Mechanical property changes, thermal stability, and water absorption capacity of poly(vinyl chloride) (PVC)/sisal fiber composites were assessed with respect to the effect of maleic anhydride chemical treatments of the sisal fiber, for five different sisal fiber contents, varying from 0 to 30% by weight in the composite. The composites prepared with the untreated sisal exhibited higher tensile modulus and hardness than the unloaded resin, while elongation and tensile strength were reduced. The deterioration in the mechanical properties of PVC blended with sisal fiber is attributed to the presence of moisture, interfacial defects at the fiber and polymer interface, and fiber dispersion in the PVC matrix. The amount of absorbed water is a function of the amount of fiber in the composite (F0 = 0 phr, F5 = 0.77 phr, and F20 = 4.83 phr). The comparison of the results of characterization of F5, F20, and F30 formulations prepared with the untreated fibers and the treated ones showed a reduction in absorbed water after the chemical treatment of fiber with maleic anhydride (F0 = 0 phr, F5 = 0.28 phr, and F20 = 2.99 phr), thus improving the mechanical properties of composites prepared with the treated sisal. © 2006 Wiley Periodicals, Inc. (1) Background: Biophysical techniques applied to serum samples characterization could promote the development of new diagnostic tools. Fluorescence spectroscopy has been previously applied to biological samples from cancer patients and differences from healthy individuals were observed. Dendronized hyperbranched polymers (DHP) based on bis(hydroxymethyl)propionic acid (bis-MPA) were developed in our group and their potential biomedical applications explored. (2) Methods: A total of 94 serum samples from diagnosed cancer patients and healthy individuals were studied (20 pancreatic ductal adenocarcinoma, 25 blood donor, 24 ovarian cancer, and 25 benign ovarian cyst samples). (3) Results: Fluorescence spectra of serum samples (fluorescence liquid biopsy, FLB) in the presence and the absence of DHP-bMPA were recorded and two parameters from the signal curves obtained. A secondary parameter, the fluorescence spectrum score (FSscore), was calculated, and the diagnostic model assessed. For pancreatic ductal adenocarcinoma (PDAC) and ovarian cancer, the classification performance was improved when including DHP-bMPA, achieving high values of statistical sensitivity and specificity (over 85% for both pathologies). (4) Conclusions: We have applied FLB as a quick, simple, and minimally invasive promising technique in cancer diagnosis. The classification performance of the diagnostic method was further improved by using DHP-bMPA, which interacted differentially with serum samples from healthy and diseased subjects. These preliminary results set the basis for a larger study and move FLB closer to its clinical application, providing useful information for the oncologist during patient diagnosis. 相似文献

Extracellular vesicles (EVs) are a heterogeneous population of secreted membrane vesicles, with distinct biogenesis routes, biophysical properties and different functions both in physiological conditions and in disease. The release of EVs is a widespread biological process, which is conserved across species. In recent years, numerous studies have demonstrated that several bioactive molecules are trafficked with(in) EVs, such as microRNAs, mRNAs, proteins and lipids. The understanding of their final impact on the biology of specific target cells remains matter of intense debate in the field. Also, EVs have attracted great interest as potential novel cell-free therapeutics. Here we describe the proposed physiological and pathological functions of EVs, with a particular focus on their molecular content. Also, we discuss the advances in the knowledge of the mechanisms regulating the secretion of EV-associated molecules and the specific pathways activated upon interaction with the target cell, highlighting the role of EVs in the context of the immune system and as mediators of the intercellular signalling in the brain. 相似文献

Neurotrophins are growth factors which bind to signal-transducing receptors called Trk proteins. The neurotrophins and their receptor proteins are present in the mammalian and avian lymphoid organs, thus suggesting that these factors could act upon cells of the immune system. Nevertheless, little is known about the cellular distribution of neurotrophins and their receptor proteins in avian lymphoid tissues. In this study we use immunohistochemistry to detect the cellular localisation of neurotrophins and their receptor proteins in the pigeon caecal tonsil, used as a model for avian secondary lymphoid organs. Rabbit polyclonal antibodies against neurotrophins (nerve growth factor -NGF-, brain-derived neurotrophic factor -BDNF- and neurotrophin -3 NT-3-) and against specific epitopes of TrkA, TrkB and TrkC proteins were used. Cytokeratins, vimentin, S-100 protein and chromogranin A were studied in parallel to identify cells which seemed to express neurotrophins and Trk proteins. TrkA-like protein was seen in the intestinal epithelium, whereas TrkB-like and TrkC-like proteins was found in cells which we identified as dendritic cells and macrophages. BDNF-like and NT-3-like reactivity was localised in intestinal epithelial cells, especially endocrine cells. Present results add further evidence to the presumptive immune role of neurotrophins and their receptors and the possible functions of these peptides in the caecal tonsil are discussed. 相似文献

