X-ray diffraction patterns of pyrolytically sprayed aluminium doped zinc oxide films have been recorded and X-ray line profile analysis studies have been carried out after correcting for instrumental broadening. Different microstructural parameters such as crystallite size, r.m.s. strain and dislocation density have been determined from the variance analysis of X-ray line profiles. Texture coefficient, the degree of preferred orientation of the crystallites and stacking fault have been estimated from the X-ray diffraction data. It is observed that the figure of merit as a transparent conductor depends on the dopant concentration and microstructural parameters of the films deposited under identical growth conditions. 相似文献

Wheat blast, caused by the fungus Magnaporthe oryzae Triticum (MoT) pathotype, is a devastating disease persistent in South America and Bangladesh. Since MoT generally fails to cause visual symptoms in wheat until the heading stage when the infection would have advanced, disease control by fungicide application solely based on the detection of visual symptoms is ineffective. To develop an accurate and sensitive method to detect MoT at the seedling and vegetative stages for disease control, we sequenced the genomes of two MoT isolates from Brazil and identified two DNA fragments, MoT-6098 and MoT-6099, that are present in the MoT genome but not in the genome of the rice-infecting Magnaporthe oryzae Oryzae (MoO) pathotype. Using polymerase chain reaction (PCR), we confirmed the specificity of the two markers in 53 MoT and MoO isolates from South America and Bangladesh. To test the efficiency of the two markers, we first established a loop-mediated isothermal amplification (LAMP) method to detect MoT at isothermal conditions, without the use of a PCR machine. Following this, we used the Cas12a protein and guide RNAs (gRNAs) to target the MoT-6098 and MoT-6099 sequences. The activated Cas12a showed indiscriminate single-stranded deoxyribonuclease (ssDNase) activity. We then combined target-dependent Cas12a ssDNase activation with recombinase polymerase amplification (RPA) and nucleic acid lateral flow immunoassay (NALFIA) to develop a method that accurately, sensitively, and cost-effectively detects MoT-specific DNA sequences in infected wheat plants. In recent years, bayberry decline disease has caused significant damage to the bayberry industry. In order to evaluate whether humic acid can be used to effectively control the disease, this research examined the nutritional growth and fruit quality of bayberry, soil physical and chemical properties, soil microbial community structure, and metabolites. Results indicated that the application of humic acid not only improved the vigor and fruit quality of diseased trees, but also increased the diversity of microbial communities in the rhizosphere soil. A great increase was observed in the relative abundance of bacterial genus Mycobacterium and Crossiella; fungal genus Fusarium and Coniosporium. In contrast, a significant decrease was observed in the relative abundance of bacterial genus Acidothermus, Bryobacter, Acidibacter, fungal genus of Geminibasidium and Mycena. Analysis of redundancies (RDA) for microbial communities and soil characteristics showed that the main four variables, including available nitrogen, phosphorus, potassium, and calcium, had a great effect on the composition of bacterial and fungal communities in bayberry rhizosphere soil at the genus level. The main four variables had a greater effect on bacterial communities than on fungal communities. In addition, ABC transporter, arginine and proline metabolism, galactose metabolism, and glutathione metabolism were significantly affected by humic acid, which changed the content of 81 metabolites including 58 significantly down-regulated metabolites such as isohexonic acid and carinitine, and 23 significantly up-regulated metabolites such as acidic acid, guaninosuccinate, lyxose, 2-monoolein, epicatechin, and pentonolactone. These metabolites also significantly correlated with rhizosphere soil microbiota at the phylum, order, and genus levels. In conclusion, the results demonstrated the role of humic acid on plant growth and fruit quality, as well as rhizosphere soil characteristics, microbiota, and secondary metabolites, which provides novel insights into the control of bayberry decline disease. 相似文献

