An intracellular 3-hydroxybutyrate-oligomer hydrolase was purified from a poly(3-hydroxybutyrate)-degrading bacterium, Paucimonas lemoignei. It hydrolyzed the 3-hydroxybutyrate dimer with the highest specific activity of any of the enzymes reported so far. The gene was cloned and sequenced. The deduced amino acid sequence showed that the enzyme is a homolog of the PhaZc of Ralstonia eutropha H16.  相似文献

Previous in vitro studies have suggested that calreticulin (CALR), which is responsible for the folding and quality control of glycoproteins, may be associated with decidualization. However, its precise role in regulating decidualization has not been explored in vivo. Here, we used pregnant rat models to examine endometrial CALR expression during the peri-implantation period. We also examined whether polypectomy, a procedure that could ameliorate infertility, alters the endometrial expression levels of CALR and several implantation factors in women diagnosed as infertile. In rats, uterine CALR was expressed at a high level at the implantation site, and a marked increase in CALR expression was observed in decidual cells of normal pregnancy. In addition, endometrial CALR expression was enhanced by either administration of estradiol-17β in the delayed implantation rat model or the artificial induction of decidualization in the pseudopregnant rat. In cultured stromal cells, siRNA-mediated silencing of CALR inhibited the decidual stimulus-induced expression of prolactin, decidual/trophoblast prolactin-related protein, and connexin 43. In humans, the endometrial expression levels of the mRNAs encoding CALR and the implantation-related factor insulin-like growth factor binding protein (IGFBP)-7 tended to increase after polypectomy. The strongest positive correlation between expression levels before polypectomy was observed for IGFBP-7 and CALR, and the strength of this correlation increased after the surgery. Thus, endometrial CALR may play a role in the formation of decidua, and the polypectomy of infertile patients may result in the co-operative expression of endometrial factors, including CALR, that could enhance endometrial receptivity.  相似文献

An ultrafast X‐ray computed tomography (CT) scanner has been developed. This scanner overcomes problems that occur in a transient or unsettled state, which make the conventional CT scanner inappropriate. To reduce the scanning time, this X‐ray CT system uses electronic switching of electron beams for X‐ray generation instead of the mechanical motion adopted by conventional CT scanners. The mechanical motion is a major obstacle to improving scanning speed. A prototype system with a scanning time of 3.6 ms was initially developed and confirmed to measure the dynamic events of two‐phase flow. However, an increased scanning speed is generally required for practical use in the thermal hydraulics research field. Therefore, an advanced type which can operate under the scanning time of 0.5 ms and can measure two‐phase flow with a velocity up to 4 to 5 m/s was developed. © 2000 Scripta Technica, Heat Trans Asian Res, 29(3): 155–165, 2000  相似文献

This paper presents the results of an investigation on the loop current of a thyristor-controlled series capacitor (TCSC). The knowledge of loop current characteristics is important for understanding analytically the equivalent impedance of TCSC, estimating harmonics produced by TCSC, and determining ratings of the TCSC circuit components. However, little has been written on the loop current of TCSC circuit. Therefore, using a laboratory-scale TCSC installed on a laboratory power system, the TCSC currents and voltages were measured and analyzed to investigate the behavior of loop current. A mathematical model of TCSC was also developed to compare to the experimental results and further clarify the relationship between loop current and steady-state impedance characteristics, including effects of TCSC circuit component size on the characteristics. We have developed a robust vision system for 2D positioning of industrial parts stable to changes of lighting conditions. A wide dynamic range vision sensor that we had developed previously was used to avoid the saturation of object images. Additionally, a gray scale pattern matching technique was employed for robust image processing. Performance of the system with a wide dynamic range vision sensor was investigated experimentally in comparison with that of a system with a conventional video camera. The probability of correct positioning of an object under changing lighting conditions, simulating those in a factory, was 100% for the wide dynamic range vision sensor and 83% for the conventional video camera. The experimental results confirm the effectiveness of the developed system, revealing that dynamic range expansion of the video cameras is very effective for realizing robust robot vision systems. © 1997 Scripta Technica, Inc. Thermotropic liquid-crystalline compounds were applied as membrane materials (membrane solvent and neutral carrier) for neutral carrier-type ion sensors to investigate how the ordered arrangement of neutral carriers affects the property of the resulting ion sensors. Nematic, smectic, and cholesteric liquid-crystalline compounds were used as the membrane solvents and crown ether derivatives with a molecular structure similar to the liquid-crystalline solvent as the K+ neutral carriers. Polarized IR spectroscopy and X-ray diffraction experiments confirmed that the highly ordered arrangement of membrane components was retained in the liquid-crystal-based ion-sensing membranes containing a neutral carrier and a lipophilic salt. The ordered arrangement of neutral carriers in the liquid-crystalline membranes enhanced the ion selectivity significantly, probably due to the efficient cooperation of two adjacent crown ether moieties in the highly ordered and aggregated state.  相似文献

