The complexity of the transition from solution to gel for low molecular weight gelators in apolar solvents makes it incredibly difficult to assess the gel point. Since both nucleation and crystal growth occurs prior to the formation of a continuous three dimensional network numerous techniques, such as calorimetry and inflection point of the complex modulus, are invalid when probing the gel point. However, monitoring the frequency dependence of G′ and G″ illustrates the transition from a dilute solution to a weak gel which may be differentiated from the gel point observed by the decreasing value of the complex viscosity as a function of frequency. For 3% 12-hydroxystearic acid in mineral oil once the gelator nucleates the material behaves as a dilute solution, upon crystal growth (67 °C) we observe the true cross over point which is independent of frequency and finally a strong gel is formed at 64 °C where G′ and G″ are independent of frequency. 相似文献

The volatile compositions of 192 olive oil samples from five different European countries were investigated by PTR-MS sample headspace analysis. The mass spectra of all samples showed many masses with high abundances, indicating the complex VOC composition of olive oil. Three different PLS-DA models were fitted to the data to classify samples into ‘country’, ‘region’ and ‘district’ of origin, respectively. Correct classification rates were assessed by cross-validation. The first fitted model produced an 86% success rate in classifying the samples into their country of origin. The second model, which was fitted to the Italian oils only, also demonstrated satisfactory results, with 74% of samples successfully classified into region of origin. The third model, classifying the Italian samples into district of origin, yielded a success rate of only 52%. This lower success rate might be due to either the small class set, or to genuine similarities between olive oil VOC compositions on this tight scale. 相似文献

Carbon capture combined with utilization and storage has the potential to serve as a near-term option for CO(2) emissions reduction. CO(2) capture by carbon-based sorbents and CO(2) storage in geologic formations such as coal and shale both require a thorough understanding of the CO(2) adsorption properties in microporous carbon-based materials. Complex pore structures for natural organic materials, such as coal and gas shale, in addition to general carbon-based porous materials are modeled as a collection of independent, noninterconnected, functionalized graphitic slit pores with surface heterogeneities. Electronic structure calculations coupled with van der Waals-inclusive corrections have been performed to investigate the electronic properties of functionalized graphitic surfaces. With Bader charge analysis, electronic structure calculations can provide the initial framework comprising both the geometry and corresponding charge information required to carry out statistical modeling. Grand canonical Monte Carlo simulations were carried out to determine the adsorption isotherms for a given adsorbent-adsorbate interaction at temperature/pressure conditions relevant to carbon capture applications to focus on the effect of the surface functionalities. On the basis of the current work, oxygen-containing functional groups were predicted to enhance CO(2) adsorption in microporous carbon materials in the absence of water vapor, and the hydrated graphite was found to hinder CO(2) adsorption. 相似文献

The influence of feeding system and lactation period on the gross composition, macroelements (Ca, P, Mg, and Na), and trace elements (Zn, Fe, Cu, Mo, Mn, Se, and Co) of bovine milk was investigated. The feeding systems included outdoor grazing on perennial ryegrass pasture (GRO), outdoor grazing on perennial ryegrass and white clover pasture (GRC), and indoors offered total mixed ration (TMR). Sixty spring-calving Holstein Friesian dairy cows were assigned to 3 herds, each consisting of 20 cows, and balanced with respect to parity, calving date, and pre-experimental milk yield and milk solids yield. The herds were allocated to 1 of the 3 feeding systems from February to November. Milk samples were collected on 10 occasions over the period June 17 to November 26, at 2 or 3 weekly intervals, when cows were on average 119 to 281 d in lactation (DIL). The total lactation period was arbitrarily sub-divided into 2 lactation periods based on DIL, namely mid lactation, June 17 to September 9 when cows were 119 to 203 DIL; and late lactation, September 22 to November 26 when cows were 216 to 281 DIL. With the exception of Mg, Na, Fe, Mo, and Co, all other variables were affected by feeding system. The GRO milk had the highest mean concentrations of total solids, total protein, casein, Ca, and P. The TMR milk had the highest concentrations of lactose, Cu, and Se, and lowest level of total protein. The GRC milk had levels of lactose, Zn, and Cu similar to those of GRO milk, and concentrations of TS, Ca, and P similar to those of TMR milk. Lactation period affected all variables, apart from the concentrations of Fe, Cu, Mn, and Se. On average, the proportion (%) of total Ca, P, Zn, Mn, or Se that sedimented with the casein on high-speed ultracentrifugation at 100,000 × g was ≥60%, whereas that of Na, Mg, or Mo was ≤45% total. In the current study, we characterized 137 Lactococcus lactis bacteriophages that had been isolated between 1997 and 2012 from whey samples obtained from industrial facilities located in 16 countries. Multiplex PCR grouping of these 137 phage isolates revealed that the majority (61.31%) belonged to the 936 group, with the remainder belonging to the P335 and c2 groups (23.36 and 15.33%, respectively). Restriction profile analysis of phage genomic DNA indicated a high degree of genetic diversity within this phage collection. Furthermore, based on a host-range survey of the phage collection using 113 dairy starter strains, we showed that the c2-group isolates exhibited a broader host range than isolates of the 936 and P335 groups. 相似文献

