Silver nanoparticles represent a threat to biota and have been shown to cause harm through a number of mechanisms, using a wide range of bioassay endpoints. While nanoparticle concentration has been primarily considered, comparison of studies that have used differently sized nanoparticles indicate that nanoparticle diameter may be an important factor that impacts negative outcomes. In considering this, the aim of the present study was to determine if different sizes of silver nanoparticles (AgNPs; 10, 20, 40, 60 and 100 nm) give rise to similar effects during embryogenesis of Mediterranean sea urchins Arbacia lixula and Paracentrotus lividus, or if nanoparticle size is a parameter that can modulate embryotoxicity and spermiotoxicity in these species. Fertilised embryos were exposed to a range of AgNP concentrations (1–1000 µg L−1) and after 48 h larvae were scored. Embryos exposed to 1 and 10 µg L−1 AgNPs (for all tested sizes) showed no negative effect in both sea urchins. The smaller AgNPs (size 10 and 20 nm) caused a decrease in the percentage of normally developed A. lixula larvae at concentrations ≥50 µg L−1 (EC50: 49 and 75 μg L−1, respectively) and at ≥100 µg L−1 (EC50: 67 and 91 μg L−1, respectively) for P. lividus. AgNPs of 40 nm diameter was less harmful in both species ((EC50: 322 and 486 μg L−1, for P. lividus and A. lixula, respectively)). The largest AgNPs (60 and 100 nm) showed a dose-dependent response, with little effect at lower concentrations, while more than 50% of larvae were developmentally delayed at the highest tested concentrations of 500 and 1000 µg L−1 (EC50(100 nm); 662 and 529 μg L−1, for P. lividus and A. lixula, respectively. While AgNPs showed no effect on the fertilisation success of treated sperm, an increase in offspring developmental defects and arrested development was observed in A. lixula larvae for 10 nm AgNPs at concentrations ≥50 μg L−1, and for 20 and 40 nm AgNPs at concentrations >100 μg L−1. Overall, toxicity was mostly ascribed to more rapid oxidative dissolution of smaller nanoparticles, although, in cases, Ag+ ion concentrations alone could not explain high toxicity, indicating a nanoparticle-size effect. 相似文献

A large metabolomics study was performed on 600 plasma samples taken at four time points before and after a single intake of a high fat test meal by obese and lean subjects. All samples were analyzed by a liquid chromatography-mass spectrometry (LC-MS) lipidomic method for metabolic profiling. A pragmatic approach combining several well-established statistical methods was developed for processing this large data set in order to detect small differences in metabolic profiles in combination with a large biological variation. Such metabolomics studies require a careful analytical and statistical protocol. The strategy included data preprocessing, data analysis, and validation of statistical models. After several data preprocessing steps, partial least-squares discriminant analysis (PLS-DA) was used for finding biomarkers. To validate the found biomarkers statistically, the PLS-DA models were validated by means of a permutation test, biomarker models, and noninformative models. Univariate plots of potential biomarkers were used to obtain insight in up- or downregulation. The strategy proposed proved to be applicable for dealing with large-scale human metabolomics studies. 相似文献

The present study was performed to evaluate the antioxidant capacity, the polyphenol and metal content of conventionally and organically produced wines, which underwent similar winemaking processes. The wine grapes were grown under well-defined organic and conventional conditions from the two different wine-growing sub-regions of Croatia, Prigorje and Southern Dalmatia. The values of antioxidant activity, as evaluated by two free radical methods, ABTS and DPPH, was found systematically higher in organic wines compared to conventional ones. In a reversed phase HPLC analysis, used in order to characterise the phenol fraction of wines, higher concentrations of chlorogenic acid, ferulic acid, catechin, trans-resveratrol, all studied hydroxybenzoic acids and flavonols were found in the organically produced wines. No apparent trend was found in the metal contents of the wines, but ICP-MS analysis confirmed that both wine productions, organic and conventional, were well within the toxicological safety limits. 相似文献

Thermodynamic studies on phase transformation of biopolymers in solution are useful to understand their nature and to evaluate their technological potentials. Thermodynamic studies should be conducted avoiding time‐related phenomena. This condition is not easily achieved in hydrophilic biopolymers. In this contribution, the simultaneous effects of pH, salt concentration, and cooling rate (Cr) on the folding from random coil to triple helical collagen‐like structures of gelatin were systematically studied. The phase transformation temperature at the absolute invariant condition of Cr = 0 °C/min (TTCr=0) is introduced as a conceptual parameter to study phase transformations in biopolymers under quasi‐thermodynamic equilibrium and avoiding interferences coming from time‐related phenomena. Experimental phase diagrams obtained at different Cr are presented. The TTCr=0 compared with pH and TTCr=0 compared with [NaCl] diagram allowed to explore the transformation process at Cr = 0 °C/min. The results were explained by electrostatic interactions between the biopolymers and its solvation milieu. 相似文献

The selective surface oxidation of the Ti-stabilized interstitial free (Ti-IF) and dual phase (DP) steel during recrystallization annealing at 820 °C under the variety of exposure time and composition of protective H2-N2 atmosphere at low dew point (−40 °C) was examined. It was found that Mn, Al, Si and Cr oxides are formed in all cases, but higher Mn and Si concentration in DP steel leads to an increase in a density of coarse nonwettable MnSiO3 particles. An increase in annealing time enhances surface coverage with oxides, while increased H2 content in gas atmosphere results in less coverage with external oxides improving the wettability. The influence of the stronger reducing atmosphere is more exhibit on Ti-IF steel where after 120 s of annealing in 15vol.%H2-85vol.%N2 atmosphere the wetting angle θ stabilized to a value of the order of 6°. The presence of surface BN and the TiN particles also affects the full surface coverage by external oxides. 相似文献

The abandoned Dúbrava Mine, situated in the northern part of the Nízke Tatry Mts in the middle of Slovakia, was the most significant producer of antimony (Sb) in the former Czechoslovakia. Mine drainage from adits (containing up to 9,300 μg/L of Sb), mine waste dumps, and the leachate from mine tailings contribute Sb and arsenic (As) into nearby Palud?anka Creek and groundwater. Some drinking water resources have been closed due to excessive Sb concentrations; the concentration of Sb in one household well (126 μg/L) far exceeds the Sb drinking water limit of 5 μg/L. Although Sb is attenuated by dilution and adsorption on ferric iron in stream sediment in the Palud?anka Creek, Sb concentrations increase downstream of the mine tailings and then remain almost constant, leading to concentrations of 128 μg/L at the northern boundary of the study area. The dissolved As concentrations in the mine drainage are much lower than Sb, ranging from 4 to 62 μg/L. Flow and transport modeling confirmed the observed contamination pattern and the major role of the mine adits. Results of this study indicate serious Sb contamination, which could be mitigated by treatment of the adit discharges. 相似文献
