Organizations operate in environments of vulnerability. While there are methodologies that guide business continuity management, it is evident that organizations remain vulnerable to disruptions emerging in times of transformation and systemic change. This work aims to compile and analyze the disruptions and related risk management strategies or activities described in the literature, and explore which disruptions are creating concern in companies and how they approach them. The methodology is based on a review of the literature and from the review we define a questionnaire to guide semi-structured interviews to managers from companies all located in a highly volatile macroeconomic context. The literature describes disruptions related with governance, cyberattacks, technology, supply chain, internal processes, and shock events and both precrises and postcrises strategies. The empirical analysis using data from companies reveals that (a) the most stressful disruptions are the currency devaluation, the high inflation rate, and government interventions and (b) managers rely on collaborative work, effective communication between stakeholders, supportive leadership, and constant adaptation to the new environment. Future research is required on techniques supporting adaptation to ‘new normalcy’. 相似文献

Ecto-nucleoside triphosphate diphosphohydrolases (NTPDases) are enzymes located on the surface of the T. cruzi plasma membrane, which hydrolyze a wide range of tri-/-diphosphate nucleosides. In this work, we used previously developed genetically modified strains of Trypanosoma cruzi (T. cruzi), hemi-knockout (KO +/−) and overexpressing (OE) the TcNTPDase-1 gene to evaluate the parasite infectivity profile in a mouse model of acute infection (n = 6 mice per group). Our results showed significantly higher parasitemia and mortality, and lower weight in animals infected with parasites OE TcNTPDase-1, as compared to the infection with the wild type (WT) parasites. On the other hand, animals infected with (KO +/−) parasites showed no mortality during the 30-day trial and mouse weight was more similar to the non-infected (NI) animals. In addition, they had low parasitemia (45.7 times lower) when compared with parasites overexpressing TcNTPDase-1 from the hemi-knockout (OE KO +/−) group. The hearts of animals infected with the OE KO +/− and OE parasites showed significantly larger regions of cardiac inflammation than those infected with the WT parasites (p < 0.001). Only animals infected with KO +/− did not show individual electrocardiographic changes during the period of experimentation. Together, our results expand the knowledge on the role of NTPDases in T. cruzi infectivity, reenforcing the potential of this enzyme as a chemotherapy target to treat Chagas disease (CD). 相似文献

This paper presents a quadratic boost converter using a single quasi-resonant network to reach soft commutation. A resonant inductor, a resonant capacitor, and an auxiliary switch form the resonant network and the main switch operates in a zero-current-switching way. A complete analysis of this converter is presented. According to the simulation and experimental results, this quadratic boost converter provides a larger conversion ratio than that provided by the conventional boost converter (for a given duty ratio D), and presents optimum performance, which operates with soft-switch commutation using a single resonant network. 相似文献

The hyperphosphorylation of microtubule‐associated protein tau (tau) in the hippocampus can be caused by central and peripheral insulin resistance and these alterations are related to the development of tauopathies, such as Alzheimer's disease. In this study, we used a high‐fat diet to induce obesity and insulin resistance in adult Swiss mice and checked whether supplementation with Myrciaria jaboticaba berry peel for 10 weeks could improve insulin sensitivity, learning/memory performance, and prevent tau phosphorylation in the hippocampus. Furthermore, adipocytokines, inflammatory markers, and oxidative stress were assessed. Myrciaria jaboticaba peel has phenolic compounds (e.g., cyanidin, ellagic acid), dietary fiber and carotenoids, which contribute to great antioxidant capacity. Supplementation of the high‐fat diet with 4% M. jaboticaba peel prevented fat weight gain and reduced peripheral insulin resistance. The treated group also showed lower tau phosphorylation in the hippocampus corroborating better learning/memory performance in the Morris water maze test. Maintenance of neuronal viability, lower levels of hippocampal inflammatory markers, and improved brain antioxidant defenses were also related to the consumption of M. jaboticaba peel. These findings contribute to a better understanding of how a high‐fat diet supplemented with jaboticaba berry peel counteracts the impairment of cognitive functions caused by high‐fat diet intake and diet‐induced insulin resistance. 相似文献

