A base non-specific and acid RNase was isolated from cellular slime mold (Dictyostelium discoideum) cells in a homogeneous state (about 2.4 kDa) by SDS-polyacrylamide gel electrophoresis. The RNase (RNase DdI) has a pH optimum of 5.0. The amino acid sequence of RNase DdI was determined by a combination of protein chemistry, a search of Data base, Dicty cDB and further sequence analysis of cDNA from the same bank. RNase DdI consists of 198 amino acid residues, and about 13.3, 0.9, 1.2, 3.3, and 1.0 residues of mannose, xylose, glucose, GlcNAc, and GalNAc, respectively. RNase DdI has two characteristic conserved segments of the RNase T2 family, and thus belongs to the RNase T2 family. Considering the fact that most of the RNase activity of D. discoideum is present in the lysosomal fraction [Wiener and Ashworth (1970) Biochem. J. 118, 505-512], it was concluded that the lysosomal RNase in D. discoideum is a member of the RNase T2 family. The amino acid sequence of RNase DdI is highly homologous with that of Physarum polycephalum RNase (RNase Phyb), and its amino acid sequence seems to be similar to those of plant/animal type RNases, rather than fungal RNases. The location of RNase DdI in the phylogenetic tree of the RNase T2 family was estimated.  相似文献

This paper describes the development of an anthropomorphic visual sensor which generates a spatially variant resolution image by using a retina-like structure. This sensor consists of a dove prism for image rotation and two linear CCD sensors with 512 pixel/line resolution and holds approximately 45 kbytes of image data. The retina-like sensor has variable resolution with increasing density towards the center of the visual field and yields a polar-coordinate image directly. The motion analysis of the object in the scene from the optical flow is considerably simplified if the velocity is represented in polar coordinates, compared to the case when the image is represented in cartesian coordinates. A calibration procedure for the proposed retina-like sensor is also presented with experimental data to verify the validity of the system. Development of this sensor holds promise in applications to high-speed tracking systems, such as the eyes of navigation robots, because it has data reduction and polar mapping characteristics.  相似文献

Intraspinal synovial cyst primarily occurs in the lumbar spine and may cause back pain and compression nerve disturbance. Percutaneous aspiration has been recommended as an alternative to surgery in three cases. We report on two patients who improved after aspiration of gelatinous material and steroid injection but whose cyst recurred within a short period of time. There are reportedly two types of synovial cyst: One contains serosanguineous fluid and the other gelatinous fluid. These two types are indistinguishable on computed tomography or magnetic resonance imaging. The results of all patients whose aspirated material was gelatinous were unsuccessful, including our cases. Thus, the outcome may depend on the pathology of the cyst.  相似文献

Bladder cancer has a high recurrence rate; therefore, frequent and effective monitoring is essential for disease management. Cystoscopy is considered the gold standard for the diagnosis and continuous monitoring of bladder cancer. However, cystoscopy is invasive and relatively expensive. Thus, there is a need for non-invasive, relatively inexpensive urinary biomarker-based diagnoses of bladder cancer. This study aimed to investigate the presence of activated protein kinase Cα (PKCα) in urine samples and the possibility of PKCα as a urinary biomarker for bladder cancer diagnosis. Activated PKCα was found to be present at higher levels in bladder cancer tissues than in normal bladder tissues. Furthermore, high levels of activated PKCα were observed in urine samples collected from orthotopic xenograft mice carrying human bladder cancer cells compared to urine samples from normal mice. These results suggest that activated PKCα can be used as a urinary biomarker to diagnose bladder cancer. To the best of our knowledge, this is the first report describing the presence of activated PKCα in the urine of orthotopic xenograft mice.  相似文献

It is possible to consider a mastered skill to be an optimal skill under some conditions, especially in the field of sports, We call an ideal skill generated in an artificial environment like a computer, an "artificially mastered skill". There must exist an objective function which represents the objective of the optimization for the skill. The objective function plays the most important role in defining the meaning of the mastered skill. In this paper, we propose a computer simulation method of generating artificially mastered motions for an upper limb during baseball pitching by using a three-dimensional mathematical model and an optimizing method. We report the consequences of simulation experiments for several objective functions that define the mastered skill in the meaning of "nonwasteful" and/or "smooth". From the consequences, we also investigate the property of an "artificially mastered skill motion" generated from each objective function, paying attention to differences among the consequent motions for each objective function from the viewpoints of kinematics and dynamics.  相似文献

Gangliosides (glycosphingolipids containing one or more sialic acids) are highly expressed in neural tissues in vertebrates, and four species (GM1a, GD1a, GD1b, GT1b) are predominant in mammalian brains. GM3 is the precursor of each of these four species and is the major ganglioside in many nonneural tissues. GM3 synthase (GM3S), encoded by ST3GAL5 gene in humans, is a sialyltransferase responsible for synthesis of GM3 from its precursor, lactosylceramide. ST3GAL5 mutations cause an autosomal recessive form of severe infantile-onset neurological disease characterized by progressive microcephaly, intellectual disability, dyskinetic movements, blindness, deafness, intractable seizures, and pigment changes. Some of these clinical features are consistently present in patients with ST3GAL5 mutations, whereas others have variable expression. GM3S knockout (KO) mice have deafness and enhanced insulin sensitivity, but otherwise do not display the above-described neurological defects reported in ST3GAL5 patients. The authors present an overview of physiological functions and pathological aspects of gangliosides based on findings from studies of GM3S KO mice and discuss differential phenotypes of GM3S KO mice versus human GM3S-deficiency patients.  相似文献

Lithium zirconate, Li2ZrO3, is known as a candidate blanket material in a fusion reactor. Various neutronics benchmark experiments for zirconium have thus been carried out so far. According to the independent benchmark studies by two parties, the neutron spectrum calculations show fairly large overestimation for most evaluated nuclear data libraries. However, the reason has not yet been made clear up to now. The author's group expects it would be due to a problem of evaluation for the natZr(n,2n) reaction cross-section, because the cross-section measurement is basically not possible with the foil activation method for zirconium isotopes except for 90Zr.In the present study, two neutrons emitted from natZr(n,2n) reaction have been measured directly to investigate the reason for the above overestimation. The measurement was done with our own special technique of detecting angle-correlated neutrons by the coincidence detection technique and the pencil-beam DT neutron source of FNS, JAEA. Angle-correlated energy differential cross-sections for natZr(n,2n) reaction were successfully measured. The obtained total cross-section above the emitted neutron energy of 800 keV was fairly larger than the one evaluated in JENDL-3.3. The total cross-section of natZr(n,2n) reaction was estimated by extrapolating the spectrum down to zero energy taking into account the nuclear temperature. The estimated cross-section value with the nuclear temperature of 1 MeV, which is larger than the one adopted in JENDL-3.3, was in acceptable agreement with JENDL-3.3. It is suggested from the result that the disagreement pointed out in the previous benchmark studies may be due to inappropriate nuclear temperature used in the evaluation. Further investigation of the nuclear temperature employed in the nuclear data evaluation should thus be carried out once again.  相似文献