We newly generated an RNA-sequencing-based microRNA (miRNA) expression signature of head and neck squamous cell carcinoma (HNSCC). Analysis of the signature revealed that both strands of some miRNAs, including miR-139-5p (the guide strand) and miR-139-3p (the passenger strand) of miR-139, were downregulated in HNSCC tissues. Analysis of The Cancer Genome Atlas confirmed the low expression levels of miR-139 in HNSCC. Ectopic expression of these miRNAs attenuated the characteristics of cancer cell aggressiveness (e.g., cell proliferation, migration, and invasion). Our in silico analyses revealed a total of 28 putative targets regulated by pre-miR-139 (miR-139-5p and miR-139-3p) in HNSCC cells. Of these, the GNA12 (guanine nucleotide-binding protein subunit alpha-12) and OLR1 (oxidized low-density lipoprotein receptor 1) expression levels were identified as independent factors that predicted patient survival according to multivariate Cox regression analyses (p = 0.0018 and p = 0.0104, respectively). Direct regulation of GNA12 and OLR1 by miR-139-3p in HNSCC cells was confirmed through luciferase reporter assays. Moreover, overexpression of GNA12 and OLR1 was detected in clinical specimens of HNSCC through immunostaining. The involvement of miR-139-3p (the passenger strand) in the oncogenesis of HNSCC is a new concept in cancer biology. Our miRNA-based strategy will increase knowledge on the molecular pathogenesis of HNSCC.  相似文献

Catalyst performance of NiO–MgO solid solution catalysts for methane reforming with CO2 and H2O in the presence of oxygen using fluidized and fixed bed reactors under atmospheric and pressurized conditions was investigated. Especially, methane and CO2 conversion in the fluidized bed reactor in methane reforming with CO2 and O2 was higher than those in the fixed bed reactor over Ni0.15Mg0.85O catalyst under 1.0 MPa. In contrast, conversion levels in the fluidized and fixed bed reactor were almost the same over MgO-supported Ni and Pt catalysts. It is suggested that the promoting effect of catalyst fluidization on the activity is related to the catalyst reducibility. On a catalyst with suitable reducibility, the oxidized and deactivated catalyst can be reduced with the produced syngas and the reforming activity regenerates in the fluidized bed reactor during the catalyst fluidization. In addition, the catalyst fluidization inhibited the carbon deposition.  相似文献

A mechanically induced current (MIC) in a polypyrrole/Au-coated membrane (PPy/Au-membrane) composite with various surface morphologies was investigated, and the electrolyte conditions were determined in an electrochemical cell. A MIC was induced on porous PPy/Au-membranes with a thin layer of PPy. Conversely, relatively small MICs were observed in non-highly porous films such as freestanding films and PPy/Au-membranes with thick PPy deposits. A MIC smaller by one order of magnitude was also observed in a Au-membrane without PPy. These results indicated that the MICs was due to a charging phenomenon in both the redox and the double layer capacitances. The MIC also varied with supporting electrolyte and their concentration. The MIC was strongly reduced in solutions with diluted electrolytes and with bulky cationic electrolytes, indicating that the number and the penetration speed of mobile ions limited the magnitude of the MIC. These characteristics indicated that the MIC was essentially a diffusion limited current. A two-electrode MIC cell was also configured to investigate a power generation film in a normal saline solution, which can possibly be utilized for biomedical applications.  相似文献

Preeclampsia (PE) is characterized by disturbed extravillous trophoblast migration toward uterine spiral arteries leading to increased uteroplacental vascular resistance and by vascular dysfunction resulting in reduced systemic vasodilatory properties. Its pathogenesis is mediated by an altered bioavailability of nitric oxide (NO) and tissue damage caused by increased levels of reactive oxygen species (ROS). Furthermore, superoxide (O2) rapidly inactivates NO and forms peroxynitrite (ONOO). It is known that ONOO accumulates in the placental tissues and injures the placental function in PE. In addition, ROS could stimulate platelet adhesion and aggregation leading to intravascular coagulopathy. ROS-induced coagulopathy causes placental infarction and impairs the uteroplacental blood flow in PE. The disorders could lead to the reduction of oxygen and nutrients required for normal fetal development resulting in fetal growth restriction. On the other hand, several antioxidants scavenge ROS and protect tissues against oxidative damage. Placental antioxidants including catalase, superoxide dismutase (SOD), and glutathione peroxidase (GPx) protect the vasculature from ROS and maintain the vascular function. However, placental ischemia in PE decreases the antioxidant activity resulting in further elevated oxidative stress, which leads to the appearance of the pathological conditions of PE including hypertension and proteinuria. Oxidative stress is defined as an imbalance between ROS and antioxidant activity. This review provides new insights about roles of oxidative stress in the pathophysiology of PE.  相似文献
