Head group behavior of nonionic amphiphilic compound, (poly(oxyethylene) hydrogenated castor oil, HCO), in aqueous dispersions were investigated by EPR (electron paramagnetic resonance) in conjunction with a modern slow-tumbling simulation. The aliphatic spin probes, 5-doxylstearic acid (5-DSA) and 3β-doxyl-5α-cholestane (CHL), were used to obtain fluidity of the surface region of the membrane. The order parameter (S 0) using the simulation for 5-DSA and CHL in the region were approximately 0.4 and 0.2, respectively. The ordering results suggest that the head group region of the membrane is somewhat fluid. The rotational diffusion coefficients (R ≈ 1/(6τR)) for the probes were 3.4 × 107 and 7.1 × 107 s−1, respectively. Activation energies, calculated using the temperature dependence of diffusion coefficients, were 18 and 17 kJ/mol for the probes. The EPR results imply that the CHL probe in the HCO membrane has quite different behavior in comparison with that of PC (phosphatidylcholine) from egg. Thus, the present EPR analyses have provided quantitative insight into the surface region of the amphiphilic membrane.  相似文献

A 12,808-nucleotide containing DNA fragment cloned from naphthalene-utilizing (Nah+) Pseudomonas aeruginosa PaK1 was analyzed and compared with the genes (pah(OUS)) of a 14,462-nucleotide DNA fragment from Pseudomonas putida OUS82. The DNA sequence analyses demonstrated that the naphthalene upper-pathway genes and their deduced enzymes were very similar between the two bacteria: nucleotide similarities, 83-93%; amino acid similarities, 79-95%. These genes were also similar to those of the nah operon of plasmid NAH7; in particular, the OUS82 genes were similar to the nah genes, whereas the PaK1 genes were almost identical to the dox genes of Pseudomonas sp. C18. A region homologous with the 84-bp repeated sequence that Eaton (J. Bacteriol., 176, 7757-7762, 1994) has found at a site upstream of he nah operon was found only in a region downstream of the pah(PaK) gene cluster in PaK1 and on both sides of the pah(OUS) gene cluster in OUS82. A PaK1 gene, corresponding to an unknown gene (nahQ) in the nah operon, is located between the 1,2-dihydroxynaphthalene dioxygenase gene and the trans-o-hydroxybenzylindenepyruvate (tHBP A) hydratase-aldolase gene (nahE), and was suggested to be involved in the conversion of naphthalene to salicylate. Just downstream of the pah(PaK) gene cluster, a portion of a region was identical to one-third of the transposase gene (tnpA) in a phenol-catabolic plasmid pEST1226.  相似文献

A gene encoding hydroxyquinol 1,2-dioxygenase was cloned from 2,4,6-trichlorophenol-degrading Ralstonia (Pseudomonas) pickettii strain DTP0602. Cell-free extracts of Escherichia coli containing a cloned 1.4-kb StuI-XhoI DNA fragment of R. pickettii DTP0602 hydroxyquinol 1,2-dioxygenase converted hydroxyquinol into maleylacetate and also degraded 6-chlorohydroxyquinol. The 1.4-kb DNA fragment contained one open reading frame (designated hadC) composed of 948 nucleotides. The molecular mass of 34,591 deduced from the gene product (HadC) was in agreement with the size (35 kDa) of the purified HadC protein determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The amino acid sequence of HadC exhibited high homology to that of the hydroxyquinol 1,2-dioxygenase of 2,4,5-trichlorophenoxyacetic acid-degrading Burkholderia cepacia AC1100 (Daubaras, D. L. et al., Appl. Environ. Microbiol., 61, 1279-1289, 1995). The active enzyme had a molecular mass of 68 kDa, suggesting that it is functional as a homodimer. The enzyme also catalyzed the oxidation of pyrogallol and 3-methylcatechol, possible intermediates in the degradation of 2,4,6-trichlorophenol, in addition to 6-chlorohydroxyquinol and hydroxyquinol. The dioxygenase catalyzed both ortho- and meta-cleavage of 3-methylcatechol.  相似文献

