Interleukin 4 (IL-4) is a pleiotropic cytokine. Of the cell types responsive to IL-4, T cells express one IL-4 receptor (IL-4R) type, IL-4Ralpha/IL-2Rgamma (class I IL-4R), whereas endothelial cells express another type, IL-4Ralpha/IL-13Ralpha (class II IL-4R). It was hypothesized that IL-4 variants could be generated that would be selective for cell types expressing the different IL-4Rs. A series of IL-4 muteins were generated that were substituted in the region of IL-4 implicated in interactions with IL-2Rgamma. These muteins were evaluated in T cell and endothelial cell assays. One of these muteins, containing the mutation Arg-121 to Glu (IL-4/R121E), exhibited complete biological selectivity for T cells, B cells, and monocytes, but showed no activity on endothelial cells. Receptor binding studies indicated that IL-4/R121E retained physical interaction with IL-2Rgamma but not IL-13Ralpha; consistent with this observation, IL-4/R121E was an antagonist of IL-4-induced activity on endothelial cells. IL-4/R121E exhibits a spectrum of activities in vitro that suggest utility in the treatment of certain autoimmune diseases.  相似文献

In this paper, we address the challenge of realizing full-body behaviors in scalable modular robots. We present an experimental study of a biologically inspired approach to organize the morphology and control of modular robots. The approach introduces a nested hierarchy that decomposes the complexity of assembling and commanding a functional robot made of numerous simple modules. The purpose is to support versatility, scalability, and provide design abstraction. The robots we describe incorporate anatomy-inspired parts such as muscles, bones, and joints, and these parts in turn are assembled from modules. Each of those parts encapsulates one or more functions, e.g., a muscle can contract. Control of the robot can then be cast as a problem of controlling its anatomical parts rather than each discrete module. To validate this approach, we perform experiments with micron-scale spherical catom modules in simulation. The robots we simulate are increasingly complex and include snake, crawler, quadruped, cilia surface, arm-joint-muscle, and grasping robots. We conclude that this is a promising approach for future microscopic many-modules systems, but also that it is not applicable to relatively weak and slow homogeneous systems such as the centimeter-scale ATRON.  相似文献

Inertial microfluidics has been attracting considerable interest for size-based separation of particles and cells. The inertial forces can be manipulated by expanding the microchannel geometry, leading to formation of microvortices for selective isolation and trapping of particles or cells from a mixture. In this work, we aim to enhance our understanding of particle trapping in such microvortices by developing a model of selective particle entrapment. Design and operational parameters including flow conditions, size of the trapping region, and target particle concentration are explored to elucidate their influence on trapping behavior. Our results show that the size dependence of trapping is characterized by a threshold Reynolds number, which governs the selective entry of particles into microvortices from the main flow. We show that concentration enhancement on the order of 100,000× and isolation of targets at concentrations as low as 1/mL is possible. Ultimately, the insights gained from our systematic investigation suggest optimization solutions that enhance device performance (efficiency, size selectivity, and yield) and are applicable to selective isolation and trapping of large rare cells as well as other applications.  相似文献

To determine whether circadian profiles of various plasma hormones are abnormal in patients with winter seasonal affective disorder (SAD), we obtained 24-hour profiles of plasma cortisol, prolactin, and thyrotropin in subsets of a sample of 22 depressed patients with SAD on and off light therapy and in subsets of a sample of 24 normal controls. Cortisol levels did not differ between patients and controls, and levels in patients were not affected by light therapy. Prolactin levels were lower in patients than in controls throughout the day (p < 0.03) but were unaffected by light therapy. Independent of patient vs. control status, prolactin levels were higher in women than in men throughout the day (p < 0.003). Thyrotropin levels were no different in patients and controls, but levels in patients were lower following light therapy (p < 0.05).  相似文献

Lean is widely applied in hospitals, but the impact tends to be limited. This paper investigates three possible explanations: 1) maturity, 2) complexity, and 3) the value concept and analyses the bearing of these in a case study of lean application in a large Danish university hospital. The results indicate that lean tends to be applied in secondary and support functions with a logistic character and therefore has had a limited impact on the overall healthcare performance. The case study shows that there are constraints related to low lean maturity, the complexity of processes and operations as well as differences in value perceptions among the different professions (e.g., doctors, nurses, and managers) within the hospital. The conclusion is that lean is useful for hospitals, but the lean concept as well as its implementation methods need to be fitted to the organizational complexity and diverging values in hospitals in order to bring about a larger impact. It is therefore necessary to develop new models for the lean concept as well as the implementation of lean adapted to the particular context of hospitals with a stronger focus on patient experiences and on coordination of social relations.  相似文献

In point-and-click interfaces the location of targets is sometimes known to the user before visually identifying it, and sometimes not. This experiment investigates how pointing is affected by whether the target location is precued so that users know it in advance or nonprecued so that users learn it only at the onset of pointing trials. The study investigates this for young, adult, and elderly participants pointing with mouse and touchpad. Target precuing affects the trial completion time, the reaction time, the sheer movement time, and multiple movement kinematics. In addition, target precuing interacts with the use of either mouse or touchpad, with target distance, and with target size, but surprisingly little with participant age. Because the target location was always made known to participants no later than at the onset of the pointing trial, the effects of target precuing must be due to the different possibilities for mental and motor preparations.  相似文献

The oxidation of ethanol was studied in low-pressure, premixed flat flames using molecular beam mass spectrometry (MBMS) in combination with electron impact ionization (EI) and resonance-enhanced multiphoton ionization (REMPI). Flame temperature profiles were measured by laser-induced fluorescence (LIF) of seeded NO. Two ethanol/oxygen/argon flames with stoichiometries of ?=1.00 and ?=2.57 were investigated at 50 mbar by EI-MBMS. Profiles of a variety of stable and radical species were measured as a function of height above the burner. The benzene profile in the fuel-rich ethanol flame was obtained by REMPI-MBMS. The same technique was used to determine the dependence of the benzene concentration on the ethanol/propene ratio in low-pressure flames with blended fuels (propene/ethanol/oxygen/argon). The C/O ratio of all blends was kept constant at C/O=0.773 or C/O=0.600. Ethanol addition ranged from 0 to 15% for flames with C/O=0.773, and from 0 to 100% for flames with C/O=0.600. In both data sets, a decrease of the benzene concentration with increasing ethanol percentage was observed. Qualitative information on some other aromatic species with higher mass was also obtained.  相似文献

An aerosol-based process for coating the surface of arbitrary “carrier” particles with other types of (smaller) “coating” particles via mutual electrostatic attraction is described. Its practical viability was tested by depositing negatively charged 12-nm palladium particles on 250-nm silica spheres carrying a charge of approximately +40 units each. At respective concentrations of 3 to 8 × 106 particles per cm3 (with a charge fraction of about 25%) and 1 × 104 particles per cm3, the deposition process runs to completion (i.e., to neutralization of the carrier particles) within less than a minute. Comparative estimates show that electrostatically enhanced deposition rates are up to 50 times higher than purely thermal collisions. Transmission electron micrographs show a fairly uniform distribution of coating particles across the surface of the carrier particles. The electrostatic coating kinetics were determined experimentally via the charge loss of the carrier particles and compared also to numerical simulations using Zebel's model for electrostatic enhancement of the collision kernel. Measured rates were generally within 10–15% of the simulations, except for the very early stages of attachment (the first 10 s), where agreement was found to be rather sensitive to the coating particle concentration, possibly due to space charge effects.
