On-line supercritical CO2 extraction - gas chromatography was applied to the isolation and identification of free fatty acids and other volatile compounds of young and ripe Swiss cheese (Emmental) produced in Finland. Extractions were carried out using a micro-cartridge at 40 °C temperature and 10 MPa pressure and the volatile fractions were analysed by DB-WAX column (polyethylene glycol phase) with flame ionization detection and mass spectrometry. The total time of analysis was less than 2.5 h. Acetic acid and propionic acid predominated over the C12–C18 acids, the longer chain fatty acids increasing in concentration during the ripening of the cheese. Due to the high proportions of fatty acids, further fractionation is required for analysis of the less abundant aroma compounds such as alcohols, carbonyls and lactones.  相似文献

BACKGROUND: alpha 2-Adrenergic agonists have been shown to reduce anesthetic requirements of other anesthetics, and they may even act as complete anesthetics by themselves at high doses in animal models. The present study was designed to define the interaction of intravenous infusion of dexmedetomidine, an alpha 2-adrenergic agonist, and isoflurane in patients having surgery by using the minimum alveolar concentration (MAC) of isoflurane as the measure of anesthetic potency. METHODS: Forty-nine women scheduled for abdominal hysterectomy were randomly allocated to receive either a placebo infusion (n = 16) or a two-stage infusion of dexmedetomidine with target plasma concentration of 0.3 ng/ml (n = 17) or 0.6 ng/ml (n = 16). The study drug infusion was commenced 15 min before induction of anesthesia with thiopental and alfentanil and was continued until skin incision. The end-tidal concentration of isoflurane for each patient was predetermined according to the "up-down" method of Dixon, and it was maintained for at least 15 min before the patient's response to skin incision was assessed. RESULTS: The MAC of isoflurane was 0.85% end-tidal in the control group, 0.55% end-tidal with the low dose of dexmedetomidine, and 0.45% end-tidal with the high dose of dexmedetomidine. CONCLUSIONS: The MAC of isoflurane in the control group was lower than that reported previously in similar patients having surgery, probably due to anesthesia induction with thiopental and alfentanil. Nevertheless, with the high dose of dexmedetomidine, the MAC of isoflurane was still 47% less than that without dexmedetomidine.  相似文献

Classic analyses of system implementations view user participation as a key element for successful implementation. However, under some conditions, avoiding user participation offers an alternative route to a successful implementation; this is advisable especially when the user network is weak and aligning user needs with the technological capabilities will take too much resource. To illustrate such situation, we analyse how a successful implementation outcome of an enterprise resource planning (ERP) system emerged in a recently established conglomeration of two previously independent universities. The ERP was used to replace several legacy student administration systems for both political and functional reasons. It was deemed successful by both project consultants and the new university's management while the users were marginalised (‘black boxed’) and left to ‘pick up the pieces’ of an incomplete system using traditional methods such as shadow systems and work‐a‐rounds. Using a process approach and an actor–network theory ‘reading’ of related socio‐technical events, we demonstrate how three networks of actors – management, the project team and the administrative users – collided and influenced the implementation outcome and how the management and project network established the ERP as a reliable ally while at the same time the users – while being enrolled in the network – were betrayed through marginalisation. Our analysis also suggests a useful way to conduct a ‘follow the network’ analysis explaining and accounting for the observed implementation outcome. We illustrate the benefits of using a socio‐technical processual analysis and show how stable actor networks must be constructed during large‐scale information technology change and how different actor groups perceive and influence differently the implementation outcome.  相似文献

Seven proton conducting membranes based on different commercial fluoropolymer films were prepared by radiation grafting with styrene followed by sulfonation. These membranes were studied as candidates for fuel cell electrolyte membranes and compared to Nafion® 105 and 117 with respect to conductivity, oxygen and hydrogen permeability, kinetics of the oxygen reduction reaction (ORR) and performance in a fuel cell. The dependence of the conductivity of the membranes on the relative humidity (RH) and temperature was also determined. The conductivity was observed to depend on the membrane thickness and the water uptake. The dependence of the conductivity on the temperature and the RH was the same for all of the experimental membranes. Reactant gas permeabilities appeared to depend only slightly on the matrix material and no major differences in the Tafel slopes and exchange current densities of the ORR were observed. Membranes with high water uptakes appeared to be less durable in the fuel cell than membranes with low water uptakes. Thus to prepare a membrane that is durable under the fuel cell conditions, the water uptake must remain low even at the expense of the conductivity.  相似文献

