The development of a computational multibody knee model able to capture some of the fundamental properties of the human knee articulation is presented. This desideratum is reached by including the kinetics of the real knee articulation. The research question is whether an accurate modeling of the condyle contact in the knee will lead to reproduction of the complex combination of flexion/extension, abduction/adduction, and tibial rotation observed in the real knee. The model is composed by two anatomic segments, the tibia and the femur, whose characteristics are functions of the geometric and anatomic properties of the real bones. The biomechanical model characterization is developed under the framework of multibody systems methodologies using Cartesian coordinates. The type of approach used in the proposed knee model is the joint surface contact conditions between ellipsoids, representing the two femoral condyles, and points, representing the tibial plateau and the menisci. These elements are closely fitted to the actual knee geometry. This task is undertaken by considering a parameter optimization process to replicate experimental data published in the literature, namely that by Lafortune and his coworkers in 1992. Then kinematic data in the form of flexion/extension patterns are imposed on the model corresponding to the stance phase of the human gait. From the results obtained, by performing several computational simulations, it can be observed that the knee model approximates the average secondary motion patterns observed in the literature. Because the literature reports considerable inter-individual differences in the secondary motion patterns, the knee model presented here is also used to check whether it is possible to reproduce the observed differences with reasonable variations of bone shape parameters. This task is accomplished by a parameter study, in which the main variables that define the geometry of condyles are taken into account. It was observed that the data reveal a difference in secondary kinematics of the knee in flexion versus extension. The likely explanation for this fact is the elastic component of the secondary motions created by the combination of joint forces and soft tissue deformations. The proposed knee model is, therefore, used to investigate whether this observed behavior can be explained by reasonable elastic deformations of the points representing the menisci in the model. 相似文献

We present a bead-based approach to microfluidic polymerase chain reaction (PCR), enabling fluorescent detection and sample conditioning in a single microchamber. Bead-based PCR, while not extensively investigated in microchip format, has been used in a variety of bioanalytical applications in recent years. We leverage the ability of bead-based PCR to accumulate fluorescent labels following DNA amplification to explore a novel DNA detection scheme on a microchip. The microchip uses an integrated microheater and temperature sensor for rapid control of thermal cycling temperatures, while the sample is held in a microchamber fabricated from (poly)dimethylsiloxane and coated with Parylene. The effects of key bead-based PCR parameters, including annealing temperature and concentration of microbeads in the reaction mixture, are studied to achieve optimized device sensitivity and detection time. The device is capable of detecting a synthetically prepared section of the Bordetella pertussis genome in as few as 10 temperature cycles with times as short as 15?min. We then demonstrate the use of the procedure in an integrated device; capturing, amplifying, detecting, and purifying template DNA in a single microfluidic chamber. These results show that this method is an effective method of DNA detection which is easily integrated in a microfluidic device to perform additional steps such as sample pre-conditioning. 相似文献

This paper explores the issues involved in moving from ethnographic explorations of work in context to a practical contribution to system design. It does so using the example of an interdisciplinary research project involving sociologists and computer scientists in the domain of air traffic control systems. It forms a pair with another paper (Sommerville et al., 1992) exploring these questions from the perspective of our computer science partners. We characterise ethnography as a research method, and consider the differences between undertaking it for strictly sociological or anthropological purposes by contrast with interdisciplinary and design purposes. We summarise some of our results in ethnographic explications of the work of air traffic controllers, and the sociality which it manifests. We describe the dialogues involved in rendering these observations informative for systems design, and the mutual translations implied in attempting to reconcile sociological with software engineering questions about supporting the work. We conclude by specifying some features of cooperative work which an engineering approach is in danger of overlooking; the ways, and limits, in which ethnographers can form a bridge between users and designers; and some of the conflicts of interest entrained in generating technical change.The research is funded by the UK MRC/SERC/ESRC Cognitive Science/HCI Initiative with the title Social Analysis of Control Systems for HCI Design, grant number SPG 8931598. Bratu's problem, which is the nonlinear eigenvalue equationu+ exp(u)=0 withu=0 on the walls of the unit square and as the eigenvalue, is used to develop several themes on applications of Chebyshev pseudospectral methods. The first is the importance ofsymmetry: because of invariance under the C4 rotation group and parity in bothx andy, one can slash the size of the basis set by a factor of eight and reduce the CPU time by three orders of magnitude. Second, the pseudospectral method is ananalytical as well as a numerical tool: the simple approximation3.2A exp(–0.64A), whereA is the maximum value ofu(x, y), is derived via collocation with but a single interpolation point, but is quantitatively accurate for small and moderateA. Third, the Newton-Kantorovich/Chebyshev pseudospectral algorithm is so efficient that it is possible to compute good numerical solutions—five decimal places—on amicrocomputer inbasic. Fourth, asymptotic estimates of the Chebyshev coefficients can be very misleading: the coefficients for moderately or strongly nonlinear solutions to Bratu's equations fall off exponentially rather than algebraically withv untilv is so large that one has already obtained several decimal places of accuracy. This paper introduces a generic model for interactive design of constructed facilities. The proposed methodology synthesizes structural problem parameters and conditions in terms of load environment, structural properties, and function of the structure into a hierarchy of possible failure modes. Design evaluations are made in terms of this behavior hierarchy. The user has a high degree of control over the design process and may move at will to any of the design tasks. In normal vision, eye movements cause the image to move on the retina. Special apparatus can stabilize the image on the retina so it cannot move, and vision fades away. Previous methods for stabilizing the image were either optical systems or complex computer-controlled feedback systems. A simpler feedback system is presented which detects eye motion and rotates a mirror through which the target is viewed, to exactly compensate for eye motion. Unlike previous optical systems, this new system has no lenses in either the viewing or the image-forming path to limit the spatial resolution. 相似文献
