There is consistent evidence that vitamin D deficiency is strongly associated with liver dysfunction, disease severity, and poor prognosis in patients with liver disease. Vitamin D and its receptor (VDR) contribute to the regulation of innate and adaptive immune responses. The presence of genetic variants of vitamin D- and VDR-associated genes has been associated with liver disease progression. In our recent work, we summarized the progress in understanding the molecular mechanisms involved in vitamin D–VDR signaling and discussed the functional significance of VDR signaling in specific cell populations in liver disease. The current review focuses on the complex interaction between immune and liver cells in the maintenance of liver homeostasis and the development of liver injury, the interplay of vitamin D and VDR in the development and outcome of liver disease, the role of vitamin D- and VDR-associated genetic variants in modulating the occurrence and severity of liver disease, and the therapeutic value of vitamin D supplementation in various liver diseases. The association of the vitamin D–VDR complex with liver dysfunction shows great potential for clinical application and supports its use as a prognostic index and diagnostic tool. 相似文献

Sarcopenia is characterised by an age-related decrease in the number of muscle fibres and additional weakening of the remaining fibres, resulting in a reduction in muscle mass and function. Many studies associate poor maternal nutrition during gestation and/or lactation with altered skeletal muscle homeostasis in the offspring and the development of sarcopenia. The aim of this study was to determine whether the musculoskeletal physiology in offspring born to mouse dams fed a low-protein diet during pregnancy was altered and whether any physiological changes could be modulated by the nutritional protein content in early postnatal stages. Thy1-YFP female mice were fed ad libitum on either a normal (20%) or a low-protein (5%) diet. Newborn pups were cross-fostered to different lactating dams (maintained on a 20% or 5% diet) to generate three groups analysed at weaning (21 days): Normal-to-Normal (NN), Normal-to-Low (NL) and Low-to-Normal (LN). Further offspring were maintained ad libitum on the same diet as during lactation until 12 weeks of age, creating another three groups (NNN, NLL, LNN). Mice on a low protein diet postnatally (NL, NLL) exhibited a significant reduction in body and muscle weight persisting up to 12 weeks, unlike mice on a low protein diet only prenatally (LN, LNN). Muscle fibre size was reduced in mice from the NL but not LN group, showing recovery at 12 weeks of age. Muscle force was reduced in NLL mice, concomitant with changes in the NMJ site and changes in atrophy-related and myosin genes. In addition, μCT scans of mouse tibiae at 12 weeks of age revealed changes in bone mass and morphology, resulting in a higher bone mass in the NLL group than the control NNN group. Finally, changes in the expression of miR-133 in the muscle of NLL mice suggest a regulatory role for this microRNA in muscle development in response to postnatal diet changes. Overall, this data shows that a low maternal protein diet and early postnatal life low-protein intake in mice can impact skeletal muscle physiology and function in early life while postnatal low protein diet favours bone integrity in adulthood. 相似文献

Human error can have potentially devastating consequences in contexts such as healthcare, but there is a rarely a simple dichotomy between errors and correct behaviour. Furthermore, there has been little consideration of how the activities of users (erroneous and otherwise) relate to the conceptual fit between user and device, despite the fact that healthcare technologies are becoming increasingly prevalent and complex. In this article, we present a study in which nurses’ conceptions of infusion device practice were elicited to identify misfits. By focusing on key concepts that users work with when setting up infusions and the extent to which the system supports them, our analysis highlights how actions are influenced by the different resources available to users including: the device itself; supporting artefacts; the conceptual understanding of the user; and the community of practice the user is part of. The findings reveal the ways in which users are resourceful in their day-to-day activities and also suggest potential vulnerabilities within the wider system that could threaten patient safety. Our approach is able to make previously under-explored aspects of practice visible, thus enabling insight into how users act and why. 相似文献

