Chagas disease is caused by the protozoan parasite Trypanosoma cruzi and affects over 6 million people worldwide. Development of new drugs to treat this disease remains a priority since those currently available have variable efficacy and frequent adverse effects, especially during the long regimens required for treating the chronic stage of the disease. T. cruzi modulates the host cell-metabolism to accommodate the cell cytosol into a favorable growth environment and acquire nutrients for its multiplication. In this study we evaluated the specific anti-T. cruzi activity of nine bio-energetic modulator compounds. Notably, we identified that 17-DMAG, which targets the ATP-binding site of heat shock protein 90 (Hsp90), has a very high (sub-micromolar range) selective inhibition of the parasite growth. This inhibitory effect was also highly potent (IC50 = 0.27 μmol L−1) against the amastigote intracellular replicative stage of the parasite. Moreover, molecular docking results suggest that 17-DMAG may bind T. cruzi Hsp90 homologue Hsp83 with good affinity. Evaluation in a mouse model of chronic T. cruzi infection did not show parasite growth inhibition, highlighting the difficulties encountered when going from in vitro assays onto preclinical drug developmental stages.  相似文献

HTML is being massively used as an interface to provide services to users. Web developers are producing and changing sites at a high pace while trying to support the latest HTML standards. In this context, it is common to find websites that do not comply with the standards and fail to be correctly processed by browsers. Considering this dynamic environment and the increasingly large diversity of browsers with frequent updates, the appearance of problems in web pages is a common, sometimes severe, and hard‐to‐track problem. In this short communication, we describe the initial design of an approach that will be used to obtain information regarding the characteristics of HTML documents on the Web and extract indicators of representative errors made by their developers. Preliminary results show nearly 90% of the pages analyzed having at least one type of error and the prevalence of a small number of error types.  相似文献

In the last decades, advances in interactive information technologies have facilitated collaborative fiction writing, which has become widespread and large-scale. This paper proposes a framework to analyze collaborative storytelling systems, made of a set of parameters divided into six conceptual areas. Four of them relate to the systems and two (process and output) to the results of the collaboration. Through this framework we can study more precisely these different factors of the systems, their interplay, and how they impact the creators’ performance. We also present a controlled extended-duration field study on collaborative storytelling, and we use this framework to comparatively analyze these observations and other relevant experiences in the field of co-creation of shared narrative spaces. As a result, we propose a human-information interaction model for collaborative narrative systems, intended to better support co-creation and address the barriers of this kind of systems turning them into new opportunities for collaboration.  相似文献

Composite materials are being used with notable effectiveness to increase and upgrade the flexural load carrying capacity of reinforced concrete (RC) members. Near-surface mounted (NSM) is one of the most promising strengthening techniques, based on the use of carbon fiber-reinforced polymer (CFRP) laminates. According to NSM, the laminates are fixed with epoxy based adhesive into slits opened into the concrete cover on the tension face of the elements to strength. Laboratory tests have shown that the NSM technique is an adequate strengthening strategy to increase the flexural resistance of RC slabs. However, in RC slabs of low concrete strength, the increase of the flexural resistance that NSM can provide is limited by the maximum allowable compressive strain in the compressed part of the slab, in order to avoid concrete crushing. This restriction reduces the effectiveness of the strengthening, thus limiting the use of the NSM technique. A new thin layer of concrete bonded to the existing concrete at the compressed region is suitable to overcome this limitation. Volumetric contraction due to shrinkage and thermal effects can induce uncontrolled cracking in the concrete of this thin layer. Adding steel fibers to concrete [steel fiber-reinforced concrete (SFRC)], the postcracking residual stress can be increased in order to prevent the formation of uncontrolled crack patterns. In the present work, the combined strengthening strategy, a SFRC overlay and NSM CFRP laminates, was applied to significantly increase the flexural resistance of existing RC slabs. Experimental results of four-point bending tests, carried out in unstrengthened and strengthened concrete slab strips, are presented and analyzed.  相似文献

