The search for new microbicide compounds is of an urgent need, especially against difficult-to-eradicate biofilm-forming bacteria. One attractive option is the application of cationic multivalent dendrimers as antibacterials and also as carriers of active molecules. These compounds require an adequate hydrophilic/hydrophobic structural balance to maximize the effect. Herein, we evaluated the antimicrobial activity of cationic carbosilane (CBS) dendrimers unmodified or modified with polyethylene glycol (PEG) units, against planktonic and biofilm-forming P. aeruginosa culture. Our study revealed that the presence of PEG destabilized the hydrophilic/hydrophobic balance but reduced the antibacterial activity measured by microbiological cultivation methods, laser interferometry and fluorescence microscopy. On the other hand, the activity can be improved by the combination of the CBS dendrimers with endolysin, a bacteriophage-encoded peptidoglycan hydrolase. This enzyme applied in the absence of the cationic CBS dendrimers is ineffective against Gram-negative bacteria because of the protective outer membrane shield. However, the endolysin—CBS dendrimer mixture enables the penetration through the membrane and then deterioration of the peptidoglycan layer, providing a synergic antimicrobial effect.  相似文献

Background: Dissemination of breast cancer (BC) cells through the hematogenous or lymphogenous vessels leads to metastatic disease in one-third of BC patients. Therefore, we investigated the new prognostic features for invasion and metastasis. Methods: We evaluated the expression of miRNAs and epithelial-to-mesenchymal transition (EMT) genes in relation to CDH1/E-cadherin changes in samples from 31 patients with invasive ductal BC including tumor centrum (TU-C), tumor invasive front (TU-IF), lymph node metastasis (LNM), and CD45-depleted blood (CD45-DB). Expression of miRNA and mRNA was quantified by RT-PCR arrays and associations with clinico-pathological characteristics were statistically evaluated by univariate and multivariate analysis. Results: We did not verify CDH1 regulating associations previously described in cell lines. However, we did detect extremely high ZEB1 expression in LNMs from patients with distant metastasis, but without regulation by miR-205-5p. Considering the ZEB1 functions, this overexpression indicates enhancement of metastatic potential of lymphogenously disseminated BC cells. In CD45-DB samples, downregulated miR-205-5p was found in those expressing epithelial and/or mesenchymal markers (CTC+) that could contribute to insusceptibility and survival of hematogenously disseminated BC cells mediated by increased expression of several targets including ZEB1. Conclusions: miR-205-5p and potentially ZEB1 gene are promising candidates for markers of metastatic potential in ductal BC.  相似文献

The standard site method was used to measure antenna factor. Based on calculation of antenna factor uncertainty, ways of more precise estimation of uncertainty, leading to uncertainty reduction, are presented. Using uncertainty standards together with known approaches to radiated emission measurement uncertainty calculation, a model of antenna factor measurement and an equation for calculation of its uncertainty were derived. Possible steps to reduce resultant measurement uncertainty are described, including an analysis of correlation, and acknowledgment of frequency dependence. The uncertainty was calculated in 25 frequency points in the range from 30 MHz to 1000 MHz. Furthermore, various calculations of uncertainty are executed to examine the suggested effects on the uncertainty value.  相似文献

The main goal of this paper is to present results of comparison study for the level set and direct Lagrangian methods for computing evolution of the Willmore flow of embedded planar curves. To perform such a study we construct new numerical approximation schemes for both Lagrangian as well as level set methods based on semi-implicit in time and finite/complementary volume in space discretizations. The Lagrangian scheme is stabilized in tangential direction by the asymptotically uniform grid point redistribution. Both methods are experimentally second order accurate. Moreover, we show precise coincidence of both approaches in case of various elastic curve evolutions provided that solving the linear systems in semi-implicit level set method is done in a precise way, redistancing is performed occasionally and the influence of boundary conditions on the level set function is eliminated. Electrophysiological experiments and modeling studies have shown that afterhyperpolarization regulates the discharge of lumbar motoneurons in anesthetized cats and is an important determinant of their firing properties. However, it is still unclear how firing properties depend on slow afterhyperpolarization, input conductance, and the fast currents responsible for spike generation. We study a single-compartment integrate-and-fire model with a slow potassium conductance that exponentially decays between spikes. We show that this model is analytically solvable, and we investigate how passive and active membrane properties control the discharge. We show that the model exhibits three distinct firing ranges (primary, secondary, and high frequency), and we explain the origin of these three ranges. Explicit expressions are established for the gain of the steady-state current-frequency (I-f) curve in the primary range and for the gain of the I-f curve for the first interspike interval. They show how the gain is controlled by the maximal conductance and the kinetic parameters of the afterhyperpolarization conductance. The gain also depends on the spike voltage threshold, and we compute how it is decreased by threshold accommodation (i.e., the increase of the threshold with the injected current). In contrast, the gain is not very sensitive to the input conductance. This implies that tonic synaptic activity shifts the current-frequency curve in its primary range, in agreement with experiments. Taking into account the absolute refractory period associated with spikes somewhat reduces the gain in the primary range. More importantly, it accounts for the approximately linear and steep secondary range observed in many motoneurons. In the nonphysiological high-frequency range, the behavior of the I-f curve is determined by the fast voltage-dependent currents, via the amplitude of the fast repolarization afterspike, the duration of the refractory period, and voltage threshold accommodation, if present.  相似文献

Examined the dynamics of interpersonal trust in group creative problem solving. Each member of 29 4-person groups of undergraduate females was led via manipulated feedback from 3 other group members to perceive a high trust, low trust, or control (no trust) manipulation condition. Two instructional sets were used: a standard brainstorming technique and synectics. Performance was measured by the number of ideas each group generated. Self-report data were taken on perceived effort, satisfaction, and group attractiveness. The high trust and control groups outperformed those in the low trust conditions on each of 3 tasks. Also, it appears that when information about trust is lacking in the group, members assume that relatively high trust exists. No differential impact due to problem-solving instructions was found. Ceramic tapes are used to build 3-dimensional components and microsystems in layer manufacturing. The tapes are individually printed and structured before being stacked and laminated. The structuring process of the tapes affects the maximal resolution of fluidic channels, suspended bridges and beams, which in turn determines the scale of miniaturization of the produced components. The aim of this paper is to investigate if the tape composition can be optimized to improve the cutting resolution of laser cutting, which is a very flexible tool for micromachining. Using the Siemens star pattern, the laser cutting resolution was measured for alumina green tapes of different binder compositions with different laser settings. For all tapes the resolution was better the higher the laser beam velocity. At higher velocity though, a higher number of cutting cycles is necessary to cut the tape. The laser cutting resolution depends on the binder composition, but the laser parameters must also be optimized to achieve high cutting resolution.  相似文献
