Summary The acid-tolerant and heat-resistant bacterium Alicyclobacillus acidoterrestris is a spoilage problem in pasteurized and heat-treated fruit juices. In this study it was shown to grow in orange juice, grapefruit juice and apple juice to produce detectable taint at levels of about 104 –105 c.f.u. ml−1 . Decimal reduction times were determined at 80 °, 90 ° and 95 °C in each juice and confirmed the heat-resistant nature of the spores under normal juice pasteurization conditions. They also confirmed that heat sensitivity increased with decreasing pH but that this effect was less pronounced at higher temperatures. The organism was, however, sensitive to the bacteriocin food preservative nisin. The presence of nisin during heating decreased the D value by up to 40% and the MIC for nisin against spores at 25 °C was only 5 International Units (IU) ml−1 . The results indicate that use of nisin is a potentially useful way of controlling this organism in fruit juices and fruit juice-containing products. 相似文献

The present work evaluated the possible role of volatile amines as indicator(s) of poultry meat spoilage. Fresh chicken meat (breast fillet) was packaged in four different atmospheres: air (A), vacuum (VP) and two modified atmospheres (MAs), namely M1, 30%/65%/5% (CO2/N2/O2) and M2, 65%/30%/5% (CO2/N2/O2). All chicken samples were kept under refrigeration (4 ± 0.5 °C) for a period of 15 days. Of the four treatments, the VP and M1 and M2 gas mixtures were the most effective for delaying the development of aerobic spoilage microbial flora. Pseudomonas spp. in chicken samples stored under M2 gas mixture and VP were significantly lower than all the other samples after 15 days of storage. Of the remaining bacterial species examined, lactic acid bacteria (LAB), Brochothrix thermosphacta, were dominant in the microbial association of both aerobically- and MA-packaged chicken, while yeasts contributed to a much lesser extent in the final microbial flora of chicken meat. On the basis of microbiological data (TVC), shelf-life extensions of 2, 4 and 9–10 days were achieved by VP and M1 and M2 gas mixtures. Results of the present work showed that the limit of sensory acceptability (a score of 6) was reached for the aerobically, vacuum-packaged and M1 gas mixture chicken samples approximately on days 6–7 and 9–10, respectively. Based on sensory (taste) analysis and with regard to chicken spoilage and freshness, TMA-N and TVB-N limit values of acceptability, namely 10.0 mg N/100 g and 40 mg N/100 g for chicken samples stored in air, may be proposed as the upper limit values for spoilage initiation of fresh chicken meat stored aerobically. Interestingly, the M2 gas mixture sample did not reach these limit values throughout the 15 day storage period. The formation of volatile amines during chill storage of chicken meat, under the packaging conditions examined in the present study, seemed to be in good agreement with the increase in microbiological count (TVC) and sensory taste score except for the M2 gas mixture. 相似文献

