The article describes an NMR spectroscopy study of interactions between vancomycin and a muramyl pentapeptide in two complexes: vancomycin and a native muramyl pentapeptide ended with D-alanine (MPP-D-Ala), and vancomycin and a modified muramyl pentapeptide ended with D-serine (MPP-D-Ser). The measurements were made in a 9:1 mixture of H2O and D2O. The obtained results confirmed the presence of hydrogen bonds previously described in the literature. At the same time, thanks to the pentapeptide model used, we were able to prove the presence of two more hydrogen bonds formed by the side chain amino group of L-lysine and oxygen atoms from the vancomycin carboxyl and amide groups. This type of interaction has not been described before. The existence of these hydrogen bonds was confirmed by the 1H NMR and molecular modeling. The formation of these bonds incurs additional through-space interactions, visible in the NOESY spectrum, between the protons of the L-lysine amino group and a vancomycin-facing hydrogen atom in the benzylic position. The presence of such interactions was also confirmed by molecular dynamics trajectory analysis.  相似文献

Betacyanins are a group of water-soluble red-violet compounds containing nitrogen in their structure. These are biosynthesized in red beetroot (Beta vulgaris L.), a widely consumed vegetable that contains significant amounts of nutritious and bioactive compounds which are also found in dietary supplements. This contribution presents results of betacyanin thermal oxidation (resulting in dehydrogenation) interrelated with decarboxylation in selected acetate/phosphate buffers at pH 3–8 and at 85 °C, which may be of particular significance for formulation and performance of foods. Most of the reaction products were detected at the highest concentrations in the acidic solutions (pH 3–4). The main dehydrogenation reaction pathways were monitored by LC-DAD-MS/MS and were associated with decarboxylation of the principal extract pigments, betanin/isobetanin and neobetanin, at carbon positions C-2 and C-17. Additional reactions are accompanied by the 2,15-decarboxylation processes at different dehydrogenation levels with 15-decarboxy-betanin and 2,15-bidecarboxy-betanin, structurally elucidated by NMR analysis, as the distinct indicators of this route type. For other novel pigments detected, 2,15-bidecarboxy-xanbetanin, 2,15-bidecarboxy-xanneobetanin and 2,15,17-tridecarboxy-neobetanin, additional high resolution mass spectrometric analyses were performed and confirmed their molecular formulas.  相似文献

The present study clarified changes in the contents of polar metabolites (amino acids, organic acids, saccharides, cyclitols, and phosphoric acid) in leaf senescence in Ginkgo biloba with or without the application of methyl jasmonate (JA-Me) in comparison with those in naturally senescent leaf blades and petioles. The contents of most amino acids and citric and malic acids were significantly higher in abaxially, and that of myo-inositol was lower in abaxially JA-Me-treated leaves than in adaxially JA-Me-treated and naturally senescent leaves. The levels of succinic and fumaric acids in leaves treated adaxially substantially high, but not in naturally senescent leaves. In contrast, sucrose, glucose, and fructose contents were much lower in leaf blades and petioles treated abaxially with JA-Me than those treated adaxially. The levels of these saccharides were also lower compared with those in naturally senescent leaves. Shikimic acid and quinic acid were present at high levels in leaf blades and petioles of G. biloba. In leaves naturally senescent, their levels were higher compared to green leaves. The shikimic acid content was also higher in the organs of naturally yellow leaves than in those treated with JA-Me. These results strongly suggest that JA-Me applied abaxially significantly enhanced processes of primary metabolism during senescence of G. biloba compared with those applied adaxially. The changes in polar metabolites in relation to natural senescence were also discussed.  相似文献

