This present study is a preliminary exploration of the affinity between a carboxylic model drug ibuprofen and aluminum hydroxide. Ibuprofen was comilled with aluminum hydroxide in different weight ratios in the solid state and was characterized by scanning electron microscopy (SEM), X-ray powder diffractometry (XRD), Fourier transform infrared spectroscopy (FTIR), and in vitro dissolution studies. XRD and SEM studies indicated complete interaction of ibuprofen with aluminum hydroxide and complete amorphization of aluminum hydroxide–ibuprofen complexed salt as well, on comilling with aluminum hydroxide at 1:2 ratio. FTIR data showed the disappearance of acid carbonyl peak with the appearance and the corresponding increase in absorbance of new signal at 1,682 cm?1 in the 1:1 and 1:2 ibuprofen–aluminum hydroxide-comilled powder. The accompanied increase in the absorbance of carboxylate peak in the ibuprofen–aluminum hydroxide physical mixture, and 1:0.1, 1:0.5, 1:1, and 1:2 (IBApm, and IB1A0.1, IB1A0.5, IB1A1, and IB1A2, respectively) comilled powder indicated an acid–base reaction between ibuprofen and aluminum hydroxide. On storage at 40°C and 75% relative humidity (RH) for 10 weeks, XRD study showed the absence of reversion to the crystalline state and FTIR data revealed continued increase of new signal at 1,682 cm?1 relative to carboxylic acid peak and no reappearance of carboxylic acid peak. In vitro dissolution studies revealed that the percent release of ibuprofen from the aluminum hydroxide-comilled powder is in the following order: IB1A2 < IB1A1 < ibuprofen crystal < ibuprofen milled alone < IB1A0.1 < IB1A0.5. Aluminum metal cation might have interacted to form a complex through the carboxyl and carbonyl groups of ibuprofen. Improved dissolution of drug associated with IB1A0.1 and IB1A0.5 is because of the absence of a new signal at 1,682 cm?1 and improved amorphization of the drug to some extent. Dissolution of drug affected in IB1A2 and IB1A1 may be because of the insoluble stable complex formation.  相似文献

Adherence of Helicobacter pylori to gastric epithelial cells is thought to be important in the pathogenesis of infection and may be essential to maintain lifelong colonization. However, the factors responsible for adherence to gastric epithelial cells in vivo have not been characterized, and the significance of adherence to standard epithelial cell lines is unclear. Hemagglutination is also thought to be important in H. pylori adherence. However, no studies have clearly linked H. pylori hemagglutination or adherence to cultured epithelial cells to primary gastric epithelial cell adherence. Furthermore, it is not clear whether laboratory strains which have undergone multiple passages lose potential colonization factors. In this study, we examined the effect of serial laboratory passage on hemagglutination and correlated the hemagglutination characteristics of H. pylori strains to primary gastric cell adherence. Variable expression of hemagglutination was seen with serial laboratory passage of 15 strains. After 100 serial laboratory passages, all strains had lost hemagglutination activity. Hemagglutination was seen in association with adherence to primary gastric cells in vitro isolated from 2 patients. An association with ultrastructural intimate adherence was seen with HEp-2 cells, but not with gastric adenocarcinoma cells. Ultrastructural adherence was seen in corresponding antral biopsies of patients whose strains were hemagglutination positive, but hemagglutination was not associated with gastric inflammation. These data indicate that H. pylori hemagglutination is lost with serial passage and that hemagglutination may play a role in the attachment of H. pylori to gastric epithelial cells, but the role of adherence to chronic gastric inflammation is unclear.  相似文献

Necrotizing enterocolitis (NEC) is a serious gastrointestinal disorder of preterm infants. Other than an association with prematurity and gastrointestinal feeding, no single factor or mechanism has been consistently linked to this disease. We have previously demonstrated that Escherichia coli isolates obtained from the stool of infants with NEC caused NEC-like injury in a weanling rabbit ileal loop model; this injury, in turn, could be blocked by coinfection with selected Gram(+) bacteria (Enterococcus faecium) isolated from asymptomatic controls. Using Caco-2 cells in a trans-well system, we now demonstrate that the same E. coli isolates can cross epithelial cell monolayers in the absence of ultrastructural change or damage. These results with E. coli contrast with those seen with Salmonella typhimurium, which passed through the monolayer at a higher rate and were associated with striking ultrastructural damage. Transcytosis of E. coli was reduced 3-5-fold in the presence of E. faecium previously shown to block NEC-like injury in the loop model. There was a mild increase in the rate of E. coli transcytosis when studies were conducted with younger, undifferentiated cells; these immature cells had no brush border, had decreased production of brush border-specific enzymes, but retained well defined tight junctions, as demonstrated by transepithelial electrical resistance and electron microscopy. A further reduction/ complete blockage of E. coli transcytosis was observed when E. faecium was used as the coinfectant in studies with these undifferentiated cells. We hypothesize that the ability of E. coli to cross epithelial cell layer is a critical initial step in the cascade of events which lead ultimately to NEC; blockage or reduction in E. coli transcytosis in the presence of certain Gram(+) organisms may play a significant role in prevention of NEC.  相似文献

