Environmental Risk Assessment of chemical products and effluents within EC countries require the use of cost effective standardized toxicity tests that in most cases are restricted to acute responses to high doses. Thus, subtle ecological effects are underestimated. Here we propose a short-term one day Daphnia magna feeding inhibition test as a cost effective and ecological relevant sublethal bioassay. The sensitivity and reliability of the proposed bioassay was tested in the laboratory against standardized bacteria, algae growth, D. magna and fish acute toxicity test by using 16 chemical mixture x water type combinations that included four different water types fortified with four complex chemical mixtures. Water types included ASTM hard water and three selected effluents diluted 1/10 in water to mimic worse field situations that many overexploited arid river ecosystems suffer during summer months when effluents are discharged into them with little dilution. The results obtained denoted a greater sensitivity of the proposed feeding bioassay in 51 out of 65 tests performed with an average sensitivity 50 fold greater than that of the standardized tests. The greater differences were obtained for mixtures that included narcotic chemicals and the lowest differences for those containing pesticides. Furthermore, feeding responses to the studied contaminant mixtures behaved differently to increasing TOC content than those based on bioluminescent bacteria and algae. Increasing TOC coming from sewage treated effluents decrease toxicity to the latter bioassays but increased those of D. magna feeding bioassays. These results empathize the need to include additional bioassays to monitor more accurately and realistically the toxicity of effluents or surface waters dominated by effluent discharges, a quite common situation in America and Mediterranean arid regions. 相似文献

Activation-induced deaminase (AID) is required for somatic hypermutation in immunoglobulin genes, but also induces off-target mutations. Follicular lymphoma (FL) and chronic lymphocytic leukemia (CLL), the most frequent types of indolent B-cell tumors, are exposed to AID activity during lymphomagenesis. We designed a workflow integrating de novo mutational signatures extraction and fitting of COSMIC (Catalogue Of Somatic Mutations In Cancer) signatures, with tridimensional chromatin conformation data (Hi-C). We applied the workflow to exome sequencing data from lymphoma samples. In 33 FL and 30 CLL samples, 42% and 34% of the contextual mutations could be traced to a known AID motif. We demonstrate that both CLL and FL share mutational processes dominated by spontaneous deamination, failures in DNA repair, and AID activity. The processes had equiproportional distribution across active and nonactive chromatin compartments in CLL. In contrast, canonical AID activity and failures in DNA repair pathways in FL were significantly higher within the active chromatin compartment. Analysis of DNA repair genes revealed a higher prevalence of base excision repair gene mutations (p = 0.02) in FL than CLL. These data indicate that AID activity drives the genetic landscapes of FL and CLL. However, the final result of AID-induced mutagenesis differs between these lymphomas depending on chromatin compartmentalization and mutations in DNA repair pathways. 相似文献

The aim of this research was to investigate the effect of the addition of peel powder and/or its aqueous extract on physical and antioxidant characteristics of carboxymethyl cellulose (CMC) edible films. Prickly pear peel powder and aqueous extract (2 and 4%) were characterized in color, bioactive compounds (betalains and total phenolic compounds), antioxidant capacity and reducing power. Edible films were prepared with CMC, glycerol, varying the dissolution medium (0, 2, or 4% aqueous extract from prickly pear peel) and peel powder (0, 1, or 2%) using a face-centered central composite design. Physical, mechanical, bioactive compounds, and antioxidant capacity properties were investigated in edible films. Prickly pear peel powder presented 58.8 ± 0.08 mg of betacyanins/100 g, 53.8 ± 0.2 mg of betaxanthins/100 g, 967.8 ± 20 mg Gallic acid equivalent (GAE)/100 g for total phenolic compounds, an antioxidant capacity equivalent to 420.9 ± 4.9 mg GAE/100 g and a reducing power of 879.1 ± 115.9 mg ascorbic acid equivalent (AAE)/100 g. High concentration of aqueous extract and peel powder in CMC films increased the bioactive compounds content, antioxidant capacity and reducing power. Films mechanical properties were not affected by the aqueous extract; nevertheless, were strongly affected by the peel powder. Response surface methodology was used to optimize edible films formulation with high antioxidant characteristics, being the optimal formulation 1.7% of peel powder plus 3.3% of aqueous extract. This study may be the base for exploitation of prickly pear peel as source of bioactive compounds with antioxidant capacity for their use in edible coatings. 相似文献

