C6 glioma strongly express nitric oxide synthase. Rats bearing C6 tumours were pre-treated with i.v. Ng-nitro-L-arginine methyl ester (L-NAME), 3-morpholinosydnonimine (SIN-1) or saline before local cerebral blood flow (LCBF) or tumour capillary permeability (TCP) was measured by the [14C]iodoantipyrine autoradiographic or [14C]alpha-amino-isobutyric acid techniques. L-NAME and SIN-1 caused significant TBF alterations (-44% and +136%, respectively) with less marked (-15% and +33%) alterations in normal brain. Calculated cerebrovascular resistance changes within tumour were indeed selective. Baseline TCP was increased compared with normal brain (20-fold). L-NAME and SIN-1 administration did not alter TCP. These effects have significant implications for human malignant glioma management. Selective i.v. manipulation of LCBF, without significant changes in TCP, could increase the efficacy of chemotherapy, radiotherapy or provide better peritumoural oedema control.  相似文献

The reinforcing elements of multilayer elastomeric isolation bearings, which are normally steel plates, are replaced by a fiber reinforcement. The fiber-reinforced isolator is significantly lighter and could lead to a much less labor-intensive manufacturing process. In contrast to the steel reinforcement, which is assumed to be rigid, the fiber reinforcement is flexible in extension. This paper presents theoretical approaches for analyzing the compressive stiffness and bending stiffness of fiber-reinforced rectangular isolators. The influence of fiber flexibility on the stiffness of isolators is studied. In addition to theoretical stiffness solutions which are expressed in series form, simplified stiffness formulas based on the solutions of an infinitely long strip pad are also presented.  相似文献

Interest in the use of quantitative structure-activity relationships (QSARs) for regulatory purposes has been growing steadily over the years, and many models have been evaluated under the guidance and acceptability criteria defined at the Setubal workshop held in March 2002. This work explores some of the practical issues related to the use of QSARs for regulatory purposes using results obtained from rat oral lethality and fish acute toxicity estimates generated from computational models (including TOPKAT, MCASE, OASIS, and ECOSAR). Using data submitted under the Environmental Protection Agency's (EPA's) High Production Volume (HPV) Challenge Program, the results on the quality of the estimations are compared using a standard statistical review and an additional classification approach in which the hazard predictions were grouped using well-defined regulatory criteria (those used in EPA's New Chemical Program). Our results indicate that an evaluation of a model's regulatory applicability and predictive power is ultimately dependent on the specific criteria used in the assessment process. This work also discusses the practical difficulties associated with defining the domain of a predictive model using the estimates of four different ready biodegradation models and experimental data submitted under the EPA's New Chemical program. Our results suggest that the method a model employs for its predictions is as important as the training set in determining its domain of applicability. Together, these results highlight the challenges associated with developing reliable and easily applied acceptability criteria for the regulatory use of QSAR models.  相似文献

Recent advances in enantioselective separation techniques have enabled scientists to investigate environmental fate processes of chiral pollutants. In this study, congener- and enantiomer-specific toxaphene residues were monitored in captive, naturally contaminated fish (Fundulus heteroclitus) to characterize the effect of temperature and compound structure on the enantioselectivity of the elimination process. A previous study performed under warm water conditions (Tmean = 25 degrees C) demonstrated relatively rapid (t(1/2) approximately = 7-14 d) and enantioselective elimination of the reductive dechlorination metabolites 2-exo,3-endo,6-exo,8,9,-10-hexachlorobornane (B6-923 or Hx-Sed) and 2-endo,3-exo,5-endo,6-exo,8,9,10-heptachlorobornane (B7-1001 or Hp-Sed). As expected, repetition of this experiment at cooler water temperatures (Tmean = 15 degrees C) resulted in a decrease in overall (i.e., both enantiomers) first-order elimination rate constants. Enantiomer fractions or ratios (EFs/ERs) during elimination, however, varied by congener, ranging from racemic for very rapidly eliminated Cl5 homologues to increasingly nonracemic for selected Cl6-Cl8 homologues (including 86-923, several unknown Cl7 compounds, B8-1414, and B8-1945). As a result, we propose a classification to describe the environmental persistence of chiral toxaphene pollutants based on congener-specific elimination kinetics and susceptibility to biotransformation as measured by EFs/ERs.  相似文献

