Transglutaminase (TGase) was separated from the culture broth of an isolated strain of Streptoverticillium mobaraense. The crude enzyme was prepared by centrifugation, ultrafiltration, precipitation by alcohol, centrifugation and freeze‐drying. The yield after these processes was 65–70%. Then the enzyme was purified to homogeneity by chromatography on CM‐cellulose and Sephadex G‐75 on which the yields were about 70% and 80%, respectively; the purified folds reached 2.5–4.7 and 1.08–2.06, respectively. The molecular weight of this TGase was 39,500–40,100 Da by gel filtration chromatography. Optimum enzyme activity was observed in the pH range of 5.0–7.0, and it was maintained stable at 20–40C. The optimal temperature and pH was 52C and 6.0, respectively. At 1 mM and 5 mM metal ion or inhibitors concentration, TGase activity was strongly inhibited by Zn2+ and NEM, and not affected obviously by Ba2+, Ca2+, Co2+, Cu2+, Fe2+, Fe3+, Mg2+, Mn2+, Na+ as well as PMSF and EDTA. The effects of these additions on this TGase were compared with those of other microbial TGases.  相似文献

The rheology of Al-30Si-5Cu and Al-30Si-5Cu-2Mg spray-formed alloys has been characterized after holding at various times and temperatures, using semisolid rapid compression to obtain load-displacement curves. Microstructures were examined after partial remelting and compression in an attempt to understand the flow behavior. The initial resistance to flow generally increased with holding time at temperature. This increase was attributed to the increased strength of a three-dimensional (3-D) solid silicon network present in these alloys in the semisolid state, with the initial resistance to flow smaller in the magnesium-containing alloys. With increased temperature, the initial resistance decreased due to increased liquid content and grain spheroidization. The viscosity vs shear rate response was studied using the approach of Laxmanan and Flemings. The materials shear thinned rapidly during the test conditions, in which the shear rate rapidly increased from approximately 2.5 to 92 s−1. The results were fitted to a power-law model, and the viscosities showed a drop of approximately two to four orders of magnitude when extrapolated to a shear rate of 1000 s−1 (a typical value in actual component manufacture). The rapid compression test, which is close to conditions present in component manufacture, was shown to have more in common with a shear rate jump experiment, rather than steady-state viscometry.  相似文献

This paper presents a valve-regulated lead acid (VRLA) battery reserve life estimation scheme. The scheme is adaptive in both type and frequency of involvement. The scheme is based on capacity trending with the support of a number of state-of-health (SOH) indicators. These SOH indicators include accumulated thermal stress, float voltage, and indicators acquired from the start-of-discharge (coup de fouet) region. An implementation test case is given, indicating that the scheme is capable of providing an accurate estimation of reserve life long before the end of life. Furthermore, the estimation accuracy improves as the end of battery life approaches.  相似文献

We study learning of predicate logics formulas from “elementary facts,” i.e. from the values of the predicates in the given model. Several models of learning are considered, but most of our attention is paid to learning with belief levels. We propose an axiom system which describes what we consider to be a human scientist's natural behavior when trying to explore these elementary facts. It is proved that no such system can be complete. However we believe that our axiom system is “practically” complete. Theorems presented in the paper in some sense confirm our hypothesis.  相似文献

A new type of bottom‐emission electroluminescent device is described in which a metal oxide is used as the electron‐injecting contact. The preparation of such a device is simple. It consists of the deposition of a thin layer of a metal oxide on top of an indium tin oxide covered glass substrate, followed by the solution processing of the light‐emitting layer and subsequently the deposition of a high‐workfunction (air‐stable) metal anode. This architecture allows for a low‐cost electroluminescent device because no rigorous encapsulation is required. Electroluminescence with a high brightness reaching 5700 cd m–2 is observed at voltages as low as 8 V, demonstrating the potential of this new approach to organic light‐emitting diode (OLED) devices. Unfortunately the device efficiency is rather low because of the high current density flowing through the device. We show that the device only operates after the insertion of an additional hole‐injection layer in between the light‐emitting polymer (LEP) and the metal anode. A simple model that explains the experimental results and provides avenues for further optimization of these devices is described. It is based on the idea that the barrier for electron injection is lowered by the formation of a space–charge field over the metal‐oxide–LEP interface due to the build up of holes in the LEP layer close to this interface.  相似文献

A tri-gate bulk MOSFET design utilizing a low-aspect-ratio channel is proposed to provide an evolutionary pathway for CMOS scaling to the end of the roadmap. 3-D device simulations indicate that this design offers the advantages of a multi-gate FET (reduced variability in performance and improved scalability) together with the advantages of a conventional planar MOSFET (low substrate cost and capability for dynamic threshold-voltage control).  相似文献

We address the problem of localizing and obtaining high-resolution footage of the people present in a scene. We propose a biologically-inspired solution combining pre-attentive, low-resolution sensing for detection with shiftable, high-resolution, attentive sensing for confirmation and further analysis. The detection problem is made difficult by the unconstrained nature of realistic environments and human behaviour, and the low resolution of pre-attentive sensing. Analysis of human peripheral vision suggests a solution based on integration of relatively simple but complementary cues. We develop a Bayesian approach involving layered probabilistic modeling and spatial integration using a flexible norm that maximizes the statistical power of both dense and sparse cues. We compare the statistical power of several cues and demonstrate the advantage of cue integration. We evaluate the Bayesian cue integration method for human detection on a labelled surveillance database and find that it outperforms several competing methods based on conjunctive combinations of classifiers (e.g., Adaboost). We have developed a real-time version of our pre-attentive human activity sensor that generates saccadic targets for an attentive foveated vision system. Output from high-resolution attentive detection algorithms and gaze state parameters are fed back as statistical priors and combined with pre-attentive cues to determine saccadic behaviour. The result is a closed-loop system that fixates faces over a 130 deg field of view, allowing high-resolution capture of facial video over a large dynamic scene.  相似文献

In this note, we investigate different concepts of nonlinear identifiability in the generic sense. We work in the linear algebraic framework. Necessary and sufficient conditions are found for geometrical identifiability, algebraic identifiability and identifiability with known initial conditions. Relationships between different concepts are characterized. Constructive procedures are worked out for both generic geometrical and algebraic identifiability of nonlinear systems. As an application of the theory developed, we study the identifiability properties of a four dimensional model of HIV/AIDS. The questions answered in this study include the minimal number of measurement of the variables for a complete determination of all parameters and the best period of time to make such measurements. This information will be useful in formulating guidelines for clinical practice.  相似文献

ZnO thin film was deposited on various metal electrodes by reactive sputtering, and c-axis preferred orientation of the film has been studied. ZnO, which has high piezoelectricity, is promising for oscillators or filter devices such as surface acoustic wave (SAW) device, gas sensor, and film bulk acoustic resonator (FBAR). But, for the application of ZnO film for these devices, the film should be grown with c-axis normal to the electrode. In this study, Pt, Al, and Au were deposited on Si wafer, and the surface roughness and crystal structure of the ZnO film on the electrode were investigated using AFM, scanning electron microscopy (SEM), and X-ray diffraction (XRD). Columnar structures of ZnO films were grown with c-axis normal to all electrodes, and among them Pt electrode showed the highest preferred orientation of ZnO film.  相似文献