We have shown in 2 independent studies that cows who received recombinant bovine interleukin-8 (rbIL-8) administered intrauterinely shortly after parturition have a significant and long-lasting increase in milk yield. In the present study, we hypothesized that the increased milk production associated with rbIL-8 treatment is a consequence of increased postpartum dry matter intake (DMI) and orchestrated homeorhetic changes that prioritize milk production. Cows were enrolled into 1 of 3 treatment groups: those assigned to the control group (CTR; n = 70) received an intrauterine (IU) administration of 500 mL of Dulbecco's phosphate-buffered saline (DPBS) solution and 1 mL of DPBS solution intravenously (IV; jugular vein), those assigned to the rbIL-8 IV group (rbIL8-IV, n = 70) received an IV injection of 167 μg of rbIL-8 and 500 mL of DPBS solution IU, and cows assigned to the rbIL-8 IU group (rbIL8-IU, n = 70) received an IU administration with 1,195 μg of rbIL-8 diluted in 499.5 mL of DPBS solution and 1 mL of DPBS solution IV. Animals were housed in a tiestall from calving to 30 d in milk (DIM) to measure DMI. Blood samples were collected daily from calving to 7 DIM and weekly until 28 DIM. Insulin resistance was evaluated using an intravenous glucose tolerance test and intravenous insulin challenge test (IVICT) in a subgroup of cows (n = 20/treatment) at 10 and 11 DIM, respectively. Additionally, liver biopsy samples were taken at 14 DIM from the same subgroup of cows to measure triglyceride levels and cell proliferation and apoptosis. Cows treated with rbIL8-IU produced more milk (CTR = 36.9 ± 1.5; rbIL8-IU = 38.5 ± 1.5; rbIL8-IV = 36.6 ± 1.5 kg/d), energy-corrected milk (CTR = 42.9 ± 0.9; rbIL8-IU = 46.1 ± 0.8; rbIL8-IV = 43.7 ± 0.9 kg/d), and fat-corrected milk (CTR = 44.3 ± 0.9; rbIL8-IU = 47.8 ± 0.9; rbIL8-IV = 45.2 ± 0.9 kg/d) yields when compared with CTR cows, and no differences were observed between rbIL8-IV and CTR cows. The administration of rbIL8-IU significantly increased DMI compared with CTR (CTR = 18.8 ± 0.3; rbIL8-IU = 19.9 ± 0.3; rbIL8-IV = 19.3 ± 0.3 kg/d). Recombinant bIL-8 treatment did not affect glucose, insulin, or fatty acids (i.e., IVICT only) concentrations or their area under the curve in response to an intravenous glucose tolerance test and IVICT when compared with CTR. Moreover, rbIL-8 treatment administered IU or IV increased liver triglyceride levels. Additionally, cows treated with rbIL8-IU tended to have lower odds of developing hyperketonemia (odds ratio = 0.46, 95% confidence interval: 0.19 to 1.10), lower odds of clinical ketosis and displaced abomasum combined (odds ratio = 0.17, 95% confidence interval: 0.03 to 0.89), and lower odds of diseases combined (odds ratio = 0.43, 95% confidence interval: 0.21 to 0.86) when compared with CTR. We conclude that the administration of rbIL8-IU increases DMI, milk production, fat-corrected milk, and energy-corrected milk while improving overall health during the postpartum period. This study supports the use of rbIL-8 administered IU shortly after calving to improve health and production responses in lactating cows. 相似文献

