We compared the primacy of affective versus semantic categorization by using forced-choice saccadic and manual response tasks. Participants viewed paired emotional and neutral scenes involving humans or animals flashed rapidly in extrafoveal vision. Participants were instructed to categorize the targets by saccading toward the location occupied by a predefined target scene. The affective task involved saccading toward an unpleasant or pleasant scene, and the semantic task involved saccading toward a scene containing an animal. Both affective and semantic target scenes could be reliably categorized in less than 220 ms, but semantic categorization was always faster than affective categorization. This finding was replicated with singly, foveally presented scenes and manual responses. In comparison with foveal presentation, extrafoveal presentation slowed down the categorization of affective targets more than that of semantic targets. Exposure threshold for accurate categorization was lower for semantic information than for affective information. Superordinate-, basic-, and subordinate-level semantic categorizations were faster than affective evaluation. We conclude that affective analysis of scenes cannot bypass object recognition. Larvae of nine species of sawflies (Symphyta) were fed with the foliage of three birch species, after which the larval hemolymph composition was studied by HPLC–DAD and HPLC–ESI–MS. The hemolymph of sawfly larvae contained high concentrations of flavonol oligoglycosides (tri-, tetra-, penta-, and hexaglycosides) that could not be found in the larval foliar diet. In addition, there were significant between-sawfly species differences in both flavonoid composition and concentration (from 0.6 to 12.3 mg/ml) of the hemolymph. This suggested that the studied species have different biosynthetic activities for the synthesis of flavonoid oligoglycosides. Variation in the foliar diets did not cause differences in the hemolymph composition. Our hypothesis is that sawflies use foliar flavonoid monoglycosides rather than flavonoid aglycones to produce these new types of oligoglycosides. These findings open up new possibilities for understanding the more holistic role of flavonoids in insect biochemistry and complex interactions between plants and herbivores. 相似文献

Betula pubescens bud flavonoid aglycones reportedly have negative effects on the performance of first instar Epirrita autumnata and, thus, may defend birch leaves from larval defoliation. We hypothesized that the detrimental effects of these lipophilic flavonoids on larvae are due to their high levels in birch buds and/or the inability of naïve neonates to glycosylate them, which we have shown to occur in fifth instars. To test the latter hypothesis, we investigated the biochemical transformation of bud flavonoids in first instar E. autumnata. We found that newly hatched larvae have the ability to glycosylate birch bud/leaf flavonoid aglycones into corresponding glycosides. Moreover, we suggest that glycosylation may depend upon the chemical character of the aglycone and is an important factor in the performance of first instars. 相似文献

The chemical reactions used to make antibody-DNA conjugates in many immunoassays diminish antigen-binding activity and yield heterogeneous products. Here, we address these issues by developing an antibody-based rolling circle amplification (RCA) strategy using a fusion of φX174 gene A* protein and Z(mab25) (A*-Zmab). The φX174 gene A* protein is an enzyme that can covalently link with DNA, while the Z(mab25) protein moiety can bind to specific species of antibodies. The DNA in an A*-Zmab conjugate was attached to the A* protein at a site chosen to not interfere with protein function, as determined by enzyme-linked immunosorbent assay (ELISA) and gel mobility shift analysis. The novel A*-Zmab-DNA conjugate retained its binding capabilities to a specific class of murine immunoglobulin γ1 (IgG1) but not to rabbit IgG. This indicates the generality of the A*-Zmab-based immuno-RCA assay that can be used in-sandwich ELISA format. Moreover, the enzymatic covalent method dramatically increased the yields of A*-Zmab-DNA conjugates up to 80% after a 15 min reaction. Finally, sensitive detection of human interferon-γ (IFN-γ) was achieved by immuno-RCA using our fusion protein in sandwich ELISA format. This new approach of the use of site-specific enzymatic DNA conjugation to proteins should be applicable to fabrication of novel immunoassays for biosensing. 相似文献

Lead titanate thin films were deposited by atomic layer deposition on Si(100) using Ph4Pb and Ti(O-i-Pr)4 as metal precursors and O3 and H2O as oxygen sources. The influence of the Ti : Pb precursor pulsing ratio on the film growth, stoichiometry and quality was studied at two different temperatures, i.e. 250 and 300 °C. Uniform and stoichiometric films were obtained using a Ti : Pb precursor pulsing ratio of 1 : 10 at 250 °C or 1 : 28 at 300 °C. The as-deposited films were amorphous but the crystalline PbTiO3 phase was obtained by rapid thermal annealing at 600-900 °C both in N2 and O2 ambient. The noradrenaline (NA)-dependent hindlimb flexor reflex that can be elicited by pinching the foot of acutely spinalized rats given nialamide-DOPA or clonidine was evaluated different time intervals (14 days-6 months) after intracisternal injections of 6-OH-dopamine (6-OH-DA) and correlated to the degree of bulbospinal catecholamine (CA) denervation as seen by Falck-Hillarp fluorescence histochemistry. Six and 14 days after 6-OH-DA, when almost all NA nerve terminals of the spinal cord had degenerated, the NA receptors where supersensitive to stimulation with clonidine as evidenced by an increased flexor reflex. This supersensitivity gradually disappeared as new nerve terminals were formed in the grey matter of the spinal cord during the following 3-6 months. The supersensitivity phenomenon 14 days after 6-OH-DA could also be demonstrated by L-DOPA given to animals pretreated with 100 mg/kg nialamide. Using this relatively low dose of nialamide, almost no reflex response was seen in the control group. Using a higher degree of monoaminoxidase inhibition (nialamide 200 mg/kg) also non-supersensitive, NA receptors became maximally stimulated. Therefore, 6-OH-DA treated rats now showed a weaker reflex than controls, the reflex response being directly correlated to the number of nerve terminals present that could form NA from the precursor. Using 5,6-dihydroxytryptamine, which selectively destroys 5-hydroxytryptamine (5-HT) nerves, it was shown that the flexor reflex changes were specifically related to the NA nerves and unchanged by the simultaneous presence or absence of 5-HT nerve terminals. This was further supported by the finding of a correlation between amount of nerve terminals and flexor reflex responses in individual animals, especially at longer survival times both in the clonidine and the nialamide-DOPA experiments. 相似文献

The microbiological quality of 30 production lots of charcoal-broiled river lampreys was studied at three lamprey processing plants (plants A, B, and C). Samples were taken directly after charcoal broiling and stored at 22 and 3 degrees C. Lampreys were examined on the day of manufacture, and those kept at 22 degrees C were examined every second day for 6 days. Samples kept at 3 degrees C were examined every fourth day for up to 24 days. On the production day, the mean aerobic plate counts (APCs) for broiled lampreys from plants A, B, and C were 2.29 log CFU/g, 1.88 log CFU/g, and undetectable (1.67 log CFU/g), respectively. At 22 degrees C, the mean APCs for samples from plants A, B, and C increased markedly within 4 days, and after 6 days the counts for samples from these plants were 8.56, 5.04, and 6.23 log CFU/g, respectively. Chilling and storage at 3 degrees C remarkably improved the shelf life of the product. The levels of bacteria in charcoal-broiled river lampreys from plant A were higher than those in lampreys from plants B and C. No significant increases in APCs were observed during storage at 3 degrees C for 24 days; mean APCs did not exceed 2.80 log CFU/g for samples from any plant. Staphylococcus aureus was found in two samples. No lactic acid bacteria, thermotolerant coliforms, enterococci, Clostridium perfringens, or Listeria monocytogenes was detected.
