In response to cardiac ischemia/reperfusion, proteolysis mediated by extracellular matrix metalloproteinase inducer (EMMPRIN) and its secreted ligand cyclophilin-A (CyPA) significantly contributes to cardiac injury and necrosis. Here, we aimed to investigate if, in addition to the effect on the funny current (I(f)), Ivabradine may also play a role against cardiac necrosis by reducing EMMPRIN/CyPA-mediated cardiac inflammation. In a porcine model of cardiac ischemia/reperfusion (IR), we found that administration of 0.3 mg/kg Ivabradine significantly improved cardiac function and reduced cardiac necrosis by day 7 after IR, detecting a significant increase in cardiac CyPA in the necrotic compared to the risk areas, which was inversely correlated with the levels of circulating CyPA detected in plasma samples from the same subjects. In testing whether Ivabradine may regulate the levels of CyPA, no changes in tissue CyPA were found in healthy pigs treated with 0.3 mg/kg Ivabradine, but interestingly, when analyzing the complex EMMPRIN/CyPA, rather high glycosylated EMMPRIN, which is required for EMMPRIN-mediated matrix metalloproteinase (MMP) activation and increased CyPA bonding to low-glycosylated forms of EMMPRIN were detected by day 7 after IR in pigs treated with Ivabradine. To study the mechanism by which Ivabradine may prevent secretion of CyPA, we first found that Ivabradine was time-dependent in inhibiting co-localization of CyPA with the granule exocytosis marker vesicle-associated membrane protein 1 (VAMP1). However, Ivabradine had no effect on mRNA expression nor in the proteasome and lysosome degradation of CyPA. In conclusion, our results point toward CyPA, its ligand EMMPRIN, and the complex CyPA/EMMPRIN as important targets of Ivabradine in cardiac protection against IR. 相似文献

Zircaloy-4 is susceptible to stress corrosion cracking (SCC) in solutions of iodine dissolved in different alcohols (methanol, ethanol, 1-propanol, 1-butanol, 1-pentanol and 1-octanol). The crack propagation rate is known to decrease as the solvent molecular weight increases, as a consequence of steric hindrance. However, the mechanism that operates during SCC is still unknown. In the present work the effect of temperature on SCC susceptibility was evaluated in 1-butanol and 1-pentanol iodine containing solutions. The dependence of the crack growth rate with temperature follows an Arrhenius law, and the activation energy obtained from experimental data is consistent with a process controlled by volume diffusion of the active species (the iodine-alcohol complex) to the crack tip. 相似文献

The phase diagram of polytetrafluoroethylene is enlarged by differential scanning calorimetry to include the concentration of hexafluoropropylene comonomer units. The two transitions near 292 and 303 K in the homopolymer move to lower temperatures and apparently become one at small concentrations. Analysis of the data yields 295 K for the temperature and 13.2 J/g for the heat of transition of an infinitely large homopolymer crystal. The heat of transition associated with the formation of a crystal defect is 0.021 eV. The qualitative features of the transition can be accounted for by a mean-field model which involves two order parameters corresponding to planar units and helix reversals. This model yields two transitions which move closer together and to lower temperatures with increasing comonomer concentration. Decreasing lamella thickness will have a qualitatively similar effect. 相似文献

The unusual stress–strain hysteresis loop shape exhibited by ceramic-matrix composites under cyclic loading has previously been explained as a result of either strain rate dependence of the frictional shear stress or crack closure. This investigation has determined that the response is due to neither mechanism. Instead, it is proposed that a variation of interfacial shear strength occurs during each cycle. A static coefficient of friction dominates immediately after loading or unloading. A much lower dynamic coefficient of friction operates once fiber sliding begins. This dynamic coefficient appears to be very dependent on surface roughness. 相似文献

A series of statistical copolymers derived from 2‐(dimethylamino)ethyl methacrylate with four different hydrophobic comonomers (ethyl, butyl, cyclohexyl, and octyl methacrylates) have been prepared via conventional free radical copolymerization under bulk conditions. The copolymers have been subsequently modified, with 1,3‐propanesultone to yield the corresponding polysulfoproylbetaine derivatives. Those copolymers exhibiting the requisite aqueous solubility have been screened with respect to their antimicrobial activity against two common and notorious pathogens, namely Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). We show that certain copolymers do indeed exhibit antimicrobial activity. The extent of activity is related to the molecular characteristics of the materials such as the molar composition and structure of the hydrophobic comonomer. © 2006 Wiley Periodicals, Inc. The mathematical formulation of mass transfer in food processes is often based on the diffusion equation with the diffusion coeffcients considered as effective parameters. In some cases, more than one specie can be simultaneously diffusing in opposite directions. Therefore, the movement of one component could affect the movement of the remaining components. The aim of this work is to describe a mathematical procedure to obtain the diffusion coeffcients of different species that simultaneously diffuse in such a situation that each mass flux is affected by the existence of the others. Linear polyethers possess unusually high CO2 solubility and, hence, selectivity due to the presence of accessible ether linkages that can interact with the quadrupolar moment of CO2 molecules. In this work, membranes derived from crosslinked poly(propylene glycol) diacrylate (PPGda) oligomers differing in molecular weight (M), as well as PPGda nanocomposites containing either an organically-modified montmorillonite clay or a methacrylate-terminated fumed silica are investigated and compared with highly CO2-selective poly(ethylene glycol) diacrylate (PEGda) homopolymer and nanocomposite membranes previously reported. The rheological and permeation properties of PPGda depend sensitively on M, with the elastic modulus decreasing, but CO2 permeability and CO2/H2 selectivity increasing, with increasing M. Incorporation of either nanofiller into PPGda enhances the elastic modulus and reduces the gas permeability in the resultant nanocomposites without strongly affecting CO2/H2 selectivity. Blending PPGda and PEGda prior to chemical crosslinking yields binary membranes that exhibit intermediate gas-transport properties accurately described by a linear rule of mixtures. 相似文献

In this paper, the differential impedance analysis (DIA) has been applied to the study of the dielectric properties of hardened Portland cement paste. Two time constants are found in the impedance spectra obtained in the frequency region form 100 kHz to 15 MHz. One time constant has been attributed to the solid matrix and the other one to the liquid phase filling the pores. The effect of the cement paste-electrodes interface has been quantified using two different experimental set-ups. Measurements using direct contact between electrodes and cement paste have been compared with measurements using an air gap technique in which the specimen “floats” between the electrodes. The two referred time constants have been found in both types of measurements. The influence of drying on the dielectric parameters is also studied. 相似文献
