by further experimental results on copper-, titanium-, aluminum as well as iron materials in the above mentioned order of succession.  相似文献

The STRATEGY project (sustainable restoration and long-term management of contaminated rural, urban and industrial ecosystems; www.strategy-ec.org.uk) addressed the need for a holistic decision framework for the selection of optimal remediation strategies for long-term sustainable management of contaminated areas in Western Europe. The project considered both technical and social aspects of implementing restoration strategies for urban and rural environments. The importance of considering socially relevant objectives in addition to the dose reduction was emphasised. A critical evaluation was carried out on 101 selected countermeasures, (including rural waste disposal options), a model was developed to aid optimising countermeasure strategies and a method of carrying out participatory decision-making suggested. The outputs of the project are described and critically evaluated.  相似文献

Many of the operations to eliminate complaints concerning respiration impairments fail. In order to improve the success rate, it is important to recognize the responsiveness of the flow field within the nasal cavities. Therefore, we are developing a computer assisted surgery (CAS) system that combines computational fluid dynamics (CFD) and virtual reality (VR) technology. However, the primary prerequisite for VR-based applications is real-time interaction. A single graphics workstation is not capable of satisfying this condition and of simultaneously calculating flow features employing the huge CFD data set. In this paper, we will present our approach of a distributed system that relieves the load on the graphics workstation and makes use of an off-the-shelf parallel Linux cluster calculating streamlines. Moreover, we introduce first results and discuss remaining difficulties.  相似文献

The purpose of this laboratory study was to evaluate the possible differences in motor strategies to a new standardized low-load repetitive work task in between healthy experienced workers and a reference group. Work task event duration, i.e. working rhythm, cutting forces, surface electromyographic (EMG) activity from four shoulder muscles, postural activity, and arm and trunk movements in 3D were recorded during low-load repetitive work simulation. The experienced group showed lower EMG activity and frequency contents (P<0.05), more abducted position of the upper arm and forward flexion of the trunk prior to work simulation (P<0.05), and increased arm and trunk range of motion (P<0.05) compared with the reference group. The results highlight that experienced butchers have a different motor strategy compared with a reference group, i.e. more variable form of coordination pattern. Furthermore, the initial implementation of a possible protective motor strategy by experienced workers might be a very important prognostic factor.  相似文献

An epoxy‐based negative‐tone photoresist, which is known as a suitable material for high‐aspect‐ratio surface micromachining, is functionalized with red‐light‐emitting CdSe@ZnS nanocrystals (NCs). The proper selection of a common solvent for the NCs and the resist is found to be critical for the efficient incorporation of the NCs in the epoxy matrix. The NC‐modified resist can be patterned by standard UV lithography down to micrometer‐scale resolution, and high‐aspect‐ratio structures have been successfully fabricated on a 100 mm scaled wafer. The “as‐fabricated”, 3D, epoxy‐based surface microstructures show the characteristic luminescent properties of the embedded NCs, as verified by fluorescence microscopy. This issue demonstrates that the NC emission properties can be conveniently conveyed into the polymer matrix without deteriorating the lithographic performance of the latter. The dimensions, the resolution, and the surface morphology of the NC‐modified‐epoxy microstructures exhibit only minor deviations with respect to that of the unmodified reference material, as examined by means of microscopic and metrologic investigations. The proposed approach of the incorporation of emitting and non‐bleachable NCs into a photoresist opens novel routes for surface patterning of integrated microsystems with inherent photonic functionality at the micro‐ and nanometer‐scale for light sensing and emitting applications.  相似文献
