The electrochemical permeation technique for studying transport and trapping of hydrogen in Fe-40 at.% Al alloy at temperatures of 5, 25, 45 and 65 °C was used in the paper. The influence of temperature on the effective hydrogen diffusion coefficient, hydrogen permeation rate and hydrogen solubility was determined. Manipulator mounted on an unmanned satellite could be used for performing orbital capture maneuver in order to repair satellites or remove space debris from orbit. Use of manipulators for such purposes presents unique challenges, as high level of autonomy is required and the motion of the manipulator influences the position and orientation of the manipulator-equipped satellite. This paper presents a new control system that consists of two modules: trajectory planning module (based on trajectory optimization algorithm) and Model Predictive Controller. Both modules take into account the free-floating nature of the satellite-manipulator system. Proposed control system was tested in numerical simulations performed for a simplified planar case. In the first set of simulations Nonlinear Model Predictive Control (NMPC) was used to ensure realization of a square reference end-effector trajectory, while in the second set control system was used for optimizing and then ensuring realization of the trajectory that leads to grasping of the rotating target satellite. Simulations were performed with disturbances and with the assumed non-perfect knowledge of parameters of the satellite-manipulator system. Results obtained with NMPC are better than results obtained with the controller based on the Dynamic Jacobian inverse and with the Modified Simple Adaptive Control (MSAC). 相似文献

MAGE (melibiose-derived advanced glycation end-product) is the glycation product generated in the reaction of a model protein with melibiose. The in vivo analog accumulates in several tissues; however, its origin still needs explanation. In vitro MAGE is efficiently generated under dry conditions in contrast to the reaction carried in an aqueous solvent. Using liquid chromatography coupled with mass spectrometry, we analyzed the physicochemical properties and structures of myoglobin glycated with melibiose under different conditions. The targeted peptide analysis identified structurally different AGEs, including crosslinking and non-crosslinking modifications associated with lysine, arginine, and histidine residues. Glycation in a dry state was more efficient in the formation of structures containing an intact melibiose moiety (21.9%) compared to glycation under aqueous conditions (15.6%). The difference was reflected in characteristic fluorescence that results from protein structural changes and impact on a heme group of the model myoglobin protein. Finally, our results suggest that the formation of in vitro MAGE adduct is initiated by coupling melibiose to a model myoglobin protein. It is confirmed by the identification of intact melibiose moieties. The intermediate glycation product can further rearrange towards more advanced structures, including cross-links. This process can contribute to a pool of AGEs accumulating locally in vivo and affecting tissue biology. 相似文献

Staphylococcal biofilms are major causative factors of non-healing wound infections. Their treatment algorithms recommend the use of locally applied antiseptic agents to counteract the spread of infection. The efficacy of antiseptics against biofilm is assessed in vitro by a set of standard quantitative and semi-quantitative methods. The development of software for image processing additionally allowed for the obtainment of quantitative data from microscopic images of biofilm dyed with propidium iodine and SYTO-9 reagents, differentiating dead cells from live ones. In this work, the method of assessment of the impact of antiseptic agents on staphylococcal biofilm in vitro, based on biofilms’ processed images, was proposed and scrutinized with regard to clinically relevant antiseptics, polyhexanide, povidone–iodine and hypochlorite. The standard quantitative culturing method was applied to validate the obtained data from processed images. The results indicated significantly higher activity of polyhexanide and povidone–iodine than hypochlorite against staphylococcal biofilm. Taking into account the fact that in vitro results of the efficacy of antiseptic agents against staphylococcal biofilm are frequently applied to back up their use in hospitals and ambulatory units, our work should be considered an important tool; providing reliable, quantitative data in this regard. 相似文献

Genetically engineered T and NK cells expressing a chimeric antigen receptor (CAR) are promising cytotoxic cells for the treatment of hematological malignancies and solid tumors. Despite the successful therapies using CAR-T cells, they have some disadvantages, such as cytokine release syndrome (CRS), neurotoxicity, or graft-versus-host-disease (GVHD). CAR-NK cells have lack or minimal cytokine release syndrome and neurotoxicity, but also multiple mechanisms of cytotoxic activity. NK cells are suitable for developing an “off the shelf” therapeutic product that causes little or no graft versus host disease (GvHD), but they are more sensitive to apoptosis and have low levels of gene expression compared to CAR-T cells. To avoid these adverse effects, further developments need to be considered to enhance the effectiveness of adoptive cellular immunotherapy. A promising approach to enhance the effectiveness of adoptive cellular immunotherapy is overcoming terminal differentiation or senescence and exhaustion of T cells. In this case, EVs derived from immune cells in combination therapy with drugs may be considered in the treatment of cancer patients, especially effector T and NK cells-derived exosomes with the cytotoxic activity of their original cells. 相似文献

This article is devoted to a novel class of antimicrobial agents: nanocomposites composed of spherical silica and silver ions located at the silica’s surface with the assumed distribution. Such materials are in high demand due to the increasing threat from bacterial strains that are becoming resistant to currently known antibiotics. In particular, we focus on materials that make it possible to limit the growth of bacterial colonies on a variety of tactile surfaces. In this paper, we present a method for preparing a silica-based nanocomposite containing silver ions and the analysis of their antimicrobial properties. Our research revealed that the presence of tested nanocomposite induces very high oxidative stress in the bacteria cell, damaging and modifying bacterial DNA, creating oxidized guanines, cytosines, or adenines, which causes its very rapid destruction, leading to cell death. 相似文献

Coaching CEO successor candidates is challenging and deeply nuanced in the best of circumstances. The stakes rise exponentially when the sitting CEO owns the company, resents having "anointed" an eventual successor, and has been phenomenally successful despite the bruising effects of his narcissism and toxic micromanagement. This case study describes how a data-driven, insight-oriented coaching methodology helped the CEO candidate accelerate his effectiveness, instill hope in the organization, and forge a more stable relationship with the CEO--a sufficiently functional relationship for the future CEO to decide to remain with the company. Furthermore, this work highlights the importance of clinical skills and 3 coaching meta principles (traction, trust, and truth-telling). Finally, this longer term coaching engagement illustrates the dynamic role shift from coach to trusted advisor. Through this deepened relationship, coaching gains were consolidated and an objective sounding board was retained for both the CEO and his eventual successor.