High-grade serous ovarian carcinoma (HGSOC) is the most frequent and malignant form of ovarian cancer. A local renin–angiotensin system (RAS) has been found in the ovary, and changes in selected components of this system were observed in pathological states and also in ovarian cancer. In the present study, we examined the effect of three peptides, Ang-(1-7), Ang-(1-9) and Ang-(3-7), on proliferation and motility of the OVPA8 cell line, a new well-defined and preclinical model of HGSOC. We confirmed the presence of mRNA for all angiotensin receptors in the tested cells. Furthermore, our findings indicate that all tested angiotensin peptides increased the metabolic serum in the medium by activation of cell defense mechanisms such as nuclear factor kappaB signaling pathway andapoptosis. Moreover, tested angiotensin peptides intensified serum starvation-induced cell cycle arrest at the G0/G1 phase. In the case of Ang-(3-7), a significant decrease in the number of Ki67 positive cells (Ki67+) and reduced percentage of activated ERK1/2 levels in ovarian cancer cells were additionally reported. The angiotensin-induced effect of the accumulation of cells in the G0/G1 phase was not observed in OVPA8 cells growing on the medium with 10% FBS. Moreover, in the case of Ang-(3-7), the tendency was quite the opposite. Ang-(1-7) but not Ang-(1-9) or Ang-(3-7) increased the mobility of reluctant-to-migrate OVAP8 cells cultured in the serum-free medium. In any cases, the changes in the expression of VIM and HIF1A gene, associated with epithelial–mesenchymal transition (EMT), were not observed. In conclusion, we speculate that the adaptation to starvation in nutrient-deprived tumors can be modulated by peptides from the renin–angiotensin system. The influence of angiotensin peptides on cancer cells is highly dependent on the availability of growth factors and nutrients.  相似文献

Protein glycosylation requires dolichyl phosphate as a carbohydrate carrier. Dolichols are α-saturated polyprenols, and their saturation in S. cerevisiae is catalyzed by polyprenyl reductase Dfg10 together with some other unknown enzymes. The aim of this study was to identify such enzymes in Candida. The Dfg10 polyprenyl reductase from S. cerevisiae comprises a C-terminal 3-oxo-5-alpha-steroid 4-dehydrogenase domain. Alignment analysis revealed such a domain in two ORFs (orf19.209 and orf19.3293) from C. albicans, which were similar, respectively, to Dfg10 polyprenyl reductase and Tsc13 enoyl-transferase from S. cerevisiae. Deletion of orf19.209 in Candida impaired saturation of polyprenols. The Tsc13 homologue turned out not to be capable of saturating polyprenols, but limiting its expression reduce the cellular level of dolichols and polyprenols. This reduction was not due to a decreased expression of genes encoding cis-prenyltransferases from the dolichol branch but to a lower expression of genes encoding enzymes of the early stages of the mevalonate pathway. Despite the resulting lower consumption of acetyl-CoA, the sole precursor of the mevalonate pathway, it was not redirected towards fatty acid synthesis or elongation. Lowering the expression of TSC13 decreased the expression of the ACC1 gene encoding acetyl-CoA carboxylase, the key regulatory enzyme of fatty acid synthesis and elongation.  相似文献

In the process of enzymatic hydrolysis of wheat starch to glucose, the presence of amylose‐lipid complexes (AML's) decreases swelling and dissolving capacity and the water binding capacity of starch, thus impeding the access of amylolytic enzymes to the starch granules. The aim of the work was to define the relationship between the stability of AML's and the conditions of the enzymatic hydrolysis of wheat starch such as the kind of enzymatic preparation (only amylolytic or both amylolytic and lipolytic) and time of hydrolysis. Hydrolysates were produced from wheat starch liquefied with bacterial α‐amylase BAN 240L, subjected to further treatment with the enzymatic preparation Spezyme GA 300W, containing glucoamylase and lysophospholipase and, for comparison, only with a glucoamylase preparation (AMG 300L). The effect of amylolytic and lipolytic enzymes on the stability of AML's in the process of wheat starch hydrolysis was estimated. The amount of fatty acids released during hydrolysis was determined with gas‐liquid chromatography (GLC) and by differential scanning calorimetry (DSC) measuring the enthalpy of decomposition of AML's. The investigations revealed a differentiated effect of the individual enzymatic preparations on the degree of degradation of AML's. Amylose‐lipid complexes were more susceptible to the attack of preparation Spezyme GA 300W than to the digestion by α‐amylase BAN 240 L and glucoamylase AMG 300L.  相似文献