The azo dye orange II is used extensively in the textile sector for coloring fabrics. High concentrations of it are released into aqueous environments through textile effluents. Therefore, its removal from textile wastewater and effluents is necessary. Herein, initially, we tested 11 bacterial strains for their capabilities in the degradation of orange II dye. It was revealed in the preliminary data that B. subtilis can more potently degrade the selected dye, which was thus used in the subsequent experiments. To achieve maximum decolorization, the experimental conditions were optimized whereby maximum degradation was achieved at: a 25 ppm dye concentration, pH 7, a temperature of 35 °C, a 1000 mg/L concentration of glucose, a 1000 mg/L urea concentration, a 666.66 mg/L NaCl concentration, an incubation period of 3 days, and with hydroquinone as a redox mediator at a concentration of 66.66 mg/L. The effects of the interaction of the operational factors were further confirmed using response surface methodology, which revealed that at optimum conditions of pH 6.45, a dye concentration of 17.07 mg/L, and an incubation time of 9.96 h at 45.38 °C, the maximum degradation of orange II can be obtained at a desirability coefficient of 1, estimated using the central composite design (CCD). To understand the underlying principles of degradation of the metabolites in the aliquot mixture at the optimized condition, the study steps were extracted and analyzed using GC-MS(Gas Chromatography Mass Spectrometry), FTIR(Fourier Transform Infrared Spectroscopy), 1H and carbon 13 NMR(Nuclear Magnetic Resonance Spectroscopy). The GC-MS pattern revealed that the original dye was degraded into o-xylene and naphthalene. Naphthalene was even obtained in a pure state through silica gel column isolation and confirmed using 1H and 13C NMR spectroscopic analysis. Phytotoxicity tests on Vigna radiata were also conducted and the results confirmed that the dye metabolites were less toxic than the parent dye. These results emphasize that B. subtilis should be used as a potential strain for the bioremediation of textile effluents containing orange II and other toxic azo dyes. 相似文献

Social media data are rapidly increasing and constitute a source of user opinions and tips on a wide range of products and services. The increasing availability of such big data on biased reviews and blogs creates challenges for customers and businesses in reviewing all content in their decision-making process. To overcome this challenge, extracting suggestions from opinionated text is a possible solution. In this study, the characteristics of suggestions are analyzed and a suggestion mining extraction process is presented for classifying suggestive sentences from online customers’ reviews. A classification using a word-embedding approach is used via the XGBoost classifier. The two datasets used in this experiment relate to online hotel reviews and Microsoft Windows App Studio discussion reviews. F1, precision, recall, and accuracy scores are calculated. The results demonstrated that the XGBoost classifier outperforms—with an accuracy of more than 80%. Moreover, the results revealed that suggestion keywords and phrases are the predominant features for suggestion extraction. Thus, this study contributes to knowledge and practice by comparing feature extraction classifiers and identifying XGBoost as a better suggestion mining process for identifying online reviews. 相似文献

Recently, Yang and Chang proposed an identity-based remote login scheme using elliptic curve cryptography for the users of mobile devices. We have analyzed the security aspects of the Yang and Chang's scheme and identified some security flaws. Also two improvements of the Yang and Chang's scheme have been proposed recently, however, it has been found that the schemes have similar security flaws as in the Yang and Chang's scheme. In order to remove the security pitfalls of the Yang and Chang and the subsequent schemes, we proposed an enhanced remote user mutual authentication scheme that uses elliptic curve cryptography and identity-based cryptosystem with three-way challenge-response handshake technique. It supports flawless mutual authentication of participants, agreement of session key and the leaked key revocation capability. In addition, the proposed scheme possesses low power consumption, low computation cost and better security attributes. As a result, the proposed scheme seems to be more practical and suitable for mobile users for secure Internet banking, online shopping, online voting, etc. 相似文献

Neisseria gonorrhoeae and Neisseria meningitidis are Gram-negative bacterial pathogens that infect human mucosal epithelia. Type IV pilus-mediated adherence of these bacteria is a crucial early event for establishment of infection. In this work, we show that the type IV pili transduce a signal into the eucaryotic host cell. Purified adherent pili, but not pili from a low binding mutant, trigger an increase in the cytosolic free calcium ([Ca2+]i) in target epithelial cells, a signal known to control many cellular responses. The [Ca2+]i increase was blocked by antibodies against CD46, a putative pilus receptor, suggesting a role for this protein in signal transduction. Pilus-mediated attachment was inhibited by depletion of host cell intracellular Ca2+ stores but not by removal of extracellular Ca2+. Further, kinase inhibition studies showed that pilus-mediated adherence is dependent on casein kinase II. In summary, these data reveal a novel function of the type IV pili, namely induction of signal transduction pathways in host cells. 相似文献
