The role of the competence protein ComEA in DNA uptake during transformation of competent Bacillus subtilis was analyzed by lysed-protoplast transformation (LP transformation). A comEA deletion mutant was constructed by a fusion polymerase chain reaction. Transformants of the mutant were obtained by LP transformation at a frequency of 1.1 × 10(2) transformants per μg DNA, representing a low relative efficiency of transformation [RET (mutant/wild type)] of 2.7 × 10(-6). This implied an important role of the protein during DNA uptake. When analyzing LP transformation of comEA with a plasmid (5.7 kb), a similar RET (mutant/wild type) of 5.6 × 10(-5) was obtained. Following addition of DNA into the comEA mutant culture, the number of transformants increased at a rate of 0.5 transformants/min, which was very low compared with the wild-type (6.9×10(4) transformants/min). However, even in the comEA mutant, DNA uptake began immediately after addition of DNA. Using co-transformation analysis of the comEA mutant, short linkages at distances of 2-156 kb could be detected, but not long linkages at distances of 671-1662 kb. Taken together, the results indicate that ComEA plays an important role in the transfer of transforming DNA into the DNA channel and in controlling the rate of DNA uptake.  相似文献

Recently, humanized animals whereby a part of the animal is biologically engineered using human genes or cells have been utilized to overcome interspecific differences. Herein, we analyzed the detail of the differentiation states of various human leukocyte subpopulations in humanized mouse and evaluated comprehensively the similarity of the leukocyte lineage between humanized mice and humans. Humanized mice were established by transplanting human CD34(+) cord blood cells into irradiated severely immunodeficient NOD/Shi-scid/IL2Rγ(null) (NOG) mice, and the phenotypes of human cells contained in bone marrow, thymus, spleen and peripheral blood from the mice were analyzed at monthly intervals until 4 months after cell transplantation. The analysis revealed that transplanted human hematopoietic stem cells via the caudal vein homed and engrafted themselves successfully at the mouse bone marrow. Subsequently, the differentiated leukocytes migrated to the various tissues. Almost all of the leukocytes within the thymus were human cells. Furthermore, analysis of the differentiation states of human leukocytes in various tissues and organs indicated that it is highly likely that the human-like leukocyte lineage can be developed in mice.  相似文献

Temperature characteristics of the open-circuit voltage (Voc) were investigated in the temperature range from 30°C to 240°C for the InGaP/InGaAs/Ge triple-junction cells. Also, single-junction cells that had the similar structure to the subcells in the triple-junction cells were studied. In the high-temperature range (from 170°C to 240°C), the temperature coefficients of Voc of the InGaP/InGaAs/Ge triple-junction solar cell (dVoc/dT) were different from those in the low-temperature range (from 30°C to 100°C). This is because photo-voltage from the Ge subcell becomes almost 0 V in the high-temperature range. It was found that the open-circuit voltage of a Ge single-junction cell reduced to almost 0 V temperatures over 120°C under 1 sun condition.  相似文献

Two-dimensional numerical computations were carried out in order to examine the effect of magnetizing force for the air in a cylindrical container with thermal and magnetic field gradients. In a gravitational field, the air was driven by both gravitational and magnetizing forces, and flow pattern and the heat transfer rate greatly depended on the axial position of the coil. Moreover, when the magnetizing and gravitational forces cancel out each other, convection became almost quiescent. On the other hand, the aerial flow was induced by the magnetizing force even in a nongravitational field, and the air behaved like an electrically conducting fluid.  相似文献

Transient radiative transfer characteristics in a three-dimensional scattering-absorbing medium subjected to collimated irradiation of a short pulse train were investigated. A basic problem in which a cube is exposed to collimated irradiation of a single unit step square pulse was solved via the transient discrete-ordinates method, and Duhamel's superposition theorem was used to construct the responses of various pulse trains. The effects of optical thickness, scattering albedo, pulse width, and pulse train interval between two successive pulses on the temporal profiles of divergence of radiative heat flux, reflectance, and transmittance were scrutinized.  相似文献

The effects of fuel droplet size on soot formation in spray flames formed in a laminar counterflow are investigated experimentally and numerically. Sauter mean diameter (SMD) of quasi-monodispersed fuel spray (n-decane) is carefully controlled independently from the other spray characteristics using a frequency-tunable vibratory orifice atomizer, and the two-dimensional spatial distributions of soot volume fraction and soot particle size are measured by laser induced incandescence (LII) and time resolved LII (TIRE-LII), respectively. In addition, the soot formation processes are examined in detail by a two-dimensional direct numerical simulation (DNS) employing a kinetically based soot model with flamelet model. The results show that the soot formation area and location are strongly affected by the SMD of the fuel spray. As the SMD of the fuel spray increases, the average soot formation area expands, whereas local suppression of soot formation is instantaneously observed in the spray flames because of the appearance of groups of unburned droplets. The size of soot particles tend to be larger in the outer part of the soot formation area compared to soot in the inner part. This is because the surface growth of soot particles markedly proceeds compared to the coagulation and oxidation.  相似文献

In directional solidification, a sample is pulled in an externally imposed temperature gradient. When the pulling direction and the temperature gradient are not perfectly aligned, a transverse temperature gradient is generated in the sample. We investigate the dynamics of lamellar eutectic patterns, both by experimental observations and by numerical simulations, and find that it is dramatically affected by a transverse gradient. Whereas, in its absence, the labyrinth patterns that are formed in the initial stage of solidification persist over the entire duration of the experiments, a transverse temperature gradient induces a drift of the pattern which rapidly generates a regular array of lamellae. The drift speed is found to be consistently larger than expected from the normal growth hypothesis, both in simulations and experiments.  相似文献

Polyaniline–silver nanocomposites were synthesized in the form of colloidal particles by the facile one-step aqueous chemical oxidative dispersion polymerization of aniline using silver nitrate as an oxidant and poly(vinyl alcohol) as a colloidal stabilizer. Aniline monomer was oxidized by silver ions, yielding polyaniline and elemental Ag simultaneously. The synthesized nanocomposite particles were colloidally stable over 2 years and transmission electron microscopy studies indicated the production of spherical, plate and rod-shaped polyaniline–silver nanocomposite particles with a silver core–polyaniline shell morphology. The conductivity of a pressed pellet of the nanocomposite particles using the conventional four-point probe technique was 1.4 × 10?2 S/cm at 25 °C. The nanocomposite particles behaved as a ‘colored’ particulate emulsifier for the stabilization of transparent oil-in-water emulsions.  相似文献

A new finishing process for metal molds by large-area electron beam (EB) irradiation is proposed in this study. In the large-area EB irradiation equipment used here, an EB with high-energy density is irradiated without focusing the beam, and so the EB with a maximum diameter of 60 mm can be used for melting or evaporating metal surface instantly. Experimental results show that the surface roughness decreases from 6 μmRz to less than 1 μmRz in just a few minutes under proper machining conditions. The corrosion resistance of metal mold surface also could be greatly improved by large-area EB irradiation. Furthermore, the surface roughness of tilting surface close to 90° could be well improved. Therefore, large-area EB irradiation method has a possibility to become a high-efficiency finishing process for metal molds.  相似文献
