Griffonia simplicifolia leaf lectin II (GSII), a plant defense protein against certain insects, consists of an N-acetylglucosamine (GlcNAc)-binding large subunit with a small subunit having sequence homology to class III chitinases. Much of the insecticidal activity of GSII is attributable to the large lectin subunit, because bacterially expressed recombinant large subunit (rGSII) inhibited growth and development of the cowpea bruchid, Callosobruchus maculatus (F). Site-specific mutations were introduced into rGSII to generate proteins with altered GlcNAc binding, and the different rGSII proteins were evaluated for insecticidal activity when added to the diet of the cowpea bruchid. At pH 5.5, close to the physiological pH of the cowpea bruchid midgut lumen, rGSII recombinant proteins were categorized as having high (rGSII, rGSII-Y134F, and rGSII-N196D mutant proteins), low (rGSII-N136D), or no (rGSII-D88N, rGSII-Y134G, rGSII-Y134D, and rGSII-N136Q) GlcNAc-binding activity. Insecticidal activity of the recombinant proteins correlated with their GlcNAc-binding activity. Furthermore, insecticidal activity correlated with the resistance to proteolytic degradation by cowpea bruchid midgut extracts and with GlcNAc-specific binding to the insect digestive tract. Together, these results establish that insecticidal activity of GSII is functionally linked to carbohydrate binding, presumably to the midgut epithelium or the peritrophic matrix, and to biochemical stability of the protein to digestive proteolysis.  相似文献

At low rotation speeds of a rotating disc electrode the diffusion-controlled inhibition of zinc deposition by lead acetate or tetrabutylammonium bromide emphasizes the existence of multiple steady states. With both zinc and nickel deposition, low frequency impedance measurements reveal an additional inductive loop associated with a diffusion-controlled inhibition of electrodeposition. From the relaxation time constant it has been concluded that this loop arises from a coupling between the inhibitor diffusion and interfacial processes slower than diffusion and involving, at least for zinc deposition, the slow renewal of active growth sites.  相似文献

Generation of wear debris is the principal obstacle limiting the durability of ultra-high molecular weight polyethylene (UHMWPE) in biomedical applications. Aiming to enhance UHMWPE wear resistance, surface modification with swift heavy ion irradiation (SHI) appears as a potential and attractive methodology. Contrary to ion implantation techniques, the swift heavy ions range can reach tens to hundreds microns and its extremely high linear energy is able to induce effective chemical modifications using low fluence values. Nano-wear performance and surface mechanical properties of samples of pristine and SHI irradiated (using N2+ ions at 33 MeV and a fluence of 1 × 1012 ions/cm2) were characterized by depth sensing indentation (DSI) and scanning probe microscopy (SPM). It turned out that modifications induced by irradiation at the surface layers were successful to reduce nano-wear volume and creep deformation. These improvements were related to beneficial changes in hardness, elastic modulus, hardness to elastic modulus ratio and friction coefficient.  相似文献

Growth hormone and insulin-like growth factors (GH/IGF axis) regulate somatic growth in mammals and fish, although their action on metabolism is not fully understood in the latter. An intraperitoneal injection of extended-release recombinant bovine growth hormone (rbGH, Posilac®) was used in gilthead sea bream fingerlings and juveniles to analyse the metabolic response of liver and red and white muscles by enzymatic, isotopic and proteomic analyses. GH-induced lipolysis and glycogenolysis were reflected in liver composition, and metabolic and redox enzymes reported higher lipid use and lower protein oxidation. In white and red muscle reserves, rBGH increased glycogen while reducing lipid. The isotopic analysis of muscles showed a decrease in the recycling of proteins and a greater recycling of lipids and glycogen in the rBGH groups, which favoured a protein sparing effect. The protein synthesis capacity (RNA/protein) of white muscle increased, while cytochrome-c-oxidase (COX) protein expression decreased in rBGH group. Proteomic analysis of white muscle revealed only downregulation of 8 proteins, related to carbohydrate metabolic processes. The global results corroborated that GH acted by saving dietary proteins for muscle growth mainly by promoting the use of lipids as energy in the muscles of the gilthead sea bream. There was a fuel switch from carbohydrates to lipids with compensatory changes in antioxidant pathways that overall resulted in enhanced somatic growth.  相似文献