This work describes the determination of aluminum in tea infusions and iced tea by tungsten coil atomic emission spectrometry (WCAES). The instrumentation is simple, inexpensive, and potentially portable. It uses a tungsten filament extracted from 150 W, 15 V commercially available slide projector light bulbs as electrothermal atomizer. A simple power supply that provides a constant potential is used to heat the coil. Tea infusions were directly analyzed by application of an optimized heating program. In order to avoid matrix interferences, the iced tea samples were diluted with an oxidizing mixture composed of hydrogen peroxide and nitric acid and also analyzed by a tailored heating program. Aluminum was quantified in all samples, and the results were in agreement with those obtained by inductively coupled plasma optical emission spectrometry at a 95 % confidence level. The limit of detection and the relative standard deviation for a solution containing 2.5 mg L?1 Al (n?=?10) were determined, and the values were 0.09 mg L?1 and 5.1 %, respectively.  相似文献

The emissive power (E) of a flame depends on the size of the fire and the type of fuel. In fact, it changes significantly over the flame surface: the zones of luminous flame have high emittance, while those covered by smoke have low E values. The emissive power of each zone (that is, the luminous or clear flame and the non-luminous or smoky flame) and the portion of total flame area they occupy must be assessed when a two-zone model is used. In this study, data obtained from an experimental set-up were used to estimate the emissive power of fires and its behaviour as a function of pool size. The experiments were performed using gasoline and diesel oil as fuel. Five concentric circular pools (1.5, 3, 4, 5 and 6m in diameter) were used. Appropriate instruments were employed to determine the main features of the fires. By superimposing IR and VHS images it was possible to accurately identify the luminous and non-luminous zones of the fire. Mathematical expressions were obtained that give a more accurate prediction of E(lum), E(soot) and the average emissive power of a fire as a function of its luminous and smoky zones. These expressions can be used in a two-zone model to obtain a better prediction of the thermal radiation. The value of the radiative fraction was determined from the thermal flux measured with radiometers. An expression is also proposed for estimating the radiative fraction.  相似文献

Computational fluid dynamics (CFD) is increasingly used to analyze wind turbines, and the next logical step is to develop CFD‐based optimization to enable further gains in performance and reduce model uncertainties. We present an aerodynamic shape optimization framework consisting of a Reynolds‐averaged Navier Stokes solver coupled to a numerical optimization algorithm, a geometry modeler, and a mesh perturbation algorithm. To efficiently handle the large number of design variables, we use a gradient‐based optimization technique together with an adjoint method for computing the gradients of the torque coefficient with respect to the design variables. To demonstrate the effectiveness of the proposed approach, we maximize the torque of the NREL VI wind turbine blade with respect to pitch, twist, and airfoil shape design variables while constraining the blade thickness. We present a series of optimization cases with increasing number of variables, both for a single wind speed and for multiple wind speeds. For the optimization at a single wind speed performed with respect to all the design variables (1 pitch, 11 twist, and 240 airfoil shape variables), the torque coefficient increased by 22.4% relative to the NREL VI design. For the multiple‐speed optimization, the torque increased by an average of 22.1%. Depending on the CFD mesh size and number of design variables, the optimization time ranges from 2 to 24h when using 256 cores, which means that wind turbine designers can use this process routinely. Firefly luciferase catalyzes the synthesis of H2O2 from the same substrates as the bioluminescence reaction: ATP and luciferin (D-LH2). About 80% of the enzyme-bound intermediate D-luciferyl adenylate (D-LH2-AMP) is oxidized into oxyluciferin, and a photon is emitted during this reaction. The enzyme pathway responsible for the generation of H2O2 is a side reaction in which D-LH2-AMP is oxidized into dehydroluciferyl adenylate (L-AMP). Like the bioluminescence reaction, the luciferase-catalyzed synthesis of H2O2 and L-AMP is a stereospecific process, involving only the natural D enantiomer. However, the intramolecular electron transfer postulated as essential to the light emission process is not involved in this side reaction.  相似文献
