We investigated the inhibition of Clostridium perfringens spore germination and outgrowth by two green tea extracts with low (green tea leaf powder [GTL]; 141 mg of total catechins per g of green tea extract) and high (green tea leaf extract [GTE]; 697 mg of total catechins per g of extract) catechin levels during abusive chilling of retail cooked ground beef, chicken, and pork. Green tea extracts were mixed into the thawed beef, chicken, and pork at concentrations of 0.5, 1.0, and 2.0% (wt/ wt), along with a heat-activated (75 degrees C for 20 min) three-strain spore cocktail to obtain a final concentration of approximately 3 log spores per g. Samples (5 g) of the ground beef, chicken, and pork were then vacuum packaged and cooked to 71 degrees C for 1 h in a temperature-controlled water bath. Thereafter, the products were cooled from 54.4 to 7.2 degrees C in 12, 15, 18, or 21 h, resulting in significant increases (P < 0.05) in the germination and outgrowth of C. perfringens populations in the ground beef, chicken, and pork control samples without GTL or GTE. Supplementation with 0.5 to 2% levels of GTL did not inhibit C. perfringens growth from spores. In contrast, the addition of 0.5 to 2% levels of GTE to beef, chicken, and pork resulted in a concentration-and time-dependent inhibition of C. perfringens growth from spores. At a 2% level of GTE, a significant (P < 0.05) inhibition of growth occurred at all chill rates for cooked ground beef, chicken, and pork. These results suggest that widely consumed catechins from green tea can reduce the potential risk of C. perfringens spore germination and outgrowth during abusive cooling from 54.4 to 7.2 degrees C in 12, 15, 18, or 21 h of cooling for ground beef, chicken, and pork.  相似文献

ABSTRACT:  We have evaluated bactericidal activities against Bacillus cereus , Escherichia coli O157:H7, Listeria monocytogenes , and Salmonella enterica of several antimicrobial wine recipes, each consisting of red or white wine extracts of oregano leaves with added garlic juice and oregano oil. Dose-response plots were used to determine the percentage of the recipes that resulted in a 50% decrease in colony-forming units (CFU) at 60 min (BA50). Studies designed to optimize antibacterial activities of the recipes demonstrated that several combinations of the naturally occurring plant-derived ingredients rapidly inactivated the above mentioned 4 foodborne pathogens. We also showed that (a) incubation temperature affected activities in the following order: 37 °C > 21 °C > 4 °C; (b) varying the initial bacterial concentrations from 103 to 104 to 105 CFU/well did not significantly affect BA50 values; (c) storage of 3 marinades up to 2 mo did not change their effectiveness against Salmonella enterica ; and (d) polyphenolic compounds isolated by chromatography from red wine exhibited exceptional activity at nanogram levels against 2 strains of Bacillus cereus . These observations suggest that antimicrobial wine formulations have the potential to improve the microbiological safety of foods.  相似文献

A 9-wk trial was conducted to study the performance of 24 Holstein cows during the transition period (3 wk prepartum to 6 wk postpartum). Cows were assigned to either a control or liquid-flavored (0.52 mL/kg of feed) total mixed ration in a randomized complete block design. The diets contained corn silage, alfalfa haylage, cottonseed, and a grain mix based on ground corn and soybean meal. Cows were fed to ensure 10% orts, and the diet provided (on a dry matter basis) 13% crude protein, 32% acid detergent fiber, 44% neutral detergent fiber, and 1.54 Mcal/kg of NEL prepartum and 17.5% crude protein, 30% acid detergent fiber, 40% neutral detergent fiber, and 1.57 Mcal/kg of NEL postpartum. An additional 2.3 kg of alfalfa hay was fed during the first 5 d postpartum. Weekly means of dry matter intake (DMI), milk yield, milk protein, milk fat, SNF, somatic cell counts, and body weight (BW) were analyzed using a repeated measures procedure. There was no effect of treatment on these variables, and least squares means were 16.9 and 15.7 kg/d for DMI, 38 and 35.3 kg/d for milk yield, 3.10 and 3.11% for milk protein, 3.69 and 3.74% for milk fat, 8.37 and 8.16% for SNF, 1.99 x 10(5) and 4.33 x 10(5) for somatic cell count, and 631 and 651 kg for BW for cows fed control and flavored diets, respectively. Individual cow daily DMI data were fitted to an exponential model describing pre- and postpartum feed consumption [DMI = a - b x e(-c x t), where DMI was measured in kg, a = asymptotic DMI, b = potential fractional increase in DMI, c = fractional rate of increase in DMI, and t = days prior to calving or days in milk]. Fractional rates of increase in DMI were similar: 0.139 and 0.123/d for control and flavored diets, respectively. Data for both groups were separately analyzed using multiple regression with 3.5% fat-corrected milk as the dependent variable and BW and DMI as independent variables. More BW was mobilized per unit increase in 3.5% fat-corrected milk in cows fed the control than in cows fed the flavored diet. Cows fed the control diet tended to be in more negative energy balance during early lactation than cows fed the flavored diet. It was concluded that feeding flavor improved energy balance of cows in early lactation and may reduce the risk of health or reproductive problems.  相似文献