Follicle stimulating hormone (FSH) is produced by the pituitary gland in a coordinated hypothalamic–pituitary–gonadal (HPG) axis event, plays important roles in reproduction and germ cell development during different phases of reproductive development (fetal, neonatal, puberty, and adult life), and is consequently essential for fertility. FSH is a heterodimeric glycoprotein hormone of two dissociable subunits, α and β. The FSH β-subunit (FSHβ) function starts upon coupling to its specific receptor: follicle-stimulating hormone receptor (FSHR). FSHRs are localized mainly on the surface of target cells on the testis and ovary (granulosa and Sertoli cells) and have recently been found in testicular stem cells and extra-gonadal tissue. Several reproduction disorders are associated with absent or low FSH secretion, with mutation of the FSH β-subunit or the FSH receptor, and/or its signaling pathways. However, the influence of FSH on germ cells is still poorly understood; some studies have suggested that this hormone also plays a determinant role in the self-renewal of germinative cells and acts to increase undifferentiated spermatogonia proliferation. In addition, in vitro, together with other factors, it assists the process of differentiation of primordial germ cells (PGCLCs) into gametes (oocyte-like and SSCLCs). In this review, we describe relevant research on the influence of FSH on spermatogenesis and folliculogenesis, mainly in the germ cell of humans and other species. The possible roles of FSH in germ cell generation in vitro are also presented.  相似文献

Factors associated with adiposity in normal weight female adolescents with adequate and high percent body fat: elaborating a risk model. This study was carried out to evaluate the factors associated with adiposity in normal weight adolescents presenting appropriate or high percent body fat, who attended the public schools in Vi?osa county-Minas Gerais/Brazil. A total of 118 female adolescents at age range from 14 to 19 years and have already presented the menarche were evaluated. The adolescents were divided into 2 groups: G1 with high percent body fat and G2 with appropriate percent body fat. The following variables were evaluated: anthropometric, body composition, lifestyle and the family history of non-communicable chronic diseases. In subsample, the basal energy consumption was determined by indirect calorimetry. The G1-grouped adolescents showed higher values for most anthropometric and body composition variables (p < 0.001). No differences were observed (p > 0.05) for basal metabolism and metabolism of lean body mass, smoking habit, total energy consumption and protein and lipid as well between groups. When comparing the physical activity level, the G1 adolescents spent more time with 1-level sedentary activities whereas the G2 ones showed higher total caloric consumption with daily activities (p < 0.001). According to risk factors under analysis, the variables related to lifestyle, as distinguishing the use of sweeteners (OR = 13.47), provided higher contribution to excessive adiposity in the normal weight adolescents. The detailed analysis of the body composition as well as the risk factors associated with excessive body fat makes possible the early diagnosis and the development of more appropriate intervention means.  相似文献

Two series of Mo/Al2O3 catalysts were prepared by equilibrium adsorption and incipient wetness impregnation methods. The effect of preparation method on the surface coverages of the calcined catalysts was investigated by the combined use of CO2 chemisorption, low temperature CO adsorption and ion scattering spectroscopy (ISS). For a given Mo loading, the CO2 and CO adsorption results showed little difference between the two preparation methods. As previously noted, the CO2 chemisorption method overestimated the Mo surface coverage. In contrast to the adsorption methods, the ISS technique gave different Mo surface coverage values for a given Mo content of the two series of catalysts. This apparent discrepancy was attributed to different repartition of the Mo phase between the internal and external surfaces which can only be detected by ISS. This interpretation is supported by the observed agreement between the coverage values measured from ISS and low temperature CO adsorption for presumably uniform catalysts obtained by the equilibrium adsorption method.  相似文献

Helicobacter pylori promotes the secretion of cytokines that regulate inflammation and carcinogenesis. Immune cells secrete cytokines into the extracellular medium or packaged in exosomes. The objective of this study was to analyze the profile of soluble and exosomal cytokines that were secreted by human peripheral blood mononuclear cells (PBMCs) that were infected with H. pylori and to build a network of interaction between cytokines and cellular proteins. PBMCs were obtained by density gradient centrifugation and infected with H. pylori for 24 h. The infection was verified by immunofluorescence and Western blot for CagA. The exosomes were obtained from culture supernatant by ultracentrifugation and characterized by transmission electron microscopy, particle size analysis, and Western blot for CD9 and CD81. Cytokines were quantified using a multiplex immunoassay in the culture supernatant, intact exosomes, and lysed exosomes. H. pylori adheres to lymphocytes and translocates CagA. In PBMCs, H. pylori induces an increase in the soluble and exosomal IL-1β, IL-6, TNF-α, IL-10, IL-17A, IL-21, and IL-22. The protein–protein interaction (PPI) network shows that soluble and exosomal cytokines interact with proteins that participate in signaling pathways such as NF-κB, MAPK, PI3K-Akt, Jak-STAT, FoxO, and mTOR, that are related to carcinogenesis; moreover, TNF-α had the highest number of interactions. Cytokine-loaded exosomes represent another means of intercellular communication that is activated by H. pylori to stimulate inflammation, carcinogenesis, or cancer progression. Cytokine-loaded exosomes are likely to be associated with extragastrointestinal diseases of inflammatory origin.  相似文献
