The authors describe a transient phase during training on a movement sequence wherein, after an initial improvement in speed and decrease in variability, individual participants' performance showed a significant increase in variability without change in mean performance speed. Subsequent to this phase, as practice continued, variability again decreased, performance significantly exceeded the gains predicted by extrapolation of the initial learning curve, the type of errors committed changed, and performance became more coherent. The transient phase of increased variability may reflect a mixture of 2 (or more) performance routines before the more effective one is set and mastered, presumably the setting up of a sequence-specific representation. Both group and individual analyses indicated a departure from the single process (e.g., power-law) model of learning. However, although similar phases appeared in the mean group data, there was little correspondence to individual participants' time courses, and the individuals' gains in the second low-variability phase were masked. The accumulation of arsenic in fur and feathers has been used as an indicator of environmental quality and animal health. However, difficulties remain in distinguishing between arsenic present from external sources versus ingestion. In addition, low extraction efficiencies limit the complete characterization of arsenic compounds in such tissues by conventional analytical techniques (e.g. high performance liquid chromatography inductively coupled plasma mass spectrometry, HPLC-ICP-MS). X-ray absorption spectroscopy (XAS) provides an alternative method for determining the speciation of arsenic compounds directly. Inorganic arsenic is hypothesized to bind to thiol groups present in keratin-rich fur and feathers; however, arsenic-sulphur compounds are rarely reported in extracts of these tissues. Here we report that 5-58% of the total detected arsenic in rodent fur (vole, deer mouse, red squirrel) and bird feathers (gray jay, American tree sparrow, dark-eyed junco) from animals living in areas of elevated arsenic best resembled an arsenic(III)-sulphur compound as determined using XAS. In addition, fur and feathers with non-detectable levels of arsenic by XAS, were able to reduce pentavalent arsenic applied as either contaminated soil or an arsenate solution. XAS-imaging was used to localize dominant trivalent (AsIII) and pentavalent (AsV) arsenic compounds, and results were used to produce a "map" of arsenic in the sample. It is believed that the some of the reduced arsenic was absorbed, while external AsV compounds associated with soil/dust particles were easily distinguished on goose feathers. However, distinguishing whether internal arsenic arose from exogenous (from the environment) or endogenous (from the body) sources proved difficult with this technique.  相似文献

Fast and accurate monitoring of chemical and microbiological parameters in drinking water is essential to safeguard the consumer and to improve the understanding of treatment and distribution systems. However, most water utilities and drinking water guidelines still rely solely on time-requiring heterotrophic plate counts (HPC) and plating for faecal indicator bacteria as regular microbiological control parameters. The recent development of relative simple bench-top flow cytometers has made rapid and quantitative analysis of cultivation-independent microbial parameters more feasible than ever before. Here we present a study using a combination of cultivation-independent methods including fluorescence staining (for membrane integrity, membrane potential and esterase activity) combined with flow cytometry and total adenosine tri-phosphate (ATP) measurements, to assess microbial viability in drinking water. We have applied the methods to different drinking water samples including non-chlorinated household tap water, untreated natural spring water, and commercially available bottled water. We conclude that the esterase-positive cell fraction, the total ATP values and the high nucleic acid (HNA) bacterial fraction (from SYBR((R)) Green I staining) were most representative of the active/viable population in all of the water samples. These rapid methods present an alternative way to assess the general microbial quality of drinking water as well as specific events that can occur during treatment and distribution, with equal application possibilities in research and routine analysis.  相似文献

Here, we present the formation of a fully addressable DNA nanostructure that shows the potential to be exploited as, for example, an information storage device based on pH-driven triplex strand formation or nanoscale circuits based on electron transfer. The nanostructure is composed of two adjacent hexagonal unit cells (analogous to naphthalene) in which each of the eleven edges has a unique double-stranded DNA sequence, constructed using novel three-way oligonucleotides. This allows each ten base-pair side, just 3.4 nm in length, to be assigned a specific address according to its sequence. Such constructs are therefore an ideal precursor to a nonrepetitive two-dimensional grid on which the "addresses" are located at a precise and known position. Triplex recognition of these addresses could function as a simple yet efficient means of information storage and retrieval. Future applications that may be envisaged include nanoscale circuits as well as subnanometer precision in nanoparticle templating. Characterization of these precursor nanostructures and their reversible targeting by triplex strand formation is shown here using gel electrophoresis, atomic force microscopy, and fluorescence resonance energy transfer (FRET) measurements. The durability of the system to repeated cycling of pH switching is also confirmed by the FRET studies.  相似文献

The 2‐ethoxycarbonylallyl 5‐(1,2‐dithiolane‐3‐yl)‐pentanoate monomer (AODS) includes in its molecular structure C?C and S? S reactive bonds allowing it to behave as a bi‐functional monomer, possessing two groups, however, with different reactivity for use in polymer chain building. The polymerization‐specific features of this monomer are the absence of auto‐acceleration and polymer chain crosslinking. Polymerization proceeds readily through most free‐radical initiators. One exception, carboxy‐peroxides are rapidly decomposed without the production of free radicals. AODS is partially converted to a gel without the consumption of double bonds during monomer dissolution in certain organic solvents and after being mixed in solution with carboxy‐peroxides. The determined AODS‐co‐MMA copolymerization parameters are r1 = 2.61, r2 = 0.23 if Luperco peroxide is used as a polymerization initiator, and r1 = 2.71, r2 = 0.38 if AIBN is used.

