Polymer‐clay nanocomposites are materials with many interesting structures, properties, and potential applications. Microstructural evaluation of a nanocomposite is not an easy task, as clay may form hierarchical structures which may look different when observed at various magnifications under a microscope, and also as the concepts of “intercalation” and “exfoliation” are not self‐sufficient to describe its morphology. In this work polymer‐clay nanocomposites of polystyrene and two styrene‐containing block copolymers (styrene‐butadiene‐styrene and styrene‐ethylene/butylene‐styrene) were prepared using three different techniques. Clay dispersion was evaluated by a recently developed microscopy image analysis procedure, combining the analysis of optical and transmission electron micrographs, and the characterization was complemented by X‐ray diffraction and rheological measurements. The results showed better clay dispersion for both block copolymers nanocomposites, mainly due to their molecular architectures. The yeast Candida deformans CBS 2071 produces an extracellular lipase which was shown to catalyse the production of various esters by the esterification of free fatty acids, even in the presence of a large molar excess of water. To clone the gene encoding this extracellular lipase, Saccharomyces cerevisiae was transformed with C. deformans genomic libraries and screened for lipolytic activity on a medium containing rapeseed oil emulsion and rhodamine B. Three members of a lipase gene family (CdLIP1, CdLIP2 and CdLIP3) were cloned and characterized. Each deduced lipase sequence has a Gly-His-Ser-Leu-Gly-(Gly/Ala)-Ala conserved motif, eight cysteine residues and encodes an N-terminal signal sequence. MALDI-TOF mass spectrometry analysis of a proteolytic digest of the lipase produced was used to obtain experimental evidence that the CdLIP1 gene encoded the extracellular lipase. Recombinant expression studies confirmed that the cloned genes encoded functional lipases. The three lipases are very similar to lipases from the related species Yarrowia lipolytica. Significant homologies were also found with several yeast and fungal lipases. As C. deformans CBS 2071 was previously considered to be synonymous with Y. lipolytica, the strains were compared for the extent of nucleotide divergence in the variable regions (D1/D2) at the 5'-end of the large-subunit (26S) ribosomal DNA (rDNA) gene. This rDNA region has diverged sufficiently to suggest that C. deformans is a separate species. The nucleotide sequences of the CdLIP1, CdLIP2 and CdLIP3 genes will appear in the EMBL nucleotide sequence database under Accession Nos AJ428393, AJ428394 and AJ428395, respectively.  相似文献

The number and mass distribution of airborne particles were recorded in several industrial plants. From the data obtained, particle deposition was estimated in three regions of the respiratory tract using the ICRP grand average deposition model based on Hinds' (1999) parameterization. The median diameter was 30–70 nm (number distributions), and >4 μm (mass distributions) near most work activities, resulting in linear relationships between the deposited number/mass concentrations and the number/mass concentrations in the air. Welding and laser cutting produced particles in the 200–500-nm range; total deposition was small, not in accordance with the linear relationship observed for the other work activities. The elemental content varied between particle sizes in some workplaces, causing different elements to deposit in different respiratory regions. Iron was the most abundant element in the particles in many of the workplaces; in an iron foundry, however, Fe was most abundant only in the micron-sized particles whereas the nanoparticles mainly comprised Pb and Sb.  相似文献

We consider the problem of testing hypotheses on the copula density from a two-dimensional random sample. We test the null hypothesis of a parametric class against a composite nonparametric alternative. Each density under the alternative is separated in the L 2-norm from any density lying in the null hypothesis. The copula densities under consideration are assumed to belong to a range of Besov balls. According to the minimax approach, the testing problem is solved in an adaptive framework: it leads to a log log  loss term in the minimax rate of testing in comparison with the non-adaptive case. A smoothness-free test statistic that achieves the minimax rate is proposed. The lower bound is also proved.  相似文献

Osteosarcoma-derived cell lines (SaOs-2, MG63) have recently been shown to deform their nucleus considerably in response to surface topography. Such a deformation had not been described previously. Here we present results on additional cell lines, including cancerous (OHS4, U2OS), immortalized (F/STRO-1+A and FHSO6) and healthy cells (HOP). The cancerous cells were found to deform extensively, the immortalized cells showed small deformations, whereas the healthy cells showed deformation only at short incubation times. These results suggest a strong link between the malignant transformation of cells and the state of the cytoskeletal network. We propose mechanisms to explain the deformation in which the cytoskeleton either pushes down on the nucleus during spreading or pulls it down upon adhesion to the pillars.  相似文献

The purpose of the present study is to examine the relation between quality of mother-child interaction in a lab and home setting, and quality of attachment of school-age children. A second objective of the study is to evaluate the associations between quality of mother-child interactions, attachment and maternal psychosocial measures (social support, depression, and parental stress). Security of attachment (Separation-Reunion procedure, Main & Cassidy, 1988) and the quality of mother-child interaction was evaluated for a sample of 38 children (mean age = 6 years). Mothers also completed self-report measures for depression, stress, and social support. Concurrent to the lab assessment, quality of mother-child interaction was also evaluated during a home visit. Results indicated a strong association between interactive patterns in both settings. Moreover, interactive patterns differed in terms of attachment classification with secure children showing the most harmonious patterns and disorganized/controlling children showing the most dysfunctional patterns. Maternal psychosocial measures were not related to child security of attachment, but mothers of insecure children reported marginally more stress related to the child. The development of a photo‐responsive micellar system capable of triggering cell death is reported. Precursors of the micelles are synthesized by connecting a lipophilic chain to a hydrophilic polyethylene glycol via a photo‐labile nitrobenzyl group. The resulting amphiphilic units are self‐assembled in water forming 12 nm micelles that are readily internalized into cells. Upon photo‐irradiation, micelles undergo cleavage and yield a cytotoxic nitrosobenzaldehyde derivative, which significantly inhibits the proliferation of MDA‐MB‐231 cells under standard in vitro conditions.  相似文献

Surface complexation of copper(II) on soil particles: EPR and XAFS studies   总被引:1,自引:0,他引:1  
The interactions of transition metals with natural systems play an important role in the mobility and the bioavailability of these metals in soils. In this study, the adsorption of copper(II) onto natural soil particles was studied as a function of pH and metal concentration. The retention capacity of soil particles was determined at pH 6.2 to be equal to 6.7 mg of copper/g of solid. The Langmuir and Freundlich isotherm equations were then used to describe the partitioning behavior of the system at different pH values. A combination of EPR, extended X-ray absorption fine structure (EXAFS), and X-ray absorption near-edge structure (XANES) spectroscopies was used to probe the Cu atomic environment at the soil particles/aqueous interface. The spectroscopic study revealed that copper(II) ions are held in inner-sphere surface complexes. It also revealed that Cu was in an octahedral coordination with first-shell oxygen atoms. A weak tetragonal distortion was pointed out due to the Jahn-Teller effect, with a mean Cu-Oequatorial bond distance of 1.96 A and a Cu-Oaxial bond distance of 2.06 A. A detailed analysis of the spectroscopic data suggested that Cu(II) was bonded to organic matter coated onto the mineral fraction of soil particles.  相似文献
