Reservosomes are late endosomes present only in members of the Schizotrypanum subgenus of the Trypanosoma genus and are defined as the site of storage of endocytosed macromolecules and lysosomal enzymes. They have been extensively described in Trypanosoma cruzi epimastigote: are bounded by a membrane unit, present an electron-dense protein matrix with electron-lucent lipid inclusions, being devoid of inner membranes. Here we performed a detailed ultrastructural analysis of these organelles using a variety of electron microscopy techniques, including ultrathin sectioning, uranyl acetate stained preparations, and freeze fracture, either in intact epimastigotes or in isolated reservosomes. New informations were obtained. First, both isolated and in situ reservosomes presented small profiles of inner membranes that are morphologically similar to the membrane surrounding the organelle. In uranyl acetate stained preparations, internal membrane profiles turned out to be longer than they appeared in ultrathin section images and traversed the organelle diameter. Internal vesicles were also found. Second, endocytosed cargo are not associated with internal vesicles and reach reservosomes on board of vesicles that fuse with the boundary membrane, delivering cargo directly into reservosome lumen. Third, electron-lucent bodies with saturated lipid core surrounded by a membrane monolayer and with unusual rectangular shape were also observed. Fourth, it was possible to demonstrate the presence of intramembranous particles on the E face of both internal vesicles and the surrounding membrane. Collectively, these results indicate that reservosomes have a complex internal structure, which may correlate with their multiple functions.  相似文献

The design and performance of two orthogonal extraction time-of-flight mass spectrometers are reported that were adapted to existing focused ion beam microscopes for secondary ion mass spectrometry. The performances of these designs were compared to that of a prototype previously described by our group. The differences include newly designed transfer ion optics and in the use of a larger microscope chamber. The two new prototypes allow a mass resolving power of either 600 Th/Th (compact design) or 3000 Th/Th (high resolution design) while simultaneously achieving a lateral spatial resolution of less than 50 nm. The spectrometers and their performance (effective ion yield, mass resolving power, lateral, and depth resolution) are described and compared. Additionally, example applications are presented with multivariate statistical methods to visualize the data sets. Both time-of-flight mass analyzers use orthogonal extraction which avoids the need to pulse the primary ion beam, and the of use monoisotopic gallium to preserve the mass resolution. The goal of the design was a cost-effective accessory to augment typical focused ion beam-scanning electron microscopy applications as an alternative to the cost of a dedicated secondary ion mass spectrometer. The modified instrument allows excellent non destructive imaging and easy sample access, and benefits from the presence of complementary non destructive analytical and imaging techniques that exploit the presence of an electron microscope.  相似文献

For a prime number \(p\) , Bergman (Israel J Math 18:257–277, 1974) established that \(\mathrm {End}(\mathbb {Z}_{p} \times \mathbb {Z}_{p^{2}})\) is a semilocal ring with \(p^{5}\) elements that cannot be embedded in matrices over any commutative ring. In an earlier paper Climent et al. (Appl Algebra Eng Commun Comput 22(2):91–108, 2011), the authors presented an efficient implementation of this ring, and introduced a key exchange protocol based on it. This protocol was cryptanalyzed by Kamal and Youssef (Appl Algebra Eng Commun Comput 23(3–4):143–149, 2012) using the invertibility of most elements in this ring. In this paper we introduce an extension of Bergman’s ring, in which only a negligible fraction of elements are invertible, and propose to consider a key exchange protocol over this ring.  相似文献

In this work three mathematical models for methane steam reforming in membrane reactors were developed. The first one is a steady state, non isothermal, non isobaric and one dimensional model derived from material and energy balances and validated using experimental data from the literature. It is referred as full model. The influence of two different intrinsic kinetics available, as well as, the influence of five important parameters on methane conversion (XCH4_{mathrm{CH}_{4}}) and hydrogen recovery (YH2_{mathrm{H}_{2}}) were parametrically evaluated through simulations. The second model, referred as meta-model, was obtained though the response surface technique. This meta-model was included into a constrained optimization problem solved using NPSOL. The third model, referred as a simplified model, takes into account only mass balances from the full model. Using this model, a gradient based method (DIRCOL) was used to perform the optimization of the sum of methane conversion and hydrogen recovery. High methane conversions and hydrogen recoveries were reached through these methodologies.  相似文献

It is well known that the use of passive energy dissipation devices such as friction dampers reduces the dynamic response of structures subjected to earthquakes. However, the parameters of each damper as well as the best position of these devices in the structure remain difficult to determine. Although articles on optimum design of tuned mass dampers and viscous dampers have been published, there is a lack of studies on the optimization of friction dampers. In previous contributions, the authors proposed a method for optimum design of this kind of damper. The proposed method is very useful; however, the computational time required is high. Thus, in this article, a new methodology for the simultaneous optimization of placement and forces of friction dampers is proposed. As this new method is developed in the frequency domain, the computational time is considerably reduced. For this purpose, the search group algorithm, recently developed by the authors, is employed, which is able to deal with optimization problems involving mixed discrete and continuous variables. For illustrative purposes, a six-storey shear building is analysed. Forces and positions of friction dampers are the design variables, while the objective function is to minimize the root mean square displacement at the top of the building. The results showed the excellent performance of the proposed method, reducing the root mean square displacement by more than 82%, with only three friction dampers and in a relatively short computational time.  相似文献

Bioactive glasses represent an interesting class of bone substitute's biomaterials. The present study investigated the repair of bone defects filled with a novel bioactive vitroceramic (Biosilicate(?)), alone or in association with particulate autogenous bone grafts in calvaria defects of rabbits. After 7, 14, and 30 days the specimens were retrieved for histological, histomorphometric and immunohistochemistry analysis. Satisfactory bone formation was observed in all groups, and direct bone-biomaterial surface was noted. Histomorphometric assessment did not show statistically significant differences in bone formation among the groups and periods, except for BG group at day 14. Immunoexpression of Runx-2 was similar among the groups containing the graft and the biomaterial, being more intense than in control group. Similar result was observed for VEGF expression, especially in the last experimental period. These results revealed that Biosilicate(?) presented a favorable behavior, comparable to autogenous bone graft.  相似文献

The aim of this article is to analyze the influence of microstructural parameters on the mechanical properties and corrosion behavior of a hypoeutectic Al–Ni alloy. Experimental results include secondary dendrite arm spacing, corrosion potential, current density, pitting potential, ultimate tensile strength and yield strength. It was found that cooling rates during solidification of about 0.6 °C/s and 8 °C/s can provide secondary dendritic spacings of 7 μm and 16 μm, respectively. Although the microstructure having their phases finely and homogeneously distributed was shown to induce better mechanical properties and higher pitting potential, its general corrosion resistance decreased when compared with the corresponding results of the coarser microstructure.  相似文献

The aim of this article is to compare the electrochemical corrosion resistance of two as-cast Al–6 wt.% Cu–1 wt.% Si and Al–8 wt.% Cu–3 wt.% Si alloys considering both the solutes macrosegregation profiles and the scale of the microstructure dendritic arrays. A water-cooled unidirectional solidification system was used to obtain the as-cast samples. Electrochemical impedance spectroscopy (EIS) and potentiodynamic anodic polarization techniques were used to analyze the corrosion resistance in a 0.5 M NaCl solution at 25 °C. It was found that the Al–8Cu–3Si alloy has better electrochemical corrosion resistance than the Al–6Cu–1Si alloy for any position along the casting length. At the castings regions where the Cu inverse profile prevailed (up to about 10 mm from the castings surface) the corrosion current density decreased up to 2.5 times with the decrease in the secondary dendrite arm spacing.  相似文献