In recent years, many methods of model reference adaptive control system (MRACS) for a linear time‐varying (LTV) plant have been proposed. These methods assumed that the structure of plant parameters is known in advance. However, it is difficult to get a priori information of plant parameters. In this paper, an MRACS design for an LTV system based on high‐order estimator (HOE) is proposed. By applying dynamic certainty equivalence (DyCE) to LTV plants, a new MRAC law of LTV system is derived without knowing the structure of the plant parameters. The MRACS law is generated by using high‐order derivatives of an estimated parameter, so that robust HOE with a normalization signal and σ modification for the system introduced. Our proposed method can attain better performance than conventional methods, such as estimation with variable forgetting factor (VF) and the gradient projection method (GPM). The robust HOE establishes the boundedness of all of the estimated parameters under the condition that the estimated parameter and the first derivative of the parameter are bounded. It is shown that all signals in the adaptive loop are bounded and the output error converges to a closed set. The proposed method is compared to the familiar schemes, the gradient projection method and the estimation based on forgetting factor through numerical simulations, and the effectiveness of our proposed method is shown. © 2000 Scripta Technica, Electr Eng Jpn, 130(4): 87–98, 2000  相似文献

In-situ observations on α/γ phase transformation were made to study the effects of grain boundary microstructures on the formation of a new phase and the migration of α/γ interphase boundary in an iron-4. 2%Cr alloy. It was found that triple junctions with more random boundaries could be the primary nucleation sites for a new phase, while triple junctions with low angle or low ∑ coincidence boundaries did not play any role as preferential sites. The migration of α/γ interphase boundary during heating over the transformation temperature range showed the two stage behaviour characterized by a stage with a migration velocity of 0. 33~0. 75 mm/s and secondly by a stage with 3. 7~7. 6 mm/s. It was also found that abnormal grain growth and a high density of ∑3 coincidence boundaries could occur in a phase with bcc structure after cycling of α/γ phase transformation. A new mechanism of nucleation and growth of a new phase in α/γ phase transformation is proposed on the basis of roles of plane-matching interphase boundaries, as previously discussed on the origin of anisotropy of grain growth due to the migration of {110} plane-matching boundaries in Fe-3%Si alloy. The most recent theoretical work on the distribution of plane-matching boundaries in solids with different crystal structures was found to be useful for the understanding of nucleation and growth during α/γ phase transformation.  相似文献

A three-dimensional (3D) digital holographic display system with image processing is presented. By use of phase-shifting digital holography, we obtain the complex amplitude of a 3D object at a recording plane. Image processing techniques are introduced to improve the quality of the reconstructed 3D object or manipulate 3D objects for elimination and addition of information by modifying the complex amplitude. The results show that the information processing is effective in such manipulations of 3D objects. We also show a fast recording system of 3D objects based on phase-shifting digital holography for display with image processing. The acquisition of 3D object information at 500 Hz is demonstrated experimentally.  相似文献

ARV1 is involved in regulating lipid homeostasis but also in the biosynthesis of glycosylphosphatidylinositol (GPI) in Saccharomyces cerevisiae. Here, we examined whether human ARV1 can complement the role of yeast ARV1 in GPI biosynthesis. Overexpression of human ARV1 could rescue the phenotypes associated with GPI anchor synthesis defect in the yeast arv1Δ mutant. The results suggest that Arv1 function in GPI biosynthesis may be conserved in all eukaryotes, from yeast to humans. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

To increase aerodynamic performance, the geometric porosity of a ringsail spacecraft parachute canopy is sometimes increased, beyond the “rings” and “sails” with hundreds of “ring gaps” and “sail slits.” This creates extra computational challenges for fluid–structure interaction (FSI) modeling of clusters of such parachutes, beyond those created by the lightness of the canopy structure, geometric complexities of hundreds of gaps and slits, and the contact between the parachutes of the cluster. In FSI computation of parachutes with such “modified geometric porosity,” the flow through the “windows” created by the removal of the panels and the wider gaps created by the removal of the sails cannot be accurately modeled with the Homogenized Modeling of Geometric Porosity (HMGP), which was introduced to deal with the hundreds of gaps and slits. The flow needs to be actually resolved. All these computational challenges need to be addressed simultaneously in FSI modeling of clusters of spacecraft parachutes with modified geometric porosity. The core numerical technology is the Stabilized Space–Time FSI (SSTFSI) technique, and the contact between the parachutes is handled with the Surface-Edge-Node Contact Tracking (SENCT) technique. In the computations reported here, in addition to the SSTFSI and SENCT techniques and HMGP, we use the special techniques we have developed for removing the numerical spinning component of the parachute motion and for restoring the mesh integrity without a remesh. We present results for 2- and 3-parachute clusters with two different payload models.  相似文献
