(1) Background: Melanoma is an aggressive neoplasm derived from melanocyte precursors with a high metastatic potential. Responses to chemotherapy and immunotherapy for melanoma remain weak, underlining the urgent need to develop new therapeutic strategies for the treatment of melanoma. (2) Methods: The viability of NHDF and A375 cell cultures after the administration of the tested isoxazole derivatives was assessed after 24-h and 48-h incubation periods with the test compounds in the MTT test. ROS and NO scavenging analyses, a glycoprotein-P activity analysis, a migration assay, a test of apoptosis, and a multiple-criteria decision analysis were also performed. (3) Results: All compounds that were tested resulted in a slower migration of melanoma neoplastic cells. The mechanism of the antitumor activity of the tested compounds was confirmed—i.e., the pro-apoptotic activity of the compounds in A375 cell cultures. Compound O7K qualified for further research. (4) Conclusions: All the tested compounds inhibited the formation of melanoma metastases and demonstrated the ability to reduce the risk of developing drug resistance in the tumor. The MCDA results showed that O7K showed the strongest antitumor activity.  相似文献

Activated carbon (AC) fiber cloths and a hydrophobic microporous polypropylene (PP) membrane, both modified with lithiated acetone oligomers, were used as electrodes and a separator in electric double layer capacitors (EDLCs) with aqueous lithium hydroxide (LiOH) as the electrolyte. Electrochemical characteristics of EDLCs were investigated by cyclic voltammetry (CV), galvanostatic charge-discharge cycle tests and impedance spectroscopy (EIS), compared with a case of the capacitor with aqueous potassium hydroxide (KOH) as an electrolyte. As a result, the capacitor with LiOH aqueous solution and a modified separator and electrodes was found to exhibit higher specific capacitance, maximum energy stored and maximum power than that with KOH aqueous solution.  相似文献

Properties of capacitors working with the same carbon electrodes (activated carbon cloth) and three types of electrolytes: aqueous, organic and ionic liquids were compared. Capacitors filled with ionic liquids worked at a potential difference of 3.5 V, their solutions in AN and PC were charged up to the potential difference of 3 V, classical organic systems to 2.5 V and aqueous to 1 V. Cyclic voltammetry, galvanostatic charging/discharging and impedance spectroscopy were used to characterize these capacitors. The highest specific energy was recorded for the device working with ionic liquids, while the highest power is characteristic for the device filled with aqueous H2SO4 electrolyte. Aqueous electrolytes led to energy density an order of magnitude lower in comparison to that characteristic of ionic liquids.  相似文献

Environmental stress is one of the most important factors affecting plant growth and development. Recent studies have shown that epigenetic mechanisms, such as DNA methylation, play a key role in adapting plants to stress conditions. Here, we analyzed the dynamics of changes in the level of DNA methylation in Arabidopsis thaliana (L.) Heynh. (Brassicaceae) under the influence of heat stress. For this purpose, whole-genome sequencing of sodium bisulfite-treated DNA was performed. The analysis was performed at seven time points, taking into account the control conditions, heat stress, and recovery to control conditions after the stress treatment was discontinued. In our study we observed decrease in the level of DNA methylation under the influence of heat stress, especially after returning to control conditions. Analysis of the gene ontology enrichment and regulatory pathways showed that genes characterized by differential DNA methylation are mainly associated with stress response, including heat stress. These are the genes encoding heat shock proteins and genes associated with translation regulation. A decrease in the level of DNA methylation in such specific sites suggests that under the influence of heat stress we observe active demethylation phenomenon rather than passive demethylation, which is not locus specific.  相似文献

Psoriasis is accompanied by disturbed redox homeostasis, with systemic and local oxidative stress promoting the modification of basic components of cellular membranes. Therefore, the aim of the study was to investigate the effect of development of psoriasis vulgaris and psoriatic arthritis on the composition and physicochemical properties of skin cell membranes (keratinocytes and fibroblasts) and blood cells (lymphocytes, granulocytes and erythrocytes). Both forms of psoriasis are characterized by decreased levels and changes in the localization of membrane phospholipids, and an increased level of sialic acid as well as the lipid peroxidation product (malondialdehyde), which resulted in an increase in the zeta potential of skin cells and blood cells, with granulocytes and lymphocytes affected more than erythrocytes. Using theoretical equations and the dependence of the cell membrane surface charge density as a function of pH, it was shown that patients with psoriatic arthritis have a greater increase in the concentration of negatively charged groups on the membrane surface and reduced the value of the association constant with H+ compared to patients with psoriasis vulgaris. Therefore, it can be suggested that the physicochemical parameters of membranes, skin and blood cells, especially lymphocytes, can be used to assess the severity of the disease.  相似文献

