A great deal of effort has been focused on exploring the underlying molecular mechanism of osteoarthritis (OA) especially at the cellular level. We report a confocal Raman spectroscopic investigation on human osteoarthritic chondrocytes. The objective of this investigation is to identify molecular features and the stage of OA based on the spectral signatures corresponding to bio-molecular changes at the cellular level in chondrocytes. In this study, we isolated chondrocytes from human osteoarthritic cartilage and acquired Raman spectra from single cells. Major spectral differences between the cells obtained from different International Cartilage Repair Society (ICRS) grades of osteoarthritic cartilage were identified. During progression of OA, a decrease in protein content and an increase in cell death were observed from the vibrational spectra. Principal component analysis and subsequent cross-validation was able to associate osteoarthritic chondrocytes to ICRS Grade I, II and III with specificity 100.0%, 98.1%, and 90.7% respectively, while, sensitivity was 98.6%, 82.8%, and 97.5% respectively. The overall predictive efficiency was 92.2%. Our pilot study encourages further use of Raman spectroscopy as a noninvasive and label free technique for revealing molecular features associated with osteoarthritic chondrocytes. 相似文献

Growing evidence links household air pollution exposure from biomass cookstoves with elevated blood pressure. We assessed cross‐sectional associations of 24‐hour mean concentrations of personal and kitchen fine particulate matter (PM2.5), black carbon (BC), and stove type with blood pressure, adjusting for confounders, among 147 women using traditional or cleaner‐burning Justa stoves in Honduras. We investigated effect modification by age and body mass index. Traditional stove users had mean (standard deviation) personal and kitchen 24‐hour PM2.5 concentrations of 126 μg/m3 (77) and 360 μg/m3 (374), while Justa stove users’ exposures were 66 μg/m3 (38) and 137 μg/m3 (194), respectively. BC concentrations were similarly lower among Justa stove users. Adjusted mean systolic blood pressure was 2.5 mm Hg higher (95% CI, 0.7‐4.3) per unit increase in natural log‐transformed kitchen PM2.5 concentration; results were stronger among women of 40 years or older (5.2 mm Hg increase, 95% CI, 2.3‐8.1). Adjusted odds of borderline high and high blood pressure (categorized) were also elevated (odds ratio = 1.5, 95% CI, 1.0‐2.3). Some results included null values and are suggestive. Results suggest that reduced household air pollution, even when concentrations exceed air quality guidelines, may help lower cardiovascular disease risk, particularly among older subgroups. 相似文献

Models, which are developed to determine the effective properties of thermal spray coatings, require the material properties of each constituent of the coating as well as the information about the spatial positions and the geometries of these constituents as input parameters. The complex microstructure of thermally sprayed Yttria-stabilized zirconia (YSZ) coatings consists of irregular voids which are distributed non-uniformly in the coating. It is a common practice in the literature to employ two-dimensional (2D) cross-sectional images of the coatings to derive the geometrical model of the microstructure and conduct the simulations in 2D. In the context of this study, contrary to the 2D approach, a new three-dimensional (3D) reconstruction approach has been developed to model the microstructure of thermally sprayed coatings in 3D. The effective properties of an YSZ coating have been calculated by means of asymptotic homogenization and virtual testing methods. The results of the models, which have been conducted in 2D and 3D, are compared with each other. Finally, the capabilities of these methods with respect to the modeling approach (in 3D and in 2D) are analyzed on the basis of reference measurements. 相似文献

The aim of the present intervention study was to examine if increased influence on working hours among shift workers led to better sleep quality.391 employees were categorized into groups based on the performed activities: High (self-rostering), moderate (education and/or policy for working hours), and low intensity intervention (meetings and discussions) and reference. Sleep quality was assessed by Karolinska Sleep Questionnaire (KSQ) at baseline and follow-up (12 months). To elucidate the process of the intervention interviews were conducted.Influence on one’s own working hours increased only in the high intensity group (p < 0.001). No effects of interventions on sleep quality were observed. Thus, sleep quality was not improved by increasing work time influence in the present group of Danish elder care workers. This was partly due to program failure (failed intervention), but may also be due to other factors such as few participants working night and few working full time. 相似文献

Analysis of In‐Flight Particle Properties of Thermal Sprayed Ultrafine Powders Demands on functional coatings with high dimensional accuracy and high surface quality has led to increasing interest in processing of very fine powder grades in a particle size range < 20 μm in past years. Fine powders are not only showing a distinct potential for application of thin and dimensionally accurate coatings, but are also very promising for the production of dense and homogeneous coatings with improved mechanical properties. The large specific surface of fine powders is allowing for relatively low thermal energy levels that are introduced into the process. Nevertheless is also requires a very sensitive temperature control, to prevent overheating of the particles. The reduction of the thermal energy level is resulting in significant advantages particularly for the usability of the HVOF process for coating of inner diameters. Within this work in‐flight particle properties of ultrafine carbide powders were analyzed. The studied HVOF process allows the adjustment of a broad parameter range by utilization of a hydrogen stabilized liquid fuel combustion process. A conventional straight nozzle type as well as a curved nozzle for internal spraying was studied. Whey protein films plasticised with sucrose have excellent oxygen barrier properties and high gloss that are reduced with time as sucrose crystallises. Whey protein isolate (WPI) films plasticised with sucrose were stored in 53% relative humidity for up to 60 days. The oxygen permeability, tensile properties and gloss of the films were measured periodically following ASTM (American Society for Testing Materials) methodology. Changes in properties were compared with changes in WPI films plasticised with glycerol (no crystallisation) or plasticised with sucrose (crystallisation) plus a crystallisation inhibitor. The two inhibitors studied were lactose and raffinose. Crystallisation in WPI/sucrose films decreased tensile strength and elongation at break. However, the inhibitors hindered sucrose crystallisation, and the desired film properties were maintained for a longer period of time. Raffinose was the more effective inhibitor, maintaining the film flexibility and barrier properties for over 28 days and maintaining gloss at almost 90% of the initial value for 60 days of storage. 相似文献

