The skin is the largest organ in the human body, comprising the main barrier against the environment. When the skin loses its integrity, it is critical to replace it to prevent water loss and the proliferation of opportunistic infections. For more than 40 years, tissue-engineered skin grafts have been based on the in vitro culture of keratinocytes over different scaffolds, requiring between 3 to 4 weeks of tissue culture before being used clinically. In this study, we describe the development of a polymerizable skin hydrogel consisting of keratinocytes and fibroblast entrapped within a fibrin scaffold. We histologically characterized the construct and evaluated its use on an in vivo wound healing model of skin damage. Our results indicate that the proposed methodology can be used to effectively regenerate skin wounds, avoiding the secondary in vitro culture steps and thus, shortening the time needed until transplantation in comparison with other bilayer skin models. This is achievable due to the instant polymerization of the keratinocytes and fibroblast combination that allows a direct application on the wound. We suggest that the polymerizable skin hydrogel is an inexpensive, easy and rapid treatment that could be transferred into clinical practice in order to improve the treatment of skin wounds.  相似文献

Web mapping has become a popular way of distributing interactive digital maps over the internet. Instead of dynamically generating map images on the fly, those can be pre-generated and served from a server-side cache for faster retrieval. However, these caches can grow unmanageably in size when the cartography covers mid to large areas for multiple rendering scales. This forces modest organizations to use partial caches containing just a subset of the total tiles, and makes their services less attractive than other mapping services like Google Maps or Microsoft Bing Maps. This work proposes a neural-network-based intelligent system that predicts which areas are likely to be requested in the future from a catalog of geographic features and a short history of past requests. These priority regions can be used by a tile prefetching policy to achieve an optimal population of the cache. Neural networks are trained and validated using supervised learning with real data-sets from a public nation-wide web map service. Trace-driven simulations demonstrate that accurate long-term predictions, up to 90% in terms of cache-hit ratio, can be obtained with the proposed model by prefetching a low fraction, only the 20% of the total tiles, and with a short training period.  相似文献

Model management is essential for coping with the complexity introduced by the increasing number and varied nature of artifacts involved in model-driven engineering-based projects. Global model management (GMM) addresses this issue by enabling the representation of artifacts, particularly transformation composition and execution, within a model called a megamodel. Type information about artifacts can be used for preventing type errors during execution. Built on our previous work, in this paper we present the core elements of a type system for GMM that improves its original typing approach and enables both typechecking and type inference on artifacts within a megamodel. This type system is able to deal with non-trivial situations such as the use of higher order transformations. We also present a prototypical implementation of such a type system.  相似文献

The influence of high intensity pulsed electric fields (HIPEF) treatment (35 kV/cm, bipolar 4 μs square wave pulses at 200 Hz) on the inactivation of Listeria innocua in fruit juice–whole (FJ–WM) or skim milk (FJ–SM) beverages was assessed. As well, the effects of HIPEF or conventional thermal (90 °C 60 s) processing in terms of microbial and enzymatic stability and changes on physicochemical parameters of the beverages during refrigerated storage were also studied. HIPEF and thermal processing ensured the microbial stability of the beverages during 56 days at 4 °C without significant changes on pH, acidity and soluble solid content values. Thermal processing was more effective than HIPEF in reducing Pectin Methyl Esterase (PME) activity in FJ–SM and FJ–WM beverages since its activity was reduced by 70.66% and 87.33%, respectively. With regards to Polygalacturonase (PG), HIPEF treatment reduced by 26.92% and 20.93% the enzyme activity in the FJ–WM and FJ–SM beverages, respectively, whereas thermal treatment did not inactivate PG in both beverages. HIPEF or thermally-treated samples showed higher viscosity than the untreated ones, being this increment more pronounced in the beverages with whole milk. Therefore, HIPEF processing may be a feasible technology in order to obtain shelf stable and fresh-like fruit juice-milk beverages.  相似文献

