In interphase eukaryotic cells, molecular transport between the cytoplasm and the nucleus is mediated by the nuclear pore complex (NPC), which perforates the double-membraned nuclear envelope (NE). Local permeability of the NE at large intact nuclei (approximately 400 microm in diameter) isolated from Xenopus laevis oocytes was studied by scanning electrochemical microscopy (SECM). Steady-state tip current versus tip-nucleus distance curves (approach curves) were measured with 10- and 2-microm-diameter Pt disk microelectrodes at the nuclei in isotonic buffer solutions containing redox-active molecules. The approach curves in the normalized form are independent of the tip diameter, indicating diffusion-limited membrane transport of the redox molecules. SECM chronoamperometry demonstrated that a decrease in the steady-state tip current at short tip-nucleus distances is due to smaller diffusion coefficients and concentrations of the redox molecules in the nucleus than those in the buffer solution. The experimental approach curves fit very well with theoretical ones for freely permeable membranes, yielding the NE permeability to the molecules that is at least 2 orders of magnitude larger than permeability of bilayer lipid membranes and cell membranes. This result indicates that passive transport of the redox molecules across the NE is facilitated by open NPC pores. The flux of the redox molecules sustainable by a single NPC channel (>9.8 x 10(6) molecules per NPC per second) and the diameter of the channel pore (>15 nm) were estimated from the SECM data by assuming the NE as an array of nanometer-sized NPC pores. The effects of the redox molecules on the nucleus and the NPC function were examined by studying signal-mediated nuclear import of rhodamine-labeled bovine serum albumin with and without nuclear localization signals by fluorescence microscopy. 相似文献

A simulation code was developed to evaluate the performance of the cesium adsorption instrument operating in Fukushima Daiichi Nuclear Power Station. Since contaminated water contains seawater whose salinity is not constant, a new model was introduced to the conventional zeolite column simulation code to deal with the variable salinity of the seawater. Another feature of the cesium adsorption instrument is that it consists of several columns arranged in both series and parallel. The spent columns are replaced in a unique manner using a merry-go-round system. The code is designed by taking those factors into account. Consequently, it enables the evaluation of the performance characteristics of the cesium adsorption instrument, such as the time history of the decontamination factor, the cesium adsorption amount in each column, and the axial distribution of the adsorbed cesium in the spent columns. The simulation is conducted for different operation patterns and its results are given to Tokyo Electric Power Company (TEPCO) to support the optimization of the operation schedule. We report a Fabry-Pérot resonator with spherical and flat mirrors to allow simultaneous electron-spin resonance (ESR) and nuclear magnetic resonance (NMR) measurements that could be used for double magnetic resonance (DoMR). In order to perform simultaneous ESR and NMR measurements, the flat mirror must reflect millimeter wavelength electromagnetic waves and the resonator must have a high Q value (Q?>?3000) for ESR frequencies, while the mirror must simultaneously let NMR frequencies pass through. This requirement can be achieved by exploiting the difference of skin depth for the two frequencies, since skin depth is inversely proportional to the square root of the frequency. In consideration of the skin depth, the optimum conditions for conducting ESR and NMR using a gold thin film are explored by examining the relation between the Q value and the film thickness. A flat mirror with a gold thin film was fabricated by sputtering gold on an epoxy plate. We also installed a Helmholtz radio frequency coil for NMR and tested the system both at room and low temperatures with an optimally thick gold film. As a result, signals were obtained at 0.18 K for ESR and at 1.3 K for NMR. We investigated the effects of Al concentration on the reciprocated motion of twin boundaries in pre-strained Mg-Al-Zn alloys through a combination of applied compression and tension,in-situ electron-backscattering diffraction observations,and high-angle annular dark-field scanning transmission electron microscopy observations.The twin growth was restricted by increased Al concentration,which resulted in the occurrence of smaller-sized twins.The reverse motion of twin boundaries was also restricted,resulting in the formation of higher fractions of secondary twins and 2-5° boundaries during reverse tension.The secondary twins and 2-5° boundaries mainly contributed to the increased ultimate tensile strength of the pre-strained Mg alloys.This effect is more significant in Mg alloys with larger pre-compression.Moreover,the increased amount of the Al solute atoms,rather than the precipitates,mainly contributed to the increased strengthening effect on the twin boundary motion.Our research contributes to development of high-strength Mg alloys by stabilizing twin boundaries. 相似文献
