通过成分分析、硬度测试和金相观察等手段对齿轮精锻模具进行失效分析,该模具失效形式为热疲劳失效,表现为齿顶龟裂、开裂和磨损。模具表面发生回火软化,金相观察发现该精锻模存在严重带状组织和热处理组织不均匀现象,机械性能下降。可采用高温扩散处理、多向锻造和超细化正火处理,减少因带状组织产生的各向异性;通过严格控制热处理工艺,保证模具得到良好组织,提高模具使用寿命。 相似文献

目的:探讨沉默碳酸酐酶1(CA1)对人肺癌A549细胞增殖、凋亡、侵袭和迁移的影响。方法:采用脂质体转染法将CA1特异性siRNA(si-CA1组)及阴性对照(si-NC组)转染肺癌A549细胞,同时以转染空脂质体的A549细胞为空白对照组(Blank组)。采用实时荧光定量PCR(qPCR)和蛋白印迹法(Western blot)检测CA1 mRNA和蛋白表达水平,细胞计数试剂盒法(CCK-8)、流式细胞术、Transwell实验分别检测A549细胞增殖、凋亡、侵袭和迁移能力。结果:qPCR和Western blot检测结果显示,转入CA1 siRNA的A549细胞中CA1 mRNA和蛋白的表达水平均明显下调(P<0.05);CCK-8实验结果显示,沉默CA1后si-CA1组A549细胞在24、48和72 h时光密度(OD)值明显低于si-NC组(P<0.05);流式细胞术结果发现,si-CA1组A549细胞凋亡率明显高于si-NC组(P<0.05);Transwell实验结果显示,si-CA1组侵袭和迁移细胞数均明显少于si-NC组(P<0.05)。与Blank组相比,si-NC组以上各指标差异均不显著(P>0.05)。结论:沉默CA1能够抑制肺癌A549细胞增殖、侵袭和迁移,并促进细胞凋亡。 相似文献

This paper investigated the mechanism of precipitation and its influence upon prop-erties of ultra-thin hot strips of low carbon steel produced by CSP techniques using experiment and thermodynamics theory. The experimental results show that there are lots of fine and dispersive precipitates in microstructures. By analysis, most of aluminum nitrides are in grains, while coexisted precipitates of MnS are along grain boundaries. Coexisted precipitates compose cation-vacancy type oxides such as Al2O3 in the core, while MnS is at the fringe of surface. The precipitation behavior of AlN and MnS in the hot strip is studied by thermodynamic calculation. Epitaxial and polycrystalline Bi4Ti3O12 thin films were prepared on single crystal (100) MgO substrates by a chemical solution deposition process using metal naphthenates as starting materials. Pyrolyzed films (at 500°C) were annealed for 30 min in air at 650, 700, 750 and 800°C, respectively. The effects of annealing temperature on the crystallinity, epitaxy and surface morphology of the films were investigated by X-ray diffraction θ-2θ scans, pole-figure analysis, and atomic force microscopy (AFM). Epitaxially grown films annealed at 700 and 750°C, respectively, showed growth of three-dimensional needle-shaped grains. During annealing at 800°C, grain growth of Bi4Ti3O12 may be suppressed by the formation of a titanium-rich phase such as Bi2Ti2O7 owing to Bi volatilization, resulting in lower root mean square roughness than that of film annealed at 750°C. 相似文献

Systemic Acquired Resistance (SAR) improves immunity of plant systemic tissue after local exposure to a pathogen. Guard cells that form stomatal pores on leaf surfaces recognize bacterial pathogens via pattern recognition receptors, such as Flagellin Sensitive 2 (FLS2). However, how SAR affects stomatal immunity is not known. In this study, we aim to reveal molecular mechanisms underlying the guard cell response to SAR using multi-omics of proteins, metabolites and lipids. Arabidopsis plants previously exposed to pathogenic bacteria Pseudomonas syringae pv. tomato DC3000 (Pst) exhibit an altered stomatal response compared to control plants when they are later exposed to the bacteria. Reduced stomatal apertures of SAR primed plants lead to decreased number of bacteria in leaves. Multi-omics has revealed molecular components of SAR response specific to guard cells functions, including potential roles of reactive oxygen species (ROS) and fatty acid signaling. Our results show an increase in palmitic acid and its derivative in the primed guard cells. Palmitic acid may play a role as an activator of FLS2, which initiates stomatal immune response. Improved understanding of how SAR signals affect stomatal immunity can aid biotechnology and marker-based breeding of crops for enhanced disease resistance. 相似文献

Marine antifouling coating using functional polymers has emerged as an important tool to combat marine fouling. Owing to their natural abundance, polysaccharides represent a more sustainable option than synthetic polymers and carrageenan, a sulfated polysaccharide, is identified as a promising candidate for further research based on its excellent marine antifouling properties. However, existing research has only explored the application of carrageenan-based coatings for 2D objects, using techniques such as spin-coating. Here, a spray-coating method is proposed to apply carrageenan-based coatings to the surfaces of 2- and 3-D objects.