Magnetic resonance imaging is frequently complicated by the presence of motion and susceptibility gradients. Also, some biologic tissues have short T2s. These problems are particularly troublesome in fast spin-echo (FSE) imaging, in which T2 decay and motion between echoes result in image blurring and ghost artifacts. The authors reduced TE in conventional spin-echo (SE) imaging to 5 msec and echo spacing (E-space) in FSE imaging to 6 msec. All magnetic gradients (except readout) were kept at a maximum, with data sampling as fast as 125 kHz and only ramp waveforms used. Truncated sinc radio-frequency pulses and asymmetric echo sampling were also used in SE imaging. Short TE (5.8 msec) SE images of the upper abdomen were compared with conventional SE images (TE = 11 msec). Also, FSE images with short E-space were compared with conventional FSE images in multiple body sites. Short TE significantly improved the liver-spleen contrast-to-total noise ratio (C/N) (7.9 vs 4.1, n = 9, P < .01) on T1-weighted SE images, reduced the intensity of ghost artifacts (by 34%, P < .02), and increased the number of available imaging planes by 30%. It also improved delineation of cranial nerves and reduced susceptibility artifacts. On short E-space FSE images, spine, lung, upper abdomen, and musculoskeletal tissues appeared crisper and measured spleen-liver C/N increased significantly (6.9 vs 4.0, n = 12, P < .01). The delineation of tissues with short T2 (eg, cartilage) and motion artifact suppression were also improved. Polymer composites can be benefited in many ways through the addition of carbon nanotubes (CNT). For instance, CNT can build up a percolated network within the polymer matrix, which results in a composite material with electrical conductivity and piezoresistive characteristics. This has very important implications for the realization of self-stress sensing structural composites. Moreover, the remarkable optical and transport properties of CNT permit to obtain information about the stress state of the composite at different scales. In the present work, the local and global stress response of SWCNT-epoxy composites is characterised by simultaneous Raman spectroscopic and electrical measurements on nanocomposite specimens submitted to different levels of surface strain. Both the Raman G′-band resonance frequency and the electrical resistance of the composite are found to change monotonically with strain until an inflection point is reached at ∼1.5% strain. Increased sensitivity of the piezoresistive network and lower load transfer efficiency occur beyond this strain level, and are considered to be the result of CNT slippage from the polymer. The reversibility of the stress sensitivity of the composites is verified by performing cyclic loading tests. Hysteresis loop are found to develop earlier on the Raman curves as in the resistance curves, which indicates that even at low strain levels, permanent damage is induced in the vicinity of carbon nanotubes. The use of Raman spectroscopy in combination with electrical methods provides a further insight on the stress sensing capabilities of CNT and the factors which affect the sensitivity and reproducibility of this behaviour. 相似文献

We report the results of density-functional calculations of the structures, binding energies and magnetic moments of the clusters Mo(N) (N = 2-13), Mo(12)Fe, Mo(12)Co and Mo(12)Ni that were performed using the SIESTA method within the generalized gradient approximation for exchange and correlation. For pure Mo(N) clusters, we obtain collinear magnetic structures in all cases, even when the self-consistent calculations were started from non-collinear inputs. Our results for these clusters show that both linear, planar and three-dimensional clusters have a strong tendency to form dimers. In general, even-numbered clusters are more stable than their neighbouring odd-numbered clusters because they can accommodate an integer number of tightly bound dimers. As a consequence, the binding energies of pure Mo(N) clusters, in their lowest-energy states, exhibit an odd-even effect in all dimensionalities. Odd-even effects are less noticeable in the magnetic moments than in the binding energies. When comparing our results for pure Mo clusters with those obtained recently by other authors, we observe similarities in some cases, but striking differences in others. In particular, the odd-even effect in three-dimensional Mo clusters was not observed before, and our results for some clusters (e.g.?for planar Mo(3) and Mo(7) and for three-dimensional Mo(7) and Mo(13)) differ from those reported by other authors. For Mo(12)Fe and Mo(12)Ni, we obtain that the icosahedral configuration with the impurity atom at the cluster surface is more stable than the configuration with the impurity at the central site, while the opposite occurs in the case of Mo(12)Co. In Mo(12)Co and Mo(12)Ni, the impurities exhibit a weak magnetic moment parallely coupled to the total magnetic moment of the Mo atoms, whereas in Mo(12)Fe the impurity shows a high moment with antiparallel coupling. 相似文献

Penile squamous cell carcinoma (PSCC) is a rare but aggressive neoplasm with dual pathogenesis (human papillomavirus (HPV)-associated and HPV-independent). The development of targeted treatment is hindered by poor knowledge of the molecular landscape of PSCC. We performed a thorough review of genetic alterations of PSCC focused on somatic mutations and/or copy number alterations. A total of seven articles have been identified which, overall, include 268 PSCC. However, the series are heterogeneous regarding methodologies employed for DNA sequencing and HPV detection together with HPV prevalence, and include, in general, a limited number of cases, which results in markedly different findings. Reported top-ranked mutations involve TP53, CDKN2A, FAT1, NOTCH-1 and PIK3CA. Numerical alterations involve gains in MYC and EGFR, as well as amplifications in HPV integration loci. A few genes including TP53, CDKN2A, PIK3CA and CCND1 harbor both somatic mutations and copy number alterations. Notch, RTK-RAS and Hippo pathways are frequently deregulated. Nevertheless, the relevance of the identified alterations, their role in signaling pathways or their association with HPV status remain elusive. Combined targeting of different pathways might represent a valid therapeutic approach in PSCC. This work calls for large-scale sequencing studies with robust HPV testing to improve the genomic understanding of PSCC. 相似文献
