Porous carbon materials represent prospective materials for absorbers, filters, and electronic applications. Carbon fibers with high surface areas can be produced from polyacrylonitrile and spun as thin fibers from solution. The resulting polymer fibers are first stabilized to obtain conjugated ribbons and then carbonized to graphitic structures in a second high‐temperature step in an inert atmosphere. In this study, we investigated a previously described fast laser‐heating process that delivered fibers with a higher crystallinity and surface area compared to the thermally carbonized fibers. In a subsequent KOH‐activation step, the crystalline domains were exfoliated, and the surface of the fibers became macroporous. This led to a reduced specific surface area but a higher capacitance compared to thermally carbonized nanofibers. The artificial regulation of proteins by light is an emerging subdiscipline of synthetic biology. Here, we used this concept to photocontrol both catalysis and allostery within the heterodimeric enzyme complex imidazole glycerol phosphate synthase (ImGP‐S). ImGP‐S consists of the cyclase subunit HisF and the glutaminase subunit HisH, which is allosterically stimulated by substrate binding to HisF. We show that a light‐sensitive diarylethene (1,2‐dithienylethene, DTE)‐based competitive inhibitor in its ring‐open state binds with low micromolar affinity to the cyclase subunit and displaces its substrate from the active site. As a consequence, catalysis by HisF and allosteric stimulation of HisH are impaired. Following UV‐light irradiation, the DTE ligand adopts its ring‐closed state and loses affinity for HisF, restoring activity and allostery. Our approach allows for the switching of ImGP‐S activity and allostery during catalysis and appears to be generally applicable for the light regulation of other multienzyme complexes. 相似文献

Iterative type I polyketide synthases (PKSs) from fungi are multifunctional enzymes that use their active sites repeatedly in a highly ordered sequence to assemble complex natural products. A phytotoxic macrolide with anticancer properties, 10,11‐dehydrocurvularin (DHC), is produced by cooperation of a highly reducing (HR) iterative PKS and a non‐reducing (NR) iterative PKS. We have identified the DHC gene cluster in Alternaria cinerariae, heterologously expressed the active HR PKS (Dhc3) and NR PKS (Dhc5) in yeast, and compared them to corresponding proteins that make DHC in Aspergillus terreus. Phylogenetic analysis and homology modeling of these enzymes identified variable surfaces and conserved motifs that are implicated in product formation. 相似文献

Isothermal oscillations in the rate of decomposition of N2O were studied on an over-exchanged Cu-ZSM-5 catalyst by mass spectroscopy and in situ transient FTIR. Oscillations in the production of O2 and N2 were observed to occur in a temperature range of 410–490°C at a total pressure of 1.0 Torr pure N2O. FTIR has provided the first spectroscopic evidence that surface nitrate species are present under oscillatory conditions. This study confirmed a previously proposed model that predicts a slow build-up of surface nitrates, followed by a rapid nitrate decomposition coupled with an increase in the rate of N2O decomposition. The IR signature of the surface nitrates suggests they are monodentate nitrate species bound to Cu2+ ions. Temperature-programmed desorption studies reveal a strong correlation between the stability of the surface nitrate species and the temperature range in which oscillations occur. 相似文献

Free-standing polyelectrolyte multilayer membranes have been formed by the layer-by-layer technique using a dip-coating apparatus. The polymer-chain mobility has been studied by 1H relaxation in the rotating frame T 1rho NMR with 13C chemical shift resolution. For each of the individual polymers a single relaxation component has been observed for all resolved signals. In the multilayer a significantly different relaxation time T 1rho has been observed with a minor second component. The interaction between the oppositely charged polyelectrolytes influences the molecular mobility. 相似文献

Lasioglossin III (LL-III) is a cationic antimicrobial peptide derived from the venom of the eusocial bee Lasioglossum laticeps. LL-III is extremely toxic to both Gram-positive and Gram-negative bacteria, and it exhibits antifungal as well as antitumor activity. Moreover, it shows low hemolytic activity, and it has almost no toxic effects on eukaryotic cells. However, the molecular basis of the LL-III mechanism of action is still unclear. In this study, we characterized by means of calorimetric (DSC) and spectroscopic (CD, fluorescence) techniques its interaction with liposomes composed of a mixture of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and 1-palmitoyl-2-oleoyl-sn-glycero-3-rac-phosphoglycerol (POPG) lipids as a model of the negatively charged membrane of pathogens. For comparison, the interaction of LL-III with the uncharged POPC liposomes was also studied. Our data showed that LL-III preferentially interacted with anionic lipids in the POPC/POPG liposomes and induces the formation of lipid domains. Furthermore, the leakage experiments showed that the peptide could permeabilize the membrane. Interestingly, our DSC results showed that the peptide-membrane interaction occurs in a non-disruptive manner, indicating an intracellular targeting mode of action for this peptide. Consistent with this hypothesis, our gel-retardation assay experiments showed that LL-III could interact with plasmid DNA, suggesting a possible intracellular target. 相似文献

New developments in the field of thermal spraying systems (increased particle velocities, enhanced process stability) are leading to improved coatings. Innovations in the field of feedstock materials are supporting this trend. The combination of both has led to a renaissance of Fe-based feedstocks. Using modern APS or HVOF systems, it is now possible to compete with classical materials for wear and corrosion applications like Ni-basis or metal-matrix composites. This study intends to give an analysis of the in-flight particle and spray jet properties achievable with two different modern thermal spraying systems using Fe-based powders. The velocity fields are measured with the Laser Doppler Anemometry. Resulting coatings are analyzed and a correlation with the particle in-flight properties is given. The experiments are accompanied by computational fluid dynamics simulations of spray jet and particle velocities, leading to a comprehensive analysis of the achievable particle properties with state-of-the-art HVOF and APS systems. 相似文献

Mutations within the glucocorticoid receptor (GR) gene locus lead to glucocorticoid resistance which is characterized by several clinical symptoms such as adrenal gland hyperplasia and salt-sensitive hypertension, although the underlying mechanisms are still unknown. We studied GR haploinsufficient (GR+/−) Sprague Dawley rats which, on a standard diet, showed significantly increased plasma aldosterone and corticosterone levels and an adrenocortex hyperplasia accompanied by a normal systolic blood pressure. Following a high salt diet, these rats developed salt-sensitive hypertension and maintained elevated enzyme-soluble epoxide hydrolase (sEH) in adrenal glands, while sEH was significantly decreased in wild-type rats. Furthermore, GR+/− rats showed dysregulation of the equilibrated linoleic and arachidonic acid pathways, with a significant increase of less active metabolites such as 8,9-DiHETrE. In Sprague Dawley rats, GR haploinsufficiency induced steroid disturbances, which provoked hypertension only in combination with high salt intake, which was accompanied by disturbances in sEH and fatty acid metabolism. Our results suggest that sEH inhibition could be a potential target to treat hypertension in patients with GR haploinsufficiency. 相似文献

Recent research projects show that retained austenite, if stabilized by nitrogen, has a positive influence on the fatigue strength of work pieces. The combined diffusion profile of carbon and nitrogen applied in a carbonitriding process plays a major role, besides the process temperature. Yet today, only the carbon potential is somehow controlled and even this is not easy to achieve. This paper will present a new system able to measure and control both the carbon potential and the nitrogen potential independently. The knowledge of the activities of nitrogen and carbon in iron and the effect of alloying elements on such activities as well as the solubilities offers a way to apply the potentials on real steels. 相似文献