An in situ technique for preparing composite nanoparticles from hydrophobic cellulose acetate and hydrophilic polysaccharides using nanoprecipitation is presented. This technique allows the nanoparticles’ surface properties to be tuned very specifically. Spherical, narrow‐size‐distributed composite nanoparticles of different size, charge, functionality, and increased stability can be generated by using hydroxyethyl cellulose, carboxymethyl cellulose, low molecular weight chitosan, and amino cellulose. The influence of the pH and hydrophilic polysaccharide content in the particle formation is shown. The pH‐ and ionic strength‐ effective zeta‐potential functions are evidence of the presence of functional polysaccharides at the nanoparticle surface. The in situ technique is compared with the adsorption of hydrophilic polysaccharides onto cellulose acetate nanoparticles in two steps. The great potential of in situ prepared composite nanoparticles in the pharmaceutical industry and bio‐ or food technology, as carriers of hydrophobic substances in aqueous media and for specific surface modifications, e.g., to selectively introduce strong antimicrobial properties, is illustrated.  相似文献

Dynamics of γ-aminobutyric acid in wheat flour bread making   总被引:1,自引:0,他引:1  
The dynamics of the health-improving non-protein amino acid γ-aminobutyric acid (GABA) during bread making were studied. Wheat flour contains trace levels of GABA (<15 ppm) and ca. 160–175 ppm of its precursor, glutamic acid (GA). During dough mixing, the levels of both GA and GABA largely increased. While wheat flour endogenous glutamic acid decarboxylase (GAD) performs some minor conversion of GA into GABA, yeast is the main contributor to GABA formation. Comparison of amino acid levels of dough samples, without or with yeast, indicated that yeast favours both GA and GABA formation already during mixing. Fermentation decreased both GA and GABA contents, due to amino acid consumption by the yeast, which used more GA than GABA. Proofing and baking resulted in large GABA losses, the latter probably in Maillard browning reactions during baking. Thermal loss of GA was less pronounced than that of GABA. Breads contained only trace levels of GABA and ca. 90–130 ppm of its precursor. Exogenous supplementation of recombinantly produced GAD of Yersinia intermedia decreased GA levels in mixed and fermented dough and increased GABA levels. The highest GAD dosage used resulted in fermented doughs with ca. 300 ppm of GABA, i.e. three times higher than the level present in the reference sample (no GAD added). After baking, a significant GABA level was left in the bread samples (ca. 115 ppm) and GABA-enriched breads were obtained. Addition of sodium glutamate (100–380 ppm) to a bread recipe containing no added GAD clearly indicated that its precursor was not the limiting factor for GABA conversion during bread making since the resulting breads contained no GABA, or only trace levels (ca. 20 ppm).  相似文献

We investigate tropical cyclone wind and storm surge damage reduction for five areas along the Miami-Dade County coastline either by hardening buildings or by the hypothetical application of wind-wave pumps to modify storms. We calculate surge height and wind speed as functions of return period and sea surface temperature reduction by wind-wave pumps. We then estimate costs and economic losses with the FEMA HAZUS-MH MR3 damage model and census data on property at risk. All areas experience more surge damages for short return periods, and more wind damages for long periods. The return period at which the dominating hazard component switches depends on location. We also calculate the seasonal expected fraction of control damage for different scenarios to reduce damages. Surge damages are best reduced through a surge barrier. Wind damages are best reduced by a portfolio of techniques that, assuming they work and are correctly deployed, include wind-wave pumps.  相似文献

In this work we investigated the diffusion of thiol molecules during microcontact printing (μCP) with rigid stamps – a process we call reversal μCP. We use such stamps instead of soft polydimethylsiloxane (PDMS) stamps to overcome the problems of sagging and pairing of structures and deformation. We varied thiol concentration in ethanol as well as different contact times. It shows that the lower the concentration the thinner the lines and the longer the contact time the thicker the lines until saturation is reached. Working in this saturation region overcomes the problem of the strong dependency of line width over contact time which is usually observed in microcontact printing and which restricts the reproducibility of the process itself. Gold lines with a width down to 80 nm have been achieved using this novel method.  相似文献

Arsenic is one of the most widely encountered environmental contaminants because of a number of anthropogenic sources; in Canada the main anthropogenic release of arsenic is from mine tailings ponds. The present study is part of a series of studies to measure chemical and biological effects of exposure for meadow voles (Microtus pennsylvanicus) living on arsenic contaminated sites. Two additional objectives were addressed in the present study: the effect of higher arsenic concentrations compared with previous studies, and the comparison of chemical speciation and biological effects. To obtain the higher environmental concentrations, specimens were collected from a former gold mining site in Montague, NS that contains highly elevated concentrations of arsenic in soils and plants. Meadow voles were collected and their tissues were analyzed for total arsenic to measure uptake, and arsenic speciation to examine the chemical effects of the high arsenic exposure. In addition to the arsenic analysis, a biomonitoring study was undertaken to examine the sub-cellular effects in meadow voles resulting from the elevated arsenic exposure. Meadow voles living on the contaminated site had substantially higher concentrations of total arsenic than animals from the background (reference) location. The extractable arsenic in internal tissues was present mainly as monomethylarsonic acid (up to 14% of total arsenic). A statistically significant relationship was observed between the reduction of glutathione in vole livers and the increase in liver arsenic concentrations, and micronucleated monochromatic red blood cells were also significantly elevated in voles from the arsenic contaminated site. This is one of the few field studies where sub-cellular effects were observed, and the first to show a co-existence of such effects with relatively high proportions of monomethylarsonic acid in voles living near mine tailings.  相似文献
