Fetal stress is known to increase susceptibility to cardiometabolic diseases and hypertension in adult age in a process known as fetal programming. This study investigated the relationship between vascular RAS, oxidative damage and remodeling in fetal programming. Six-month old Sprague-Dawley offspring from mothers that were fed ad libitum (CONTROL) or with 50% intake during the second half of gestation (maternal undernutrition, MUN) were used. qPCR or immunohistochemistry were used to obtain the expression of receptors and enzymes. Plasma levels of carbonyls were measured by spectrophotometry. In mesenteric arteries from MUN rats we detected an upregulation of ACE, ACE2, AT1 receptors and NADPH oxidase, and lower expression of AT2, Mas and MrgD receptors compared to CONTROL. Systolic and diastolic blood pressure and plasma levels of carbonyls were higher in MUN than in CONTROL. Vascular morphology evidenced an increased media/lumen ratio and adventitia/lumen ratio, and more connective tissue in MUN compared to CONTROL. In conclusion, fetal undernutrition indices RAS alterations and oxidative damage which may contribute to the remodeling of mesenteric arteries, and increase the risk of adverse cardiovascular events and hypertension.  相似文献

The NAD+‐dependent deacetylases, namely sirtuins, are involved in the regulation of a variety of biological processes such as gene silencing, DNA repair, longevity, metabolism, apoptosis, and development. An enzyme from the parasite Leishmania infantum that belongs to this family, LiSIR2RP1, is a NAD+‐dependent tubulin deacetylase and an ADP‐ribosyltransferase. This enzyme's involvement in L. infantum virulence and survival underscores its potential as a drug target. Our search for selective inhibitors of LiSIR2RP1 has led, for the first time, to the identification of the antiparasitic and anticancer bisnaphthalimidopropyl (BNIP) alkyl di‐ and triamines (IC50 values in the single‐digit micromolar range for the most potent compounds). Structure–activity studies were conducted with 12 BNIP derivatives that differ in the length of the central alkyl chain, which links the two naphthalimidopropyl moieties. The most active and selective compound is the BNIP diaminononane (BNIPDanon), with IC50 values of 5.7 and 97.4 μM against the parasite and human forms (SIRT1) of the enzyme, respectively. Furthermore, this compound is an NAD+‐competitive inhibitor that interacts differently with the parasite and human enzymes, as determined by docking analysis, which might explain its selectivity toward the parasitic enzyme.  相似文献

Since 2003 India has been actively promoting the cultivation of Jatropha on unproductive and degraded lands (wastelands) for the production of biodiesel suitable as transportation fuel. In this paper the life cycle energy balance, global warming potential, acidification potential, eutrophication potential and land use impact on ecosystem quality is evaluated for a small scale, low-input Jatropha biodiesel system established on wasteland in rural India. In addition to the life cycle assessment of the case at hand, the environmental performance of the same system expanded with a biogas installation digesting seed cake was quantified. The environmental impacts were compared to the life cycle impacts of a fossil fuel reference system delivering the same amount of products and functions as the Jatropha biodiesel system under research. The results show that the production and use of Jatropha biodiesel triggers an 82% decrease in non-renewable energy requirement (Net Energy Ratio, NER = 1.85) and a 55% reduction in global warming potential (GWP) compared to the reference fossil-fuel based system. However, there is an increase in acidification (49%) and eutrophication (430%) from the Jatropha system relative to the reference case. Although adding biogas production to the system boosts the energy efficiency of the system (NER = 3.40), the GWP reduction would not increase (51%) due to additional CH4 emissions. For the land use impact, Jatropha improved the structural ecosystem quality when planted on wasteland, but reduced the functional ecosystem quality. Fertilizer application (mainly N) is an important contributor to most negative impact categories. Optimizing fertilization, agronomic practices and genetics are the major system improvement options.  相似文献

Projects are inherently uncertain and face unexpected events, from small changes in scope to unforeseen client’s bankruptcy. This paper studies how project managers respond to such events and how successful and unsuccessful responses differ from the perspective of the practitioner. We analysed 44 unexpected events faced by 22 experienced project managers in defence and defence-related organisations. The project managers compared two unexpected events that they faced, one that they considered the response successful and the other unsuccessful. We identified three pillars supporting successful responses to unexpected events: (1) responsive and functioning structure at the organisational level, (2) good interpersonal relationship at the group level and (3) competent people at the individual level. The events and respective responses analysed suggest that improvement in project management can be achieved by better managing these three pillars, allowing project and programme managers to “create their own luck”.  相似文献