The main objective of this study was to compare the proximate chemical composition and fatty acid profile of migrating allis shad (MS) and resident allis shad (RS) and to evaluate the nutritional value of fried migrating allis shad. No significant differences in the proximate chemical composition between males and females of each stock were obtained. However, MS fish had a significantly higher fat content than RS fish. The monounsaturated fatty acids group was the dominant in MS fish and oleic acid presented the highest percentage. Docosahexaenoic acid followed by eicosapentaenoic acid were the main PUFA in fish from both stocks. However, RS fish had higher total ω6 than MS fish due to its higher percentage of linoleic acid and arachidonic acid. Frying resulted in a significant reduction in water content and affected significantly the proximate chemical composition and the fatty acid profile, particularly the palmitic, oleic and linoleic acid. 相似文献

The main goal of the present study was to discuss the application of the McNemar test to the comparison of proportions in dependent samples. Data were analyzed from studies conducted to verify the suitability of replacing a conventional method with a new one for identifying the presence of Salmonella. It is shown that, in most situations, the McNemar test does not provide all the elements required by the microbiologist to make a final decision and that appropriate functions of the proportions need to be considered. Sample sizes suitable to guarantee a test with a high power in the detection of significant differences regarding the problem studied are obtained by simulation. Examples of functions that are of great value to the microbiologist are presented. 相似文献

This work was aimed at isolating and identifying the microbiota present during the semi-dry method of coffee processing using polyphasic methods and to evaluate microbial diversity with PCR-DGGE. Samples of Coffea arabica L. were collected during different processing stages in southern Minas Gerais, Brazil. The bacterial and fungal isolates were phenotypically characterised and grouped according to the ARDRA technique, in which the 16–23S and ITS1-5.8S regions of the rDNA were sequenced for species identification. The bacterial counts varied from 3.7 to 7 log CFU g−1. The yeast counts ranged from 3.4 to 6.9 log CFU g−1, and the filamentous fungal population varied from 2 to 3.7 log CFU g−1. Bacillus subtilis, Escherichia coli, Enterobacter agglomerans, Bacillus cereus and Klebsiella pneumoniae were the predominant bacteria detected during the processing of the coffee, and Pichia anomala, Torulaspora delbrueckii and Rhodotorula mucilaginosa were the dominant yeasts. All of the yeast and bacterial species detected by PCR-DGGE were isolated using culture-dependent methods, with the exception of one uncultivable bacterial species. Aspergillus was the most common genus among the filamentous fungal isolates. The use of polyphasic methods allowed a better characterization of the microbiota that is naturally present in semi-dry processed coffee. 相似文献

Microalgae are an enormous biological resource, representing one of the most promising sources for the development of new food products and applications. Pea protein/κ-carrageenan/starch gels, interesting vegetarian alternatives to dairy desserts, served as model systems to study the addition of microalgal biomass, its effect, and subsequent rheological behaviour. Spirulina and Haematococcus gels presented a markedly different rheological behaviour compared to the control mixed biopolymer gelled system. The present goal is to clarify how these microalgae affect the gelation and interact with each biopolymer present in the complex mixed gel system. Hence, the aim of the present work is to study the effect of Spirulina and Haematococcus microalgal biomass addition on the rheological behaviour of pea protein, κ-carrageenan and starch simple gels, as well as in pea protein/κ-carrageenan and pea protein/starch systems. The gelation process was monitored in-situ through dynamic oscillatory measurements (temperature, time and frequency sweep tests) for a 24 h maturation period, and rheological results were supported with fluorescence optical microscopy observations. The addition of Spirulina and Haematococcus to biopolymer gelled systems induced significant changes in the gels’ rheological behaviour and microstructure. In general, it was observed that the gelling mechanism is ruled by the biopolymers, while microalgae seem to be embedded in the gel network acting as active particle fillers. The addition of Haematococcus resulted in more structured gels in comparison to the control and Spirulina systems. In the case of κ-carrageenan gels, both microalgae induced a large increase in the rheological parameters, which should be related to the high ionic content of microalgal biomass. Spirulina addition on starch systems promoted a decrease in the gels’ rheological parameters.
