Pemphigus vulgaris is an autoimmune blistering disease that is induced by binding of antibodies to a 130/85-kD protein complex on epidermal keratinocytes. An in vivo experimental model of this disease was developed by reconstituting severe combined immunodeficiency (SCID) mice with 1-10 x 10(7) PBL from patients with naturally occurring pemphigus vulgaris. Of 49 reconstituted mice, 34 (69%) produced human IgG levels of > 0.1 mg/ml. Circulating anti-pemphigus antibodies were found in 20 of the 34 successfully reconstituted mice; 44% of these animals had deposits of human IgG in their own skin after it was traumatized by either heat or cold. Spontaneous pemphigus vulgaris-like blisters associated with human IgG deposits were rarely found in mouse skin. By contrast, allogeneic human skin grafted to 10 to 12 mice before reconstitution with patients' PBL developed pemphigus vulgaris-like lesions containing human IgG deposits. These results demonstrate that SCID mice can serve as a model of an antibody-mediated human autoimmune skin disease.  相似文献

Photosynthetic reaction center proteins purified from Rhodobacter sphaeroides purple bacterium were deposited on the surface of indium tin oxide (ITO), a transparent conductive oxide, and the photochemical/-physical properties of the composite were investigated. The kinetics of the light induced absorption change indicated that the RC was active in the composite and there was an interaction between the protein cofactors and the ITO. The electrochromic response of the bacteriopheophytine absorption at 771 nm showed an increased electric field perturbation around this chromophore on the surface of ITO compared to the one measured in solution. This absorption change is associated with the charge-compensating relaxation events inside the protein. Similar life time, but smaller magnitude of this absorption change was measured on the surface of borosilicate glass. The light induced change in the conductivity of the composite as a function of the concentration showed the typical sigmoid saturation characteristics unlike if the photochemically inactive chlorophyll was layered on the ITO. In this later case the light induced change in the conductivity was oppositely proportional to the chlorophyll concentration due to the thermal dissipation of the excitation energy. The sensitivity of the measurement is very high; few picomole RC can change the light induced resistance of the composite.  相似文献

BACKGROUND: Lactic acid has many applications in the chemical industries and it can be produced economically by microorganisms using biomass raw materials of different origins. Sweet sorghum juice is a high sugar content raw material with potential for lactic acid production because after hydrolysis of its sucrose content the remaining glucose and fructose can supply the carbon demand of most lactic acid bacteria. However, satisfying the nitrogen and B‐vitamin needs of the bacteria by supplementation with yeast extract and/or other alternative nitrogen‐containing supplements can make the process too expensive. RESULTS: Using a statistical optimization process much of the yeast extract can be replaced by a cheaper alternative nitrogen source, namely wheat gluten. This resulted in a fermentation with 99% lactic acid yield and 3.04 g L?1 h?1 volumetric productivity. CONCLUSION: Using response surface methodology (RSM) media optimization was performed for lactic acid fermentation with an industrially acceptable result, reducing the costs of raw materials by half, replacing yeast extract by an alternative nitrogen source and applying yeast extract only as a source of micro‐elements (vitamins, salts, etc.) Copyright © 2010 Society of Chemical Industry  相似文献

The permeability of carbon dioxide, oxygen, nitrogen, and air through commercial monolayer and multilayer films, based on polyethylene (PE), biaxially oriented polypropylene (BOPP), and polyamide (PA), used for food packaging is reported. The influence of temperature (from 10 to 60°C) on permeability and DSC characteristics changes was also analyzed. Literature data for gas permeability of the mentioned monofilms are quite variable due to differences in additives, thermal history, and crystallinity. In this work, the highest gas permeability is obtained for PE film at the higher temperature (50–60°C). Laminates exhibit different gas permeation behavior from that of monofilms. Generally, gas solubility coefficient increases at higher temperature (with an exception of PA/PE and BOPPcoex.met/PE), being higher for monofilms in comparison with laminates, while diffusion coefficients are lower for monofilms in comparison with laminates. The temperature dependence of the permeability, diffusivity, and solubility of gases shows two different regions in PE, BOPPcoex/PE (10–40°C and 40–60°C), PA/PE, and BOPPcoex.met (10–30°C and 40–60°C) films. In this article, models and methods for solving a real-life frequency assignment problem based on scheduling theory are investigated. A realistic frequency assignment problem involving cumulative interference constraints in which the aim is to maximize the number of assigned users is considered. If interferences are assumed to be binary, a multiple carrier frequency assignment problem can be treated as a disjunctive scheduling problem since a user requesting a number of contiguous frequencies can be considered as a non-preemptive task with a processing time, and two interfering users can be modelled through a disjunctive constraint on the corresponding tasks. A binary interference version of the problem is constructed and a disjunctive scheduling model is derived. Based on the binary representation, two models are proposed. The first one relies on an interference matrix and the second one considers maximal cliques. A third, cumulative, model that yields a new class of scheduling problems is also proposed. Computational experiments show that the case-study frequency assignment problem can be solved efficiently with disjunctive scheduling techniques.  相似文献

The study reports the results of testing the sensitivity of an early warning sampling plan for detecting milk batches with high aflatoxin AFM1 concentration. The effectiveness of the method was investigated by the analysis of 9017 milk samples collected in Italian milk processing plants that applied control plans with different action limits (AL). For those milk processing plants where 30 ng kg?1 AL has been applied, the AFM1 contamination was significantly lower at or above the 95th percentile of the milk samples when compared with plants that used 40 ng kg?1 AL. The results show that the control plan can be used effectively for early warning of occurrence of high AFM1 contamination of milk and to carry out pro-active measures to limit the level of contamination. Estimation of dietary exposure was also carried out, based on the aflatoxin M1 content of the milk samples and on Italian food consumption data. Estimated Daily Intakes (EDI) and Hazard Indices (HI) were calculated for different age groups of the population. HIs show that no adverse effects are expected for the adult population, but in the case of children under age three, the approximate HI values were considerably higher. This underlines the importance of the careful monitoring and control of aflatoxin M1 in milk and dairy products.  相似文献

PURPOSE: In order to improve the objective localization of bilateral cortical abnormalities in positron emission tomography (PET) image volumes, we developed a new three-dimensional image processing technique. The accuracy of this approach with respect to invasive subdural electroencephalography (EEG) data was assessed in a group of children with neocortical epilepsy. METHODS: Glucose PET image volumes were obtained from 12 epileptic children (mean age 5.2 +/- 4.3 years). Bilateral cortical areas of abnormal glucose metabolism were objectively determined using two conditional criteria assessed against a normal database. The normal database was derived from a group of 15 adult controls (mean age 27.6 years). The spatial relationship between seizure onset electrodes and PET abnormalities was assessed using a conventional receiver operating characteristic (ROC) analysis as well as using a newly defined spatial proximity index (SPI), which characterizes the association between adjacent, but not coincident, abnormalities. RESULTS: ROC analysis at the 2 standard deviation (SD) threshold, revealed an accuracy of 65% to detect seizure onset areas with a sensitivity of 64 +/- 17% and a specificity of 66 +/- 24%. Sensitivity decreased to 46 +/- 24% at the 3-SD threshold with a specificity of 80 +/- 21% (accuracy 75%). The average value for the SPI was determined as 3.82 +/- 1.65 which was 20% lower than the SPI value calculated using a simple in-plane two-dimensional asymmetry between homotopic cortical segments (4.52 +/- 3.82). CONCLUSION: The presented image processing technique improves localization of cortical abnormalities and provides valuable imaging clues for placement of subdural EEG grids prior to surgical resection.  相似文献