Based on the mood-behavior-model (Gendolla, 2000), this study tested the idea that moods only have effects on effort mobilization in settings that directly call for this and in which people can thus use their moods as task-relevant information. Fifty university students were randomly assigned to a 2 (Mood: negative vs. positive) × 2 (Memorizing: intentional vs. incidental) × 2 (Time: mood induction vs. task performance) mixed model design. Effort mobilization was operationalized as systolic blood pressure (SBP) reactivity. As expected, in the intentional-memorizing condition, SBP reactivity was stronger in a negative mood than in a positive mood. The mining, milling and processing of uranium and thorium bearing minerals may result in radiation doses to workers. The control of occupational exposures from these natural sources of radiation imposes a challenge to regulators and radiation protection advisers. A survey pilot programme, which included six mines in Brazil and a monazite plant, was established, consisting of the collection and analysis of concentrations of uranium, thorium and polonium in urine, faeces and air samples. Results from workers were compared to background data from their families living in the same area and from residents from the population of Rio de Janeiro. Positive exposure results were identified among the coal miners, the niobium miners and the monazite sand workers. Difficulties in the application of internal dosimetry programmes are discussed in relation to the control of NORM workers.  相似文献

The biofouling Asian clam Corbicula fluminea greatly affects freshwater-dependent facilities in Europe and North America, including in the Great Lakes region. As chlorination has become increasingly restricted, finding alternative control agents is a priority. Due to the species' epifaunal nature, the body of knowledge on Dreissena polymorpha is larger than that on the Asian clam, and there is a tendency to assume that mitigation methods should work similarly for both species. However, this generalisation is inaccurate, and the optimisation of Asian clam control relies on species-specific toxicological data. This paper reports information on the potential of three candidate biocides for C. fluminea control: (i) the cationic polydiallyldimethylammonium chloride (polyDADMAC), (ii) potassium chloride and (iii) aluminum sulphate. While these chemicals may be employed in a range of contexts, they are particularly suitable for the highly regulated drinking water industry. LT50 values ranging from 284.3 h, for polyDADMAC applied at 10 mg/L, to 855.1 h, for an aluminum sulphate concentration of 11 mg/L, were obtained. Ecotoxicological standard tests with Pseudokirchneriella subcapitata and Daphnia magna suggested that, amongst the three biocides, potassium chloride is the one representing lower potential environmental hazard (with, for example, 48 h-EC50 higher than 1 g/L for Daphnia immobilisation, which compares to values of 20.2 and 112.5 mg/L for polyDADMAC and aluminum sulphate, respectively). The three chemicals are promising control agents at dosages compatible with waterworks' operational requirements with polyDADMAC having the highest biocidal activity, but also posing more environmental risks.  相似文献

This study aimed to elaborate a set of measures to control vitamin C losses in vegetables prepared at a food service (FS). Vegetables were randomly selected for vitamin C analysis by high performance liquid chromatography (HPLC) after delivery of raw material and during distribution for consumption of the food prepared. Some principles underlying the Hazards Analysis and Critical Control Points (HACCP) were applied to identify the Nutritional Control Points (NCP) for vitamin C losses. A Nutritional Control Measure (NCM) was adopted for each NCP as well as the monitoring criteria. The vegetables were again collected for vitamin C analysis after adoption of the NCMs. The results indicated reduction of vitamin C losses with NCM adoption.  相似文献

The aim of this work was to investigate the adhesion of Listeria monocytogenes ATCC 15313 to glass, granite, marble, polypropylene from a bowl (PPb), polypropylene from a cutting board (PPcb) and stainless steel (SS), which are materials commonly used in kitchens. Marble and granite were chosen because they are applied as kitchen bench covers and pavements in many countries and there are no literature reports on their behaviour in terms of microbial adhesion. The effect of surface hydrophobicity and roughness on the adhesion process was also analysed. The results showed that the highest extent of adhesion of L. monocytogenes occurred to stainless steel, followed by glass and in less extent to the other materials studied. However, it was not possible to establish a correlation between surface hydrophobicity or roughness and the extent of adhesion of L. monocytogenes . The adherence of L. monocytogenes should be dependent on other factors, like the presence of exopolymers and surface charge.  相似文献