The potential of the tetrabutylammonium-induced liquid-liquid-phase separation in alkyl carboxylic acid vesicular solutions for the extraction of organic compounds prior to liquid chromatography was examined for the first time. The behavior of the coacervates yielded from octanoic to oleic acids as a function of the pH and salts was investigated. The time required for phase separation depended on the length of the carboxylic acid alkyl chain and the experimental procedure (i.e., standing, sonication, centrifugation, stirring, etc.). Theoretical preconcentration factors were a function of both surfactant concentration and the length of the alkyl chain, and they greatly surpassed those obtained with other surfactant-mediated separations (e.g., surfactant-rich phases from dodecanesulfonic acid or Triton X-114). Parameters affecting the extraction efficiency were assessed. Analytes in a wide polarity/charge range, (e.g., PAHs, surfactants, chlorophenols, bisphenols, phthalates, herbicides, amines, dyes, and photographic developers) were extracted with high efficiencies on the basis of the different types of interactions that the vesicular coacervates can establish (i.e., hydrophobic and ionic interactions, hydrogen bonds, and formation of mixed aggregates). The coacervates were compatible with the chromatographic determination of analytes following UV or MS detection. Their suitability for working under real conditions was checked by applying them to the extraction of nonionic surfactants [alkylphenol ethoxylates (octyl and nonyl) and alcohol ethoxylates (C12-C16)] from raw and treated sewage and to river water samples. Analytes in the coacervate were separated and quantified by liquid chromatography-ion trap mass spectrometry. No cleanup steps were necessary. A functional interaction between progesterone, Th2 cytokines and a suitable balance between nitric oxide and prostaglandins in the uterus is considered to have a major role in the success of embryo implantation and pregnancy. Non-obese diabetic (NOD) mice offer a suitable model to study the modulatory role of Th1 cytokines on uterus signalling and function, since at the prediabetic stage they develop a spontaneous Th1 autoimmune response against exocrine glands similar to Sj?gren's syndrome. Vasoactive intestinal peptide (VIP) is a vasoactive neuro- and immunopeptide that promotes Th2 profiles and contributes to the smooth muscle relaxation and vasodilation. The aim of the present study was to investigate the activities of nitric oxide synthase and cyclo-oxygenase and the effect of VIP in the uterus of NOD mice with an emerging Th1 cytokine response. We present evidence of a reduced basal and VIP-stimulated activity of both enzymes in the uterus of NOD mice compared with normal BALB/c mice in proestrus. An altered functional interaction between both enzymes is also present in NOD mice at the time when increased levels of serum interleukin (IL)-12 and tumour necrosis factor-alpha but not interferon (IFN)-gamma or IL-10 were detected. We conclude that signalling alterations in uteri of NOD mice are simultaneous to the onset of a systemic Th1 cytokine response. 相似文献

Service continuity is one of the major aspects in the definition of the quality of the electrical energy, for this reason the research in the field of faults diagnostic for distribution systems is spreading ever more. Moreover the increasing interest around modern distribution systems automation for management purposes gives faults diagnostics more tools to detect outages precisely and in short times. In this paper, the applicability of an efficient fault location and characterization methodology within a centralized monitoring system is discussed. The methodology, appropriate for any kind of fault, is based on the use of the analytical model of the network lines and uses the fundamental components rms values taken from the transient measures of line currents and voltages at the MV/LV substations. The fault location and identification algorithm, proposed by the authors and suitably restated, has been implemented on a microprocessor-based device that can be installed at each MV/LV substation. The speed and precision of the algorithm have been tested against the errors deriving from the fundamental extraction within the prescribed fault clearing times and against the inherent precision of the electronic device used for computation. A number of countries with oligopolistic power industries have used marginal cost pricing to set the price of energy for small customers. This course of action, however, does not necessarily ensure an efficient outcome when competition is imperfect. The purpose of this paper is to study how the auction of long-term contracts could reduce market power. We do so in a two-firm, two-technology, linear-cost, static model where demand is summarized by a price inelastic load curve.