This study focused on ecosystem responses to the environmental perturbations caused by the 2004 Indian Ocean Tsunami in a small lake that was a freshwater body in 1996, prior to the tsunami. The physicochemical and biological characteristics of Kokilamedu Lake (KKM) revealed drastic changes, compared with pretsunami conditions. Monthly average observations on water quality indicated the electrical conductivity of the water increased steeply to 17.41 mS cm?1 in 2009, from the lowest pretsunami value of 1.83 mS cm?1 (range of 1.83–5.25 mS cm?1). Simultaneously, the nitrate + nitrite (NO+ NO2) values increased significantly from 0.49 μmol L?1 in 1996 to 74.47 μmol L?1 in 2006. Silicate (SiO4‐Si) exhibited a dramatic increase in concentration, from an average of 64.87 μmol L?1 in the pretsunami period to 309.71 μmol L?1 the post‐tsunami period (2009–2010). Inorganic phosphate had increased to a maximum of 9.59 μmol L?1 from a pretsunami maximum of 1.09 μmol L?1. The chlorophyll‐α concentrations did not respond to the increased nutrient stoichiometry of the lake. There was a decreased chlorophyll‐α concentration under post‐tsunami conditions. The recent infilling of the lake with sediment during the tsunami, associated with wind‐driven resuspension reduced the light penetration. There was a significant improvement in dissolved oxygen concentrations (2006–2010 average of 8.27 mg L?1) in the lake, however, compared with the pretsunami values (1994–1995 average of 5.94 mg L?1). The algal component is now dominated by blue‐green algae, while green algae had dominated in the pretsunami period. Pre‐ and post‐tsunami observations from a control site did not exhibit such dramatic shifts from the 1995 and 1996 conditions, whereas a shift was apparent in the case of KKM. Certain marine fishes have adapted to this altered ecosystem. These marine species encountered (Elops machnata, Cociella punctata, Sphyraena jello, Platycephalus indicus, Glossogobius giuris) might have been recruited during the intrusion of the tsunami waves.  相似文献

Recent advancements in the area of Mobile Cloud Computing (MCC) have significantly contributed towards assisting mankind to handle varied types of emergency situations that may arise as a result of different natural calamities like earthquakes, floods, fire, etc, which may cause huge damage to public property and result in loss of wealth of the nation. In this work, we have proposed a mobile cloud assisted architecture that supports the multicloud and hybrid‐cloud environments, together with Cloud Probing Service (CPS) and Cloud Ranking Service (CRS). The proposed algorithm consumes data from the sensor nodes and offloads the data to the most suitable cloud. A three‐layered architecture has been proposed, and the anchor points facilitate in the creation of the interface between the different layers. The simulation results indicate that the proposed mobile cloud assisted architecture for handling emergency situations (MAAS) approach performs better than the baseline algorithms.  相似文献

To modify acrylamide (AAm) hydrogels for specialty applications, it was copolymerized with 2‐Hydroxypropyl methacrylate (HPMA) in different molar ratio at 25°C in 1:1 water–acetone solvent system, using ammonium persulphate (APS) and N,N,N,N‐tetramethyl ethylene diamine (TEMED) initiator–accelerator system. Two series of hydrogels were thus prepared using two different crosslinkers—ethylene glycol dimethacrylate (EGDMA) and N,N‐methylene bisacrylamide (N,N‐MBAAm). To affect property profile of the hydrogels, concentration of HPMA was varied over a range of five concentrations from 3.5 to 28 mM. Hydrogels were further functionalized by partial hydrolysis with NaOH and Hofmann amide degradation reaction. FTIR, Nitrogen analysis, and SEM were used to establish monomer reactivity and structure relationship of the hydrogels. Metal ion uptake was studied as a function of various structural aspects of the hydrogels. The synthesis of dense sintered sialon with external additives selected from the system Y2O3–AIN–SiO2 is reported. The highest density (3.21 g/cm3) was achieved at 1750°C at 90 min of sintering with 5 wt% additive. The degree of sialon substitution increased with the amount of liquid; the YSiO2N crystalline phase formed concurrently. Strength degradation occurred above 1000°C. The fracture toughness of the material sintered with a lower amount of sintering aid remained relatively unchanged to 1200°C. The material with more additive exhibited decreased toughness above 1000°C.  相似文献

The phase formation behavior and room temperature dielectric properties of bulk perovskite solid solution composition (Ba0.5Sr0.5)(Ti1 − x Zr x )O3 have been investigated. The samples with different Zr-content were prepared through solid state reaction. The XRD investigation showed that Zr+4 is systematically dissolved in Ba0.5Sr0.5TiO3 lattice up to about 60 atm.% substitution, having cubic Pm3m structure. Eighty atom percent Zr substituted composition showed to contain a cubic phase similar to that of x = 0.6 composition and a tetragonal (I4/mcm) phase. That is the solid solution breaks around at 80 atm.% Zr substitutions. Ba0.5Sr0.5ZrO3 was having orthorhombic Imma structure. Decrease in grain sizes were observed with increase in Zr content. The permittivity of the ceramics decreased with the increase in Zr substitution. The frequency dependency of dielectric loss in the frequency range 10 Hz to 10 MHz, were improved with Zr substitution in the ceramics. The room temperature ac and dc conductivity also decreased significantly with the increase in Zr-content.  相似文献

This paper presents the detailed turbulent kinetic energy budget and higher order statistics of flow behind a surface-mounted rib with and without superimposed acoustic excitation. Pattern recognition technique is used to determine the large-scale structure magnitude. It is observed that most of the turbulence contributions after the reattachment region are from the large-scale structures contrary to the belief that mostly random turbulent structures are present after reattachment. The dissipation is not a small-scale phenomena only. It may result due to the interaction between large-scale structures. From the results of higher order moments, the outer edge of the shear layer is observed to be non-Gaussian in nature with significant deviation from the Gaussian skewness and flatness value. The kinetic energy budget results show positive intermodal production in the outer edge of the shear layer indicating the presence of back scattering. The non-Gaussian velocity distribution, ejection motions and back-scattering present in the outer shear layer may be conjectured to be correlated with each other.  相似文献