1 Scope
Shellfish allergy is an increasing global health priority, frequently affecting adults. Molluscs are an important shellfish group causing food allergy but knowledge of their allergens and cross‐reactivity is limited. Optimal diagnosis of mollusc allergy enabling accurate advice on food avoidance is difficult. Allergens of four frequently ingested Asia‐Pacific molluscs are characterized: Sydney rock oyster (Saccostrea glomerata), blue mussel (Mytilus edulis), saucer scallop (Amusium balloti), and southern calamari (Sepioteuthis australis), examining cross‐reactivity between species and with blue swimmer crab tropomyosin, Por p 1.2 Methods and results
IgE ELISA showed that cooking increased IgE reactivity of mollusc extracts and basophil activation confirmed biologically relevant IgE reactivity. Immunoblotting demonstrated strong IgE reactivity of several proteins including one corresponding to heat‐stable tropomyosin in all species (37–40 kDa). IgE‐reactive Sydney rock oyster proteins were identified by mass spectrometry, and the novel major oyster tropomyosin allergen was cloned, sequenced, and designated Sac g 1 by the IUIS. Oyster extracts showed highest IgE cross‐reactivity with other molluscs, while mussel cross‐reactivity was weakest. Inhibition immunoblotting demonstrated high cross‐reactivity between tropomyosins of mollusc and crustacean species.3 Conclusion

Our objective was to evaluate how sampling strategies (i.e., how many cows to sample and which animals to include) used in 4 dairy cattle welfare assessment programs affect the classification of dairy farms relative to thresholds of acceptability for animal-based measures. We predicted that classification performance would improve when more cows were sampled and when selecting from all lactating cows versus when some pens were excluded. On 38 freestall farms, we assessed all 12,375 cows for lameness, injuries on the tarsal (hock) and carpal joints, and body condition score and calculated the farm-level prevalence for each measure. Based on approaches used in the industry, we evaluated 6 sampling strategies generated using formulas with precision (d) of 15, 10, or 5% applied to either a single high-producing pen or all lactating cows; an additional sample was included with d = 10% applied to the entire herd, selecting lactating cows in proportion to their representation in the herd. For each sampling strategy, cow records were selected randomly (in 10,000 replicates) to calculate prevalence. The strategy of assessing all cows in the high-producing pen was also compared. Farms were classified as meeting (below) or failing to meet (above) thresholds of ≤15% moderate lameness; ≤20% moderate carpal or hock injuries; <10, <5, and ≤1% severe lameness; or injuries on the carpus or hock; and <5, <3, <1, or 0% thin cows. For each measure and threshold, we calculated pooled percent agreement, kappa, sensitivity, specificity, and positive and negative predictive value for each sampling strategy using true prevalence as the gold standard for herd classification. Across measures and thresholds, classification performance increased with the number of cows sampled [i.e., when narrower precision values (d = 5 vs. 10 vs. 15%) were used in the sample size calculation]. Because narrower precision values can dramatically increase sample size, assessment programs may need to consider both feasibility and the degree of misclassification they will accept. Applying the formula directly to lactating cows performed better than applying it to the entire herd and then selecting lactating cows in proportion to their representation in the herd. Farm classifications were similar whether cows in the hospital pen were included or excluded from the sample. Selecting all cows from the high-producing pen resulted in classifications similar to when including all lactating cows, suggesting that assessing cows from the high-producing pen may serve as an acceptable proxy for all lactating cows on the farm. 相似文献