Triacylglycerol (TAG) molecular weight distribution and regioisomeric structure of selected molecular weight species in human milk and in 32 human milk substitutes was determined. Negative ion chemical ionization mass spectrometry was used to determine the molecular weight distribution and collisionally induced dissociation tandem mass spectrometry applied to identify the sn-2 and sn-1/3 positions of fatty acids in TAG. The main molecular weight species of human milk TAG in decreasing order of abundance were 52∶2, 52∶3, 52∶1, 54∶3, 50∶2, 50∶1, 54∶4, 48∶1, 54∶2, 48∶2, 46∶1, 52∶4, and 50∶3 (acyl carbon number/number of double bonds), constituting 83 mol% of total TAG molecular species. In human milk substitutes, the proportion of the corresponding molecular weight species varied from 33 to 87 mol%. The main TAG regioisomers within the molecular weight species 52∶2, 52∶3, and 50∶1 in human milk were 18∶1-16∶0-18∶1 (83 mol%), 18∶1-16∶0-18∶2 (83 mol%), and 18∶1-16∶0-16∶0 (80 mol%), respectively. In human milk substitutes, the corresponding proportions varied in a wide range of 0–82 mol%, 0–100 mol%, and 0–73 mol%, respectively. Although TAG structures in some human milk substitutes closely resembled those in human milk, the great variation among samples leads to the conclusion that it is still possible to improve the TAG composition in human milk substitutes by applying novel methods to synthesize structured TAG.  相似文献

Chemical methods for successfully coating opal particles with electrically conducting polyaniline (PANi) polymers were studied. Two different routes were proven successful. First was the in situ chemical doping reaction of PANi with dodecylbenzenesulfonic acid (DBSA) and camphorsulfonic acid dopants. The other method was placement of opal particles in a low concentration of a conducting PANi/DBSA solution in chloroform. Ultraviolet analyses of opal samples showed typical spectra for doped PANi with delocalized polarons. The electrical resistance of coated opal was measured to be 0.75 × 103 Ω. Morphological studies of the fractured surface by scanning electron microscopy indicated successful coating. Cyclic voltammetry studies confirmed the percolating behavior of infiltrated PANi. The discourse around media choice has generated a diverse array of media choice factors originating from both the media‐based and social interaction‐based approaches. The multitude of these factors hints at the adaptive nature of media choice. Alas, how a user engages with such factors and adaptively carries out media choice has remained understudied. We undertake a field study to explore the role of a plurality of choice factors and their interactions in shaping media choice processes and outcomes. In particular, we focus on how a user identifies relationships among plural choice factors while he or she works on his or her particular choice resulting in a similar outcome – email – given a large number of alternatives. Drawing upon a theory of affordances, we propose a systemic way of narrating the dynamics of media choice as a multi‐dimensional process where a user explores her or his surroundings – a niche – as to establish media affordances that will then help her or him achieve a communication goal. We identify five relational patterns of interactions among specific choice factors: reciprocity, emergence, complementarity, re‐exploration and actualisation. These patterns are shown to be emergent and highly interdependent. We conclude by reviewing future research avenues to formulate richer ‘ecological’ accounts of media choice.  相似文献

Plasma‐activated biaxially oriented polypropylene (BOPP) films and paper substrate have been coated with functional chitosan solutions. Plasma treatment increased the amount of surface peroxide groups and carboxyl groups on the BOPP films. As a result of plasma activation, the surface energy increased from 30 to 50 dynes/cm. The enzyme tyrosinase catalysed the grafting of octyl gallate and dodecyl gallate to amino groups of chitosan polysaccharide. Resulting coatings exhibited strong antimicrobial activity against Gram‐positive Staphylococcus aureus and Gram‐negative Listeria innocua. After 24 h of incubation, a total reduction in both bacteria cell numbers varied between >4.9 and 1.4 logarithmic units. Grafted dodecyl gallate and octyl gallate at pH 6 were found to have the lowest reduction values of <3 logarithmic units for S. aureus, while 1.4 logarithmic reduction value was obtained for grafted dodecyl gallate at pH 6 against L. innocua. Chitosan coatings were also effective barrier layers against oxygen transmission although the transmission rates clearly increased in high‐humidity conditions. In dry conditions, however, the transmission rate of 2 cm3/(m2 · 24 h) was obtained with chitosan‐coated BOPP. Coatings did not have any effects on water vapour transmission. Both gallates were successfully grafted at pH 6. As increased flocculation and colour formation indicated, the tyrosinase‐catalysed grafting was more powerful with octyl gallate. Dodecyl gallate containing chitosan coatings was more hydrophobic as compared to octyl gallate. Total migration of substances into 95% ethanol was ≥5 mg/dm2, thus materials may be exploitable in packaging purposes in direct contact with certain foodstuffs.