The effects of concentration and surface modification of two Brazilian bentonite clays on nanocomposites' properties based on polylactic acid – (PLA) were investigated. The samples were prepared by the extrusion/injection method to obtain biodegradable packaging plastics. The raw materials and their bionanocomposites were characterized by various techniques. Natural clay samples presented a size of around 2 μm while the modified ones' size was 5–6 μm, probably due to the presence of cetyltrimethylammonium bromide in the interlayer space. The particle size and the contact angle increased with the treatment and the clay's density decreased. The organoclays were homogeneously dispersed in PLA, which can be associated with the interactions between PLA chains' carbonyl groups and the organoclays. The bionanocomposites present modified clay particles axis aligned to the flow direction of the extruder/injector. Chocolate organoclay acts as a nucleating agent to PLA crystal growth, increasing the sample's crystallinity, while Bofe organoclay interferes with the amorphous chain's mobility and diminishes the sample's crystallization. The addition of both organoclays to PLA diminished the sample's elongation at break and strength, although the organoclays increased the sample's Young modulus, even though Bofe organoclay is more active in PLA amorphous phase and Chocolate organoclay on the crystalline one.  相似文献

The aim of the present research was to investigate the influence of different biomasses and sewage sludge ash (at 650°C for 1000 h and 2000 h, respectively) on the surface of weld cladded (Cold Metal Transfer technique, CMT) stainless steels 309 and 310. The biomass ashes were rich in K2O, CaO, SiO2, whereas sewage sludge ash in P2O5, CaO, Fe2O3, and SiO2. Characterization of the stainless steel cross-section and surface after the corrosion process inducted by the presence of ash were carried out by scanning electron microscopy (SEM) with EDAX analysis. Phase analysis of corrosion products was made by X-ray diffraction (XRD). Chromium, nickel, and iron oxides were the main oxides which occurred after the corrosion test. It has been proved that biomass ashes have the corrosive properties (because of their chemical composition) and are more aggressive than the sewage sludge ash for that kind of materials.  相似文献

ADAM10 is the main α-secretase that participates in the non-amyloidogenic cleavage of amyloid precursor protein (APP) in neurons, inhibiting the production of β-amyloid peptide (Aβ) in Alzheimer’s disease (AD). Strong recent evidence indicates the importance of the localization of ADAM10 for its activity as a protease. In this study, we investigated ADAM10 activity in plasma and CSF samples of patients with amnestic mild cognitive impairment (aMCI) and mild AD compared with cognitively healthy controls. Our results indicated that plasma levels of soluble ADAM10 were significantly increased in the mild AD group, and that in these samples the protease was inactive, as determined by activity assays. The same results were observed in CSF samples, indicating that the increased plasma ADAM10 levels reflect the levels found in the central nervous system. In SH-SY5Y neuroblastoma cells, ADAM10 achieves its major protease activity in the fraction obtained from plasma membrane lysis, where the mature form of the enzyme is detected, confirming the importance of ADAM10 localization for its activity. Taken together, our results demonstrate the potential of plasma ADAM10 to act as a biomarker for AD, highlighting its advantages as a less invasive, easier, faster, and lower-cost processing procedure, compared to existing biomarkers.  相似文献

Endometriosis is a gynecological disease, the pathogenesis of which seems to be directly associated with inflammatory processes. Serum concentrations of IL-1β, IL-6, hs-CRP, IgG, YKL 40 and PRL, in comparison to the well-known CA 125 levels, were studied with the aim of identifying an additional noninvasive inflammatory marker or set of markers characteristic for endometriosis. The study group included 43 women with endometriosis (E), 35 women with benign gynecological disorders but without endometriosis (NE, non-endometriosis) as a comparative group, and a control group consisting of 18 healthy subjects (C). The serum concentrations of IL-1β, IL-6, hs-CRP, YKL-40, PRL and CA 125 were significantly higher in the E group (median values: 0.41 pg/mL, 2.42 pg/mL, 2.33 mg/L, 79.30 ng/mL, 21.88 ng/mL and 68.00 U/mL, respectively) than in the control group (median values: 0.21 pg/mL, 0.98 pg/mL, 0.52 mg/L, 49.77 ng/mL, 12.08 ng/mL and 12.20 U/mL respectively), with the significance of p = 0.011, p < 0.001, p = 0.028, p = 0.005, p < 0.001 and p < 0.001, respectively. The IgG concentrations were significantly lower in the endometriosis group (median value: 1061.21 mg/dL) as compared to healthy women (median value: 1210.50 mg/dL; p = 0.025). Significant differences in concentrations of IL-6 (p = 0.040), hs-CRP (p = 0.007) and CA 125 (p < 0.001) were observed in stage III vs. stage IV of endometriosis. Significantly higher concentrations of IL-6 (p = 0.010), hs-CRP (p = 0.037) and PRL (p < 0.001) were observed in the NE group vs. the control group. Only CA 125 concentrations were significantly higher in endometriosis patients as compared to the non-endometriosis group (p < 0.001). The proposed panel of inflammatory markers, especially IL-6, PRL and CA 125, may become a useful tool to identify women with advanced endometriosis who could qualify for treatment.  相似文献