The recently developed 3D micro X-ray fluorescence spectroscopy (3D Micro-XRF) enables three-dimensional resolved, nondestructive investigation of elemental distribution in samples in the micrometer regime. Establishing a reliable quantification procedure is the precondition to render this spectroscopic method into a true analytical tool. One prominent field of application is the investigation of stratified material. A procedure for the quantitative reconstruction of the composition of stratified material by means of 3D Micro-XRF is proposed and validated. With the procedure, it is now possible to determine nondestructively the chemical composition and the thickness of layers. As no adequate stratified reference samples were available for validation, stratified reference material has been developed that is appropriate for 3D Micro-XRF or other depth-sensitive X-ray techniques. 相似文献

Recently, social networks have become the most prevalent IT paradigm, as the vast majority of Internet users maintain one or multiple social networking accounts. These accounts, irrespectively of the underlying service, contain rich information and data for the owner’s preferences, social skills, everyday activities, beliefs and interests. Along with these services, the computation, sensing and networking capabilities of the state of the art mobile and portable devices, with their always-on mode, assist users in their everyday lives. Thus, the integration of social networking services with current pervasive computing systems could provide the users with the potential to interact with other users that have similar interests, preferences and expectations; and in general, the same or similar context, for limited or not time periods, in order to ameliorate their overall experience, communicate, socialise and improve their everyday activities with minimal effort. This paper introduces a cross-community context management framework that is suitable for Cooperating Smart Spaces, which couple the advantages of pervasive computing and social networking. This framework goes beyond the state of the art, among others, in that cross-community context from a multitude of sources is collected and processed to enhance the end user experience and increase the perceived value of the services provided. 相似文献

In this work, we used time-resolved imaging to study the dynamics of the laser-induced forward transfer (LIFT) process of a silver nanoparticle (NP) ink (NP size: 30–50 nm). LIFT is a versatile direct write technique in which a variety of functional materials can be transferred from a donor substrate to a receiving substrate with high spatial resolution. Two different LIFT configurations were employed: (a) dynamic release layer (DRL)-assisted LIFT, in which direct expose of the silver NP ink to laser irradiation is prevented, and (b) DRL-free LIFT, in which the silver NP ink is exposed to laser irradiation. Jetting dynamic behavior, initiated by a cavitation bubble generation and expansion, was observed in both LIFT configurations. However, jetting dynamics were significantly milder in the case of DRL-assisted LIFT, resulting in a wide laser fluence processing window (100–230 mJ/cm2) for high uniformity and reproducibility printing of silver NP ink droplets. On the contrary, DRL-free LIFT resulted in smooth jetting dynamics only for a narrow laser fluence window (30–40 mJ/cm2). The explanation of the different dynamics is based on the different mechanisms that govern the conversion of the laser pulse energy to a dynamic cavitation bubble for each LIFT configuration, i.e., (a) heat diffusion, mediated by the DRL layer, in DRL-assisted LIFT and (b) microcavitation around the silver NPs due to near field enhancement when no DRL is used. In addition, the mechanism of formation of undesirable satellite droplets around the main deposited droplets was studied by using a flexible polymeric receiving substrate. The importance of the smooth jetting behavior, achieved by DRL-assisted LIFT, was highlighted for high-resolution printing of silver NP ink as well as for ensuring enhanced LIFT processing stability. 相似文献

The present study comprises the second part of an ongoing study focusing on olive oil from five less well‐known Greek cultivars for three of which there are no data available in the literature regarding their chemical composition. A total of 74 olive oil samples were collected during the harvesting periods 2012–2013 and 2013–2014. Headspace‐solid phase microextraction was applied to determine the olive oil volatile profile. Fifty‐six compounds were identified and semi‐quantified by CG–MS. Furthermore, fatty acid composition, conventional quality parameters and color parameters were determined in an effort to characterize and differentiate olive oils according to cultivar. All samples were characterized as extra virgin olive oils. Data obtained showed significant differences between the cultivars. Multi‐element analysis in combination with chemometrics resulted in a high classification rate of 86.5 % for the combination of volatiles plus color, 89.2 % for the combination of VC plus FA, and 91.9 % for the combination of FA composition plus color plus CQP. 相似文献