The purpose of this study was to investigate the effect of applying two Lactobacillus protective cultures on microbiological, chemical and odour changes during storage of refrigerated vacuum‐packed rainbow trout (Oncorrynchus mykiss) fillets. Lactobacillus sakei CECT 4808 and Lb. curvatus CECT 904T were inoculated individually or in combination into rainbow trout fillets. The samples were vacuum‐packed and stored at 4 ± 0.5 °C and were assessed during a 20‐day storage period for microbiological (Enterobacteriaceae, Pseudomonas spp., lactic acid bacteria (LAB), H2S‐producing bacteria, yeasts and moulds) and chemical (pH, total volatile basic nitrogen, lipid oxidation) parameters and off‐odour. Samples inoculated with Lb. sakei and with both strains had significantly (P≤0.05) lower counts of all microbiological spoilage indicator organisms (Enterobacteriaceae, Pseudomonas spp., H2S‐producing bacteria, yeasts and moulds) than those inoculated with Lb. curvatus or the controls. All chemical parameters examined and off‐odour scores were also significantly (P≤0.05) improved in the samples inoculated with Lb. sakei or with both strains compared with those inoculated with Lb. curvatus or the controls. Lb. sakei generally showed a better preservative effect (P≤0.05) than the combination of strains. Inoculation with Lb. sakei CECT 4808 could provide an additional hurdle to improve shelf‐life of refrigerated vacuum‐packaged trout fillets, as inoculation with this strain resulted in extension of shelf‐life by 5 days. The implementation of novel and accurate quality and safety control methods in conjunction with chemometrics in various fields of science, particularly in food science, showed that this combination stands for a very powerful tool for detecting food authenticity. The latter reflects both geographic origin and variety. Dairy products, in particular, have repeatedly worried the public authorities in terms of authentication regarding origin and in view of the many illnesses occasionally due to products of specific origin. It has been repeatedly claimed that the application of genetic engineering in the field of agricultural and food production is both beneficial and advantageous. However, biotechnology is developing in an environment where public concerns about food safety and environmental protection are steadily increasing. The present study aims at gaining an insight into Greek consumers’ beliefs, attitudes and intentions towards genetically modified (GM) food products. The objectives of this study are summarized as follows (i) to provide evidence that consumer beliefs are built around the ‘safety‐benefits’ axis, and (ii) to segment the Greek market in terms of consumer beliefs about GM food products and identify a number of clusters with clear‐cut behavioural profiles. Although the overall attitude of Greek consumers towards GM food is negative, the research very interestingly concludes that there exists a market segment of substantial size, whose beliefs about GM food appears to be positive. This finding suggests that there is not a ‘consensus’ regarding the rejection of GM foods in the Greek market as one might have expected thus ‘encouraging’ the implementation of adequate marketing strategies to target this segment of ‘early adopters’ in the first place. 相似文献

As genetically modified (GM) foods are starting to intrude in our diet concerns have been expressed regarding GM food safety. These concerns as well as the limitations of the procedures followed in the evaluation of their safety are presented. Animal toxicity studies with certain GM foods have shown that they may toxically affect several organs and systems. The review of these studies should not be conducted separately for each GM food, but according to the effects exerted on certain organs it may help us create a better picture of the possible health effects on human beings. The results of most studies with GM foods indicate that they may cause some common toxic effects such as hepatic, pancreatic, renal, or reproductive effects and may alter the hematological, biochemical, and immunologic parameters. However, many years of research with animals and clinical trials are required for this assessment. The use of recombinant GH or its expression in animals should be re-examined since it has been shown that it increases IGF-1 which may promote cancer. 相似文献

A range of rheological techniques, including small deformation isothermal runs following quenching, controlled rate heating profiles, mechanical spectra, and large deformation strain sweeps and creep compliance testing, were employed in the investigation of maltodextrin—sodium caseinate aqueous dispersions. The concentration dependence, viscoelastic ratio and melting profiles of shear moduli for concentrated maltodextrin samples suggest a highly aggregated, enthalpically linked biopolymer gel. By contrast, the caScinate particles form a ‘pasty’ solid at low temperatures with a high viscous component, which upon moderate heating reverts readily into an entropically driven liquid body. Mixing of the polymers results in a composite system whose phase inversion from a maltodextrin continuous network with discontinuous protein inclusions to a caScinate dispersion suspending the polysaccharide particles is determined by the weight ratio of the two components in the blend. The overall strength of the composite has been related to the mechanical functions of the individual components, and the idea of kinetically trapped mixed gels has been put forward to rationalize the solvent partition between the two constituent phases. The objective of the present study was to determine the effect of high pressure (HP) processing (200, 450 and 650 MPa) at various temperatures (20, 40 and 55 degrees C) on the total plasmin plus plasminogen-derived activity (PL), plasminogen activator(s) (PA) and cathepsin D activities and on denaturation of major whey proteins in bovine milk. Data indicated that transfer of both PL and PA from the casein micelles to milk serum occurred at all pressures utilized at room temperature (20 degrees C). In addition to the transfer of PL and PA from micelles, there were reductions in activities of PL (16-18%) and PA (38-62%) for the pressures 450 and 650 MPa, at room temperature. There were synergistic negative effects between pressure and temperature on residual PL activity at 450 and 650 MPa and on residual PA activity only at 450 MPa. Cathepsin D activity in the acid whey from HP-treated milk was in general baroresistant at room temperature. The residual activity of cathepsin D decreased significantly at 650 MPa and 40 degrees C and at the pressures 450 and 650 MPa at 55 degrees C. Synergistic negative effects on the amount of native beta-lactoglobulin were observed at 450 and 650 MPa and on the amount of native alpha-lactalbumin at 650 MPa. There were significant correlations between enzymatic activities (PL, PA and cathepsin D) and the residual native beta-lactoglobulin and alpha-lactalbumin in bovine milk. In conclusion, HP significantly affected the activity of indigenous proteolytic enzymes and whey protein denaturation in bovine milk. Reduction in activity of indigenous enzymes (PL, PA and cathepsin D) and transfer of PL and PA from the casein to milk serum induced by HP is expected to have a profound effect on cheese yield, proteolysis during cheese ripening and quality of UHT milk during storage. 相似文献