Household air pollution (HAP) is estimated to be an important risk factor for cardiovascular disease, but little clinical evidence exists and collecting biomarkers of disease risk is difficult in low-resource settings. Among 54 Nicaraguan women with woodburning cookstoves, we evaluated cross-sectional associations between 48-hour measures of HAP (eg, fine particulate matter, PM2.5) and C-reactive protein (CRP) via dried blood spots; secondary analyses included seven additional biomarkers of systemic injury and inflammation. We conducted sub-studies to calculate the intraclass correlation coefficient (ICC) in biomarkers collected over four consecutive days in Nicaragua and to assess the validity of measuring biomarkers in dried blood by calculating the correlation with paired venous-drawn samples in Colorado. Measures of HAP were associated with CRP (eg, a 25% increase in indoor PM2.5 was associated with a 7.4% increase in CRP [95% confidence interval: 0.7, 14.5]). Most of the variability in CRP concentrations over the 4-day period was between-person (ICC: 0.88), and CRP concentrations were highly correlated between paired dried blood and venous-drawn serum (Spearman ρ = .96). Results for secondary biomarkers were primarily consistent with null associations, and the sub-study ICCs and correlations were lower. Assessing CRP via dried blood spots provides a feasible approach to elucidate the association between HAP and cardiovascular disease risk. 相似文献

AbstractThe network analysis method, Event Analysis of Systemic Teamwork (EAST), was used to examine routine aviation operations from multiple perspectives from six key areas (i.e. Dispatch, ATC, ATM, Maintenance, Loading, and the Cockpit). Data was collected over a five-day observational field trial at an international air cargo operator. Researchers recorded the activities of agents operating within the six key areas over three outbound and two inbound flights. Three networks (i.e. social, information and task) were created for four key phases of flight: (i) pre-flight checks and engines start (ii) taxi, take-off and assent, (iii) descent, landing and taxi, and (iv) park and shut down. The networks represent a ‘work audit’ of short-haul cargo operations, which enabled a detailed understanding of the interactions and connections within the current system. Implications for the future of distributed crewing concepts are discussed.Practitioner Summary: An analysis of the aviation system was undertaken using the amalgamated data from three outbound and two inbound flights. These analyses show the social, information and task interactions for cargo operations. This has been used to specify requirements for future distributed crewing options. 相似文献

Predicting what drivers will do as vehicle control is handed over to them from automation is a relatively new challenge for the motor vehicle industry. Operator Event Sequence Diagrams (OESDs) offer a way of modeling the interactions between the driver and vehicle automation in the handover of control. In this paper, two studies are presented in which a range of handover strategies are tested. The anticipated driver strategies were modeled using OESDs to serve as predictions of driver behavior. Drivers were then observed in two separate studies: (1) using a Lower-Fidelity (vehicle seat and controls) simulator and (2) using a Higher-Fidelity (whole vehicle) simulator. Driver behavior during a takeover task was categorized according to the signal detection paradigm into hits, misses, false alarms, and correct rejections. The results showed that for all strategies in both sets of studies, the median criterion for validity was exceeded ( > 0.8), suggesting that OESDs made good predictions of driver behavior during the handover of the vehicle from automation to manual control. 相似文献

Increased outdoor concentrations of fine particulate matter (PM2.5) and oxides of nitrogen (NO2, NOx) are associated with respiratory and cardiovascular morbidity in adults and children. However, people spend most of their time indoors and this is particularly true for individuals with chronic obstructive pulmonary disease (COPD). Both outdoor and indoor air pollution may accelerate lung function loss in individuals with COPD, but it is not feasible to measure indoor pollutant concentrations in all participants in large cohort studies. We aimed to understand indoor exposures in a cohort of adults (SPIROMICS Air, the SubPopulations and Intermediate Outcome Measures in COPD Study of Air pollution). We developed models for the entire cohort based on monitoring in a subset of homes, to predict mean 2-week–measured concentrations of PM2.5, NO2, NOx, and nicotine, using home and behavioral questionnaire responses available in the full cohort. Models incorporating socioeconomic, meteorological, behavioral, and residential information together explained about 60% of the variation in indoor concentration of each pollutant. Cross-validated R2 for best indoor prediction models ranged from 0.43 (NOx) to 0.51 (NO2). Models based on questionnaire responses and estimated outdoor concentrations successfully explained most variation in indoor PM2.5, NO2, NOx, and nicotine concentrations. 相似文献