In the current study, we screened a collection of coagulase-negative staphylococci (CoNS) isolates for orthologues of staphylococcal enterotoxins (SEs) involved in S. aureus-related staphylococcal food poisoning (SFP). The amplicons corresponding to SEs were detected in S. chromogenes, S. epidermidis, S. haemolyticus, S. borealis, S. pasteuri, S. saprophyticus, S. vitulinus, S. warneri, and S. xylosus. All amplicons were sequenced and identified as parts of known S. aureus or S. epidermidis SE genes. Quantitative real-time PCR allowed determining the relative copy number of each SE amplicon. A significant portion of the amplicons of the sea, seb, sec, and seh genes occurred at low copy numbers. Only the amplicons of the sec gene identified in three isolates of S. epidermidis displayed relative copy numbers comparable to sec in the reference enterotoxigenic S. aureus and S. epidermidis strains. Consecutive passages in microbiological media of selected CoNS isolates carrying low copy numbers of sea, seb, sec, and seh genes resulted in a decrease of gene copy number. S. epidermidis isolates harbored a high copy number of sec, which remained stable over the passages. We demonstrated that enterotoxin genes may occur at highly variable copy numbers in CoNS. However, we could identify enterotoxin genes only in whole-genome sequences of CoNS carrying them in a stable form at high copy numbers. Only those enterotoxins were expressed at the protein level. Our results indicate that PCR-based detection of enterotoxin genes in CoNS should always require an additional control, like analysis of their presence in the bacterial genome. We also demonstrate S. epidermidis as a CoNS species harboring SE genes in a stable form at a specific chromosome site and expressing them as a protein.  相似文献

The influence of extensive vegetated roofs on runoff water quality   总被引:5,自引:0,他引:5  
The influence of extensive sedum-moss vegetated roofs on runoff water quality was studied for four full scale installations located in southern Sweden. The aim of the study was to ascertain whether the vegetated roof behaves as a sink or a source of pollutants and whether the age of a vegetated roof influences runoff quality. The runoff quality from vegetated roofs was also compared with the runoff quality from non-vegetated roofs located in study areas. The following metals and nutrients were investigated: Cd, Cr, Cu, Fe, K, Mn, Pb, Zn, NO3-N, NH4-N, Tot-N, PO4-P, and Tot-P. The results show that, with the exception of nitrogen, vegetated roofs behave as source of contaminants. While in lower concentrations than normally found in urban runoff, some metals appear in concentrations that would correspond to moderately polluted natural water. Nitrate nitrogen is retained by the vegetation or soil or both. Apart from the oldest, the studied vegetated roofs contribute phosphate phosphorus to the runoff. The maintenance of the vegetation systems on the roofs has to be carefully designed in order to avoid storm-water contamination; for instance, the use of easily dissolvable fertilizers should be avoided.  相似文献

Methane drainage is used in Polish coal mines in order to reduce mine methane emission as well as to keep methane concentration in mine workings at safe levels. This article describes the method of methane drainage used in longwall 2 in seam 506. In Poland, coal seams are frequently mined in difficult conditions of very high methane hazard. Under such situations, methane is drained by means of parallel ventilation headings. This paper shows the influence of a specific ventilation system on the drainage efficiency at longwall 2 in seam 506. At this longwall, measurements of methane emission and the efficiency of drained methane were conducted. They consisted in gauging methane concentration, air velocity, absolute air pressure and the amount of methane removed via a drainage system. Experimental data were used to estimate the variations in absolute methane-bearing capacity, ventilation air methane and most importantly, to gauge the efficiency of methane drainage.  相似文献

Over the last decade it has been ever increasing interest in developing alternative sanitary systems, which would be more sustainable than a conventional one is. In Sweden urine separation has been considered a promising approach towards increasing recycling of wastewater nutrients. Full scale trials are carried out and examined. This paper presents a study on implementation of urine separation system in urban environment. Technical and non-technical aspects of system performance are evaluated; system's interference with existing infrastructure is examined. Benefits and losses connected to urine separation system are analysed not only from environmental perspective but also system's owner point of view.  相似文献
