A general methodology for patterning of multiple protein ligands with lateral dimensions below those of single cells is described. It employs dip pen nanolithography (DPN) patterning of DNA oligonucleotides which are then used as capture strands for DNA‐directed immobilization (DDI) of oligonucleotide‐tagged proteins. This study reports the development and optimization of PEG‐based liquid ink, used as carrier for the immobilization of alkylamino‐labeled DNA oligomers on chemically activated glass surfaces. The resulting DNA arrays have typical spot sizes of 4–5 μm with a pitch of 12 μm micrometer. It is demonstrated that the arrays can be further functionalized with covalent DNA‐streptavidin (DNA‐STV) conjugates bearing ligands recognized by cells. To this end, biotinylated epidermal growth factor (EGF) is coupled to the DNA‐STV conjugates, the resulting constructs are hybridized with the DNA arrays and the resulting surfaces used for the culturing of MCF‐7 (human breast adenocarcinoma) cells. Owing to the lateral diffusion of transmembrane proteins in the cell's plasma membrane, specific recruitment and concentration of EGF receptor can be induced specifically at the sites where the ligands are bound on the solid substrate. This is a clear demonstration that this method is suitable for precise functional manipulations of subcellular areas within living cells. 相似文献

Adaptive algorithms are important tools for efficient finite‐element mesh design. In this paper, an error controlled adaptive mesh‐refining algorithm is proposed for a non‐conforming low‐order finite‐element method for the Reissner–Mindlin plate model. The algorithm is controlled by a reliable and efficient residual‐based a posteriori error estimate, which is robust with respect to the plate's thickness. Numerical evidence for this and the efficiency of the new algorithm is provided in the sense that non‐optimal convergence rates are optimally improved in our numerical experiments. In this work, we use dark-field microscopy to observe a new plasmon resonance effect for a single silver nanocube in which the plasmon line shape has two distinct peaks when the particles are located on a glass substrate. The dependence of the resonance on nanocube size and shape is characterized, and it is found that the bluer peak has a higher figure of merit for chemical sensing applications than that for other particle shapes that have been studied previously. Comparison of the measured results with finite difference time domain (FDTD) electrodynamics calculations enables us to confirm the accuracy of our spectral assignments. 相似文献

Affective forecasting, experienced affect, and recalled affect were compared in younger and older adults during a task in which participants worked to win and avoid losing small monetary sums. Dynamic changes in affect were measured along valence and arousal dimensions, with probes during both anticipatory and consummatory task phases. Older and younger adults displayed distinct patterns of affect dynamics. Younger adults reported increased negative arousal during loss anticipation and positive arousal during gain anticipation. In contrast, older adults reported increased positive arousal during gain anticipation but showed no increase in negative arousal on trials involving loss anticipation. Additionally, younger adults reported large increases in valence after avoiding an anticipated loss, but older adults did not. Younger, but not older, adults exhibited forecasting errors on the arousal dimension, underestimating increases in arousal during anticipation of gains and losses and overestimating increases in arousal in response to gain outcomes. Overall, the findings are consistent with a growing literature suggesting that older people experience less negative emotion than their younger counterparts and further suggest that they may better predict dynamic changes in affect. Retained austenite transformation was studied for a 5 wt pct Cr cold work tool steel tempered at 798 K and 873 K (525 °C and 600 °C) followed by cooling to room temperature. Tempering cycles with variations in holding times were conducted to observe the mechanisms involved. Phase transformations were studied with dilatometry, and the resulting microstructures were characterized with X-ray diffraction and scanning electron microscopy. Tempering treatments at 798 K (525 °C) resulted in retained austenite transformation to martensite on cooling. The martensite start (M s ) and martensite finish (M f ) temperatures increased with longer holding times at tempering temperature. At the same time, the lattice parameter of retained austenite decreased. Calculations from the M s temperatures and lattice parameters suggested that there was a decrease in carbon content of retained austenite as a result of precipitation of carbides prior to transformation. This was in agreement with the resulting microstructure and the contraction of the specimen during tempering, as observed by dilatometry. Tempering at 873 K (600 °C) resulted in precipitation of carbides in retained austenite followed by transformation to ferrite and carbides. This was further supported by the initial contraction and later expansion of the dilatometry specimen, the resulting microstructure, and the absence of any phase transformation on cooling from the tempering treatment. It was concluded that there are two mechanisms of retained austenite transformation occurring depending on tempering temperature and time. This was found useful in understanding the standard tempering treatment, and suggestions regarding alternative tempering treatments are discussed. 相似文献

Sending memorable messages to the old: Age differences in preferences and memory for advertisements.
Socioemotional selectivity theory holds that people of different ages prioritize different types of goals. As people age and increasingly perceive time as finite, they attach greater importance to goals that are emotionally meaningful. Because the goals that people pursue so centrally influence cognition, the authors hypothesize that persuasive messages, specifically advertisements, would be preferred and better remembered by older adults when they promise to help realize emotionally meaningful goals, whereas younger adults would not show this bias. The authors also predict that modifying time perspective would reduce age differences. Findings provide qualified support for each of these predictions. Three studies investigated whether self-affirmation can proceed without awareness, whether people are aware of the influence of experimental self-affirmations, and whether such awareness facilitates or undermines the self-affirmation process. The authors found that self-affirmation effects could proceed without awareness, as implicit self-affirming primes (utilizing sentence-unscrambling procedures) produced standard self-affirmation effects (Studies 1 and 3). People were generally unaware of self-affirmation’s influence, and self-reported awareness was associated with decreased impact of the affirmation (Studies 1 and 2). Finally, affirmation effects were attenuated when people learned that self-affirmation was designed to boost self-esteem (Study 2) or told of a potential link between self-affirmation and evaluations of threatening information (Study 3). Together, these studies suggest not only that affirmation processes can proceed without awareness but also that increased awareness of the affirmation may diminish its impact. This study examined psychological and physiological stress, as well as muscle tension and musculoskeletal symptoms, among 72 female supermarket cashiers. Stress levels were found to be significantly elevated at work, as reflected in the catecholamines, blood pressure, heart rate, electromyographic (EMG) activity, and self-reports. Fifty cashiers (70%) suffering from neck-shoulder pain (trapezius myalgia) were found to have higher EMG activity at work and reported more tension after work. Women who kept a diary for 1 week and reported more musculoskeletal pain (above the median) were older, had higher blood pressure, and reported more work stress and psychosomatic symptoms. The elevated stress levels at work are consistent with data from workers involved in other types of repetitive tasks and can be important for the high prevalence of neck and shoulder symptoms among the cashiers. An engagement mode involves a subject (e.g., a user of information technology, or IT) who is engaged in an activity with an object in a certain manner (the mode). The purpose of this study is to develop a general model of engagement modes that may be used for understanding how IT-related activities are shaped by properties of the user and the IT object. A questionnaire involving items on IT engagement and the experience of flow was administered to 300 participants. The results supported an engagement mode (EM) model involving 5 different engagement modes (enjoying/acceptance, ambition/curiosity, avoidance/hesitation, frustration/ anxiety, and efficiency/productivity) characterized on 3 dimensions (evaluation of object, locus of control between subject and object, and intrinsic or extrinsic focus of motivation). The flow experience follows from a balance between enjoying/ acceptance and efficiency/productivity propelled by ambition/curiosity. The EM model could provide a platform for considering how IT users, IT applications, and IT environments should work together to yield both enjoyment and efficiency.