Effect of vitamin E supplementation on neutrophil function, milk composition and plasmin activity in dairy cows in a commercial herd 总被引:1,自引:0,他引:1
Fifty-six Holstein dairy cows from a commercial dairy herd in the Northern part of Greece were used to determine the effect of vitamin E supplementation on immune parameters, milk composition and milk quality. Cows were assigned to one of two experimental groups: control (no vitamin E supplementation) and vitamin E supplementation. Supplementation of vitamin E started 4 weeks prior to and continued up to 12 weeks after parturition. Supplementation included daily oral administration of vitamin E at 3000 i.u./cow prepartum and was reduced to 1000 i.u./cow post partum. Blood samples were collected weekly for 8 weeks starting 4 weeks before parturition, neutrophils were isolated and the following parameters were determined in neutrophils activated by phorbol myristate acetate: total cell-associated and membrane-bound urokinase plasminogen activator (u-PA) activity and superoxide production. Milk samples were collected weekly and fat, protein, lactose, somatic cell count (SCC), plasmin and plasminogen-derived activity were determined. Activated neutrophils isolated from cows that received supplemental vitamin E had higher (P<0.01) total and membrane-bound u-PA activities during the first 3 weeks after parturition and higher (P<0.01) superoxide production during week 1 prepartum and week 1 post partum compared with the corresponding values of activated neutrophils isolated from control cows. Vitamin E supplementation had no effect (P=0.28) on plasminogen-derived activity in milk. Milk obtained from cows that received supplemental vitamin E had SCC lower by 25% (P<0.05) and plasmin lower by 30% (P<0.01) than corresponding values in milk obtained from control cows. The reduction in plasmin as a result of vitamin E supplementation is very beneficial to the dairy industry because plasmin reduces the cheese-yielding capacity of milk, affects the coagulating properties of milk and its overall ability to withstand processing during cheesemaking. In conclusion, vitamin E supplementation had positive effects on the function of bovine neutrophils and milk quality in a commercial dairy herd. 相似文献

The physicochemical and microbiological characteristics of spontaneously fermented sausages made by two medium-sized enterprises (MSE) located in southern Greece have been studied. A total of 300 lactic acid bacteria and 300 staphylococcal strains have been isolated and identified by their physiological characteristics. Lactobacillus plantarum strains were found to dominate the lactic acid bacteria microbiota in most of the cases with L. sakei strains prevailing in some of them and L. rhamnosus strains occasionally accompanying the dominant lactic acid bacteria microbiota. On the other hand, S. saprophyticus strains were found to dominate the staphylococcal microbiota in all spontaneously fermented sausages with of S. simulans, S. xylosus, S. gallinarum and S. cohnii cohnii strains being sporadically present. Following the identification, an evaluation of their technological properties, namely proteolytic and lipolytic capacities as well as production of biogenic amines and antimicrobial compounds, took place. None of the lactic acid bacteria and staphylococci was found to possess lipolytic activity whereas a total of 6 lactic acid bacteria and 51 staphylococci strains were found to be able to hydrolyse either the sarcoplasic, myofibrillar or both protein fractions. Furthermore, only one L. sakei strain and 185 staphylococci strains were found to possess decarboxylase activity against lysine, tyrosine, ornithine or histidine. Finally none of the staphylococcal microbiota and 3 lactic acid bacteria strains were found to be able to produce antimicrobial compounds of proteinaceous nature against Listeria monocytogenes. 相似文献
