The interaction of the alpha subunit with the beta2 subunit of tryptophan synthase is known to be necessary for the activation of each subunit and for the catalytic efficiency of the alpha2beta2 complex. To elucidate the roles of hydrogen bonds in the interaction site between the alpha and beta subunits for subunit association, eight mutant alpha subunits at five hydrogen bonding residues (N104D, N104A, N108D, N108A, E134A, E135A, N157D, and N157A) were constructed, and the thermodynamic parameters of association with the beta subunit were obtained using a titration calorimeter. The N104D and N104A mutations remarkably decreased the stimulation activities, the association constants, and the association enthalpies. Although the association constant and the stimulation activities of E134A were reduced in the absence of salt, the change in the association enthalpy was relatively small, and the addition of salt could repair its defects. The substitutions at positions 135 and 157 did not affect the stimulation activity and decreased the Gibbs energy of association corresponding to the defect in 1 mol of hydrogen bond. The present results suggest that the alpha subunit which has a mutation at position 104 cannot fold into an intact conformation upon complex formation, resulting in reduced stimulation activities. The hydrogen bond with Asn-104, which is a conserved residue among 16 microorganisms, was especially important for alpha/beta interaction and mutual activation.  相似文献

Using immunofluorescence microscopy, we have found that SeqA protein, a regulator of replication initiation, is localized as discrete fluorescent foci in E. coli wild-type cells. Surprisingly, SeqA foci were observed also in an oriC deletion mutant. Statistical analysis revealed that a SeqA focus is localized at midcell in newborn cells. The SeqA focus is duplicated and tethered at midcell until an FtsZ ring is formed. Subsequently, these foci migrate in opposite directions toward cell quarter sites and remain tethered there until the cell divides. The cell cycle-dependent bidirectional migration of SeqA-DNA complexes is quite different from the migration pattern of oriC Dna copies. MukB protein is required for correct localization of SeqA complexes by an unknown mechanism.  相似文献

GaN films have been deposited on GaAs(lOO) substrates by a novel growth technique, hot plasma chemical vapor deposition. A radio frequency N plasma source with high power, up to 5 kW, provides an abundance of nitrogen atoms during growth. In addition, strong ultraviolet emissions from the hot plasma irradiate onto the substrate and promote the dissociation of triethylgallium, this results in growth of GaN at very low temperature (even at room temperature). In this paper, we describe the characteristics of hot nitrogen plasma and present the results of the low temperature growth of GaN. In addition, we have investigated the effects of the nitridation of GaAs substrates. Reflection high energy electron diffraction indicates the formation of a surface cubic nitrided layer on the pretreated GaAs. The GaN films grown on fully nitrided GaAs(l00) substrates are of dominantly cubic structures.  相似文献

In this paper it is presented a new algorithm for the construction of panoramic images (including 360 panoramas), which has as main characteristic to avoid the distortion that occurs by joining of several successive images in a sequence. We used the SIFT and RANSAC algorithms to find overlap areas between pairs of images, as well as a Blend algorithm for smoothing the joints. As the proposed algorithm doesn’t cause distortions, the subsequent correction is not necessary, contributing to a better performance. The results of experiments using commercial software and also the proposed algorithm were compared through a visual analysis. In addition, a quantitative analysis was done using numeric measures calculated on a panoramic image, generated from an image sequence of a region mapped and georeferenced by Google Earth.  相似文献

The surface of type 304 stainless steel wire was sputter-etched to form conical protrusions, and a Ag–1 mass%Ti film was sputter-deposited on the protrusions. Bending tests of the wire showed that delamination occurred along the interface between film and substrate for the wire without protrusions at a small strain whereas only fine cracking and partial delamination (limited to the skin of film) occurred for the wire with protrusions at a much higher strain. It is suggested that the reason for the increase in delamination strength is the increase in the interfacial area and anchor effect introduced by the protrusions.  相似文献

The effects of the co-doping and the resultant co-segregation of 2 mol% TiO2 and 2 mol% GeO2 on the ionic conductivity and on the chemical bonding state in a tetragonal ZrO2 polycrystal were investigated. The conductivity data and grain boundary microstructure showed that the doped Ti4+ and Ge4+ cations segregate along the grain boundary, and this segregation causes a reduction in the conductivity of both the grain interior and grain boundary and an increase in the activation energy of the grain boundary conductivity. Overall, the data indicate that the segregation retards the diffusion of oxygen anions. A first-principle molecular orbital calculation explains the retarded diffusion of the oxygen anion from a change in the covalent bonds around the dopant cations; an increase in the strength of the covalent bond between the oxygen and doped cation should work to suppress the diffusion of the oxygen anion.  相似文献

To objectively evaluate the efficacy, safety and usefulness of the newly developed penem oral antibiotic, ritipenem acoxil (RIPM-AC), against bacterial pneumonia, we conducted a multi-center double-blind comparative study using cefotiam hexetil (CTM-HE) as the control drug. Both RIPM-AC and CTM-HE were orally administered at 200 mg t.i.d. for 14 days, in principle. The results were as follows: The total number of patients enrolled in this trial was 208, of which 152 cases (RIPM-AC group: 73, CTM-HE group: 79) were evaluable for clinical efficacy. 1. The clinical efficacy rates (excellent + good) were 91.8% (67/73) in the RIPM-AC group and 94.9% (75/79) in the CMT-HE group. There was no significant difference between the two groups, and the clinical equivalency of RIPM-AC to CTM-HE was demonstrated. 2. In the patients enrolled in the evaluation of clinical efficacy, the eradication rates of the causative organisms were 84.6% (22/26) in the RIPM-AC group and 91.7% (22/24) in the CTM-HE group, with no significant difference between the two groups. 3. Side effects were noted in 9 cases (9.6%) of the RIPM-AC group and 5 cases (4.9%) of the CTM-HE group. Abnormal laboratory test findings were observed in 23 cases (26.7%) of the RIPM-AC group and 15 cases (15.6%) of the CTM-HE group. There was no significant differences between the two groups in the incidence of side effects nor of abnormal laboratory test findings. In the safety evaluation, RIPM-AC was judged to be safe in 64 cases (68.1%) and CTM-HE in 82 cases (80.4%), with no significant difference. 4. The usefulness rates (markedly useful+useful) were 86.5% (64/74) in the RIPM-AC group and 92.5% (74/80) in the CTM-HE group. There was no significant difference between the two groups. Since RIPM-AC showed clinical efficacy similar to those of CTM-HE and posed no particular safety problems, it is expected to be a useful antibiotic for the treatment of bacterial pneumonia.  相似文献