We identified some of the sources and sinks of monomethyl mercury (MMHg) and inorganic mercury (HgII) on Ellesmere Island in the Canadian High Arctic. Atmospheric Hg depletion events resulted in the deposition of Hg(II) into the upper layers of snowpacks, where concentrations of total Hg (all forms of Hg) reached over 20 ng/L. However, our data suggest that much of this deposited Hg(II) was rapidly photoreduced to Hg(0) which then evaded back to the atmosphere. As a result, we estimate that net wet and dry deposition of Hg(II) during winter was lower at our sites (0.4-5.9 mg/ha) than wet deposition in more southerly locations in Canada and the United States. We also found quite high concentrations of monomethyl Hg (MMHg) in snowpacks (up to 0.28 ng/L), and at times, most of the Hg in snowpacks was present as MMHg. On the Prince of Wales Icefield nearthe North Water Polynya, we observed a significant correlation between concentrations of Cl and MMHg in snow deposited in the spring, suggesting a marine source of MMHg. We hypothesize that dimethyl Hg fluxes from the ocean to the atmosphere through polynyas and open leads in ice, and is rapidly photolyzed to MMHgCl. We also found that concentrations of MMHg in initial snowmelt on John Evans Glacier (up to 0.24 ng/L) were higher than concentrations of MMHg in the snowpack (up to 0.11 ng/L), likely due to either sublimation of snow or preferential leaching of MMHg from snow during the initial melt phase. This springtime pulse of MMHg to the High Arctic, in conjunction with climate warming and the thinning and melting of sea ice, may be partially responsible for the increase in concentrations of Hg observed in certain Arctic marine mammals in recent decades. Concentrations of MMHg in warm and shallow freshwater ponds on Ellesmere Island were also quite high (up to 3.0 ng/L), leading us to conclude that there are very active regions of microbial Hg(II) methylation in freshwater systems during the short summer season in the High Arctic.  相似文献

For those interested in using head-coupled PC-based immersive virtual environment (VE) technology to train, entertain, or inform, it is essential to understand the effects this technology has on its users. This study investigated potential adverse effects, including the sickness associated with exposure and extreme responses (emesis, flashbacks). Participants were exposed to a VE for 15 to 60 min, with either complete or streamlined navigational control and simple or complex scenes, after which time measures of sickness were obtained. More than 80% of participants experienced nausea, oculomotor disturbances, and/or disorientation, with disorientation potentially lasting > 24 hr. Of the participants, 12.9% prematurely ended their exposure because of adverse effects; of these, 9.2% experienced an emetic response, whereas only 1.2% of all participants experienced emesis. The results indicate that designers may be able to reduce these rates by limiting exposure duration and reducing the degrees of freedom of the user's navigational control. Results from gender, body mass, and past experience comparisons indicated it may be possible to identify those who will experience adverse effects attributable to exposure and warn such individuals. Applications for this research include military, entertainment, and any other interactive systems for which designers seek to avoid adverse effects associated with exposure.  相似文献

Pedestrian accidents are a serious health risk to children in the UK and other Western countries. There remains a considerable amount to be learned about children's behaviour in real-traffic environments. The present study was intended to help fill this gap, by observing unobtrusively the behaviours of 123 adult-child pairs as they crossed the road at pedestrian light-controlled crossings. Eight behaviours were coded, including whether or not the pair stopped at the kerb, waited for the light to change, and checked to ensure traffic flow had stopped. Results showed that the adults observed provided reasonably good models of pedestrian behaviour, but that they rarely treated the crossing event as an opportunity to teach children explicitly about road safety. The only gender difference to emerge revealed that adults were more likely to hold girls' hands than boys' hands. No differences were observed in relation to (estimated) age of child. The implications of these findings for parental training programmes are explored.  相似文献

A dynamic coordinative-directed solubilization of single-walled carbon nanotubes (SWNTs) in aqueous solutions has been achieved through a combination of a Zn(II) metalloporphyrin complex and a cis-protected Pd(II) complex, which are believed to form charged acyclic and/or cyclic adducts on or around the side walls of SWNTs. The solubilization of SWNTs in aqueous solution only occurs when these acyclic and/or cyclic complexes are allowed to enter simultaneously into a self-assembly process with SWNTs under mild conditions. The aqueous solubility properties that these dynamic complexes confer upon SWNTs are believed to involve noncovalent bonding interactions between the two entities. They have been probed in solution using ultraviolet and visible absorption spectroscopy and in thin films using high-resolution transmission electron microscopy. The supramolecular electronic effects that the individual components of their acyclic and/or cyclic complexes impart upon a single semiconducting SWNT have been probed within a nanotube field-effect transistor device.  相似文献

Holography has been of increasing interest in recent years, with developments in many areas such as data storage and metrology. Photopolymer materials provide potentially good materials for holographic recording, as they are inexpensive and self-processing. Many experiments have been reported in the literature that describe the diffraction efficiency and angular selectivity of such materials. The majority of these reports discuss the performance of the holographic optical element after the recording stage. It has been observed, however, that sometimes, during exposure, the transmitted recording beam intensities vary with time. A simple phenomenological model is proposed to explain the beam modulation, which incorporates the growth of the phase grating, time-varying absorption effects, the mechanical motion of the plate, the growth of a lossy absorption grating during the recording process, and the effects of nonideal beam ratios.  相似文献