A TaqMan real-time polymerase chain reaction (PCR) method was developed for specific detection of bovine, ovine and caprine processed animal protein (PAP) in industrial feedstuffs. The method uses species-specific primers and probes targeting short mitochondrial D-loop sequences, and a positive amplification control based on 18S rRNA gene. The applicability of the real-time PCR protocol was assessed through analysis of 126 industrial feed samples that were manufactured to reproduce rendering processes of commercial feeds destined for farmed animals. The assay successfully classified samples as positive or negative according to the ruminant composition, enabling qualitative detection of banned material in feeds at levels as low as 0.1%. Although the method provides quantitative potential, results suggest that the real quantitative capability of the assay is limited by the existing variability in terms of composition and processing treatments of the feeds, which affect the amount and quality of amplifiable DNA.  相似文献

Objective: To use multitrait analysis to determine the measurement structure of the Caregiver Empowerment Scale (CES). Participants: An American sample of 87 adult primary family caregivers of persons with traumatic brain injury (TBI). Results: A four-factor structure was identified including factor 1 (Advocacy Self-Efficacy), factor 2 (Community Self-Efficacy), factor 3 (Caregiver Self-Efficacy), and factor 4 (Personal Self-Efficacy). Conclusions: The CES provides clinicians and researchers a means to assess self-perceived coping abilities of family caregivers of persons with TBI. Objective: The authors conducted a meta-analysis of empirical studies investigating associations between indices of posttraumatic stress disorder (PTSD) and intimate relationship problems to empirically synthesize this literature. Method: A literature search using PsycINFO, Medline, Published International Literature on Traumatic Stress (PILOTS), and Dissertation Abstracts was performed. The authors identified 31 studies meeting inclusion criteria. Results: True score correlations (ρ) revealed medium-sized associations between PTSD and intimate relationship discord (ρ = .38, N = 7,973, K = 21), intimate relationship physical aggression perpetration (ρ = .42, N = 4,630, K = 19), and intimate relationship psychological aggression perpetration (ρ = .36, N = 1,501, K = 10). The strength of the association between PTSD and relationship discord was higher in military (vs. civilian) samples, and when the study was conducted in the United States (vs. other country), and the study represented a doctoral dissertation (vs. published article). The strength of the association between PTSD and physical aggression was higher in military (vs. civilian) samples, males (vs. females), community (vs. clinical) samples, studies examining PTSD symptom severity (vs. diagnosis), when the physical aggression measure focused exclusively on severe violence (vs. a more inclusive measure), and the study was published (vs. dissertation). For the PTSD–psychological aggression association, 98% of the variance was accounted for by methodological artifacts such as sampling and measurement error; consequently, no moderators were examined in this relationship. Conclusions: Findings highlight a need for the examination of models explaining the relationship difficulties associated with PTSD symptomatology and interventions designed to treat problems in both areas. Although several brief instruments are available for the emotional intelligence (EI) construct, their conceptual coverage tends to be quite limited. One notable exception is the short form of the Emotional Quotient Inventory (EQ-i:S), which measures multiple EI dimensions in addition to a global EI index. Despite the unique advantage offered by the inventory, psychometric properties of the EQ-i:S scores have not yet been systematically evaluated. Such an evaluation was the main goal of the present investigation. Using data from 2,508 undergraduates, the authors conducted 2 studies involving factor structure, internal reliability, 6-month temporal stability, and construct validity of the EQ-i:S responses, both for the total EQ scale and for each constituent dimension. The results supported the multidimensional measurement structure of the EQ-i:S, with each dimension producing internally consistent, temporally stable, and theoretically meaningful responses. Scores on the EQ-i:S were associated more strongly with performance on an ability test of EI and with a conceptually similar construct of alexithymia than with the broader dimensions of basic personality and explained nontrivial amounts of incremental variance in the criterion symptoms of attention deficit/hyperactivity disorder. Moreover, scores on each EQ-i:S dimension exhibited unique patterns of associations with the validation variables.