Ratios of n-heptane (hep) to toluene (tol) affect the solubility of the asphaltenes in heavy oil extraction processes. Consequently phase changes and time after mixing n-heptane and heavy oil in toluene are important for understanding produced emulsions. The kinetics of phase change when n-heptane is added to toluene-diluted heavy oils, and the thermodynamic properties of partially deasphalted heavy oils were studied. The methods used were monitoring precipitation in time using light microscopy, quantitative asphaltenes analysis by near infrared spectroscopy, refractive index and densities measurements, and calculated solubility parameters of mixtures. At critical mass ratios of hep/tol from 1.37 to 2.0 in diluted heavy oil the precipitated asphaltene particles were observed under the microscope after lag times from 2 h to instantly. Lag times were longer at low initial oil concentration. The floc growth time decreased as heavy oil concentration in toluene increased. The growth patterns in time appeared as dots to beads (strings) to clusters (fractal-like flocs). Final wt% precipitated asphaltenes vs. mass fraction (hep+tol)/heavy oil followed sigmoidal relationships. Curves showing wt% soluble asphaltenes vs. mass fraction hep/tol after 24 h initially followed the same shape as time zero curves and diverged at the onset ratios of hep/tol. Slope for precipitated asphaltenes vs. solubility parameters curve showed a break at 16.4 MPa1/2. Linear correlations were established for concentrations of soluble asphaltenes in residual oils and density, for refractive index and density and for refractive index and solubility parameter. The latter correlation was in accordance with Lorenz-Lorentz theory. These equations provided a means by which oil density, refractive index and solubility parameter can be predicted when these measurements are difficult to measure practically.  相似文献

Porous γ-alumina with well arranged secondary mesopores has been contrived using nanosized templating units. The pore size of templated mesopores is precisely controlled as the pore shrinkage is insignificant. The primary pore diameter is ca. 4 nm and the secondary pore diameter is ca. 50 nm. The porous material was characterized using N2 adsorption/desorption, TEM, XRD and FT-IR. γ-alumina with bimodal pore size distribution shows improved intra-particle diffusion compared to γ-alumina with unimodal pore size distribution in a simple dye adsorption test. γ-alumina with different porous structures were then impregnated with vanadium oxide for catalytic effect comparison. It was perceived that secondary pores improve the styrene oxidation rate after the conversion of styrene reaches 30%.  相似文献

Medullary thyroid carcinoma (MTC) is a tumor deriving from the thyroid C cells. Vandetanib (VAN) and cabozantinib (CAB) are two tyrosine kinase inhibitors targeting REarranged during Transfection (RET) and other kinase receptors and are approved for the treatment of advanced MTC. We aim to compare the in vitro and in vivo anti-tumor activity of VAN and CAB in MTC. The effects of VAN and CAB on viability, cell cycle, and apoptosis of TT and MZ-CRC-1 cells are evaluated in vitro using an MTT assay, DNA flow cytometry with propidium iodide, and Annexin V-FITC/propidium iodide staining, respectively. In vivo, the anti-angiogenic potential of VAN and CAB is evaluated in Tg(fli1a:EGFP)y1 transgenic fluorescent zebrafish embryos by analyzing the effects on the physiological development of the sub-intestinal vein plexus and the tumor-induced angiogenesis after TT and MZ-CRC-1 xenotransplantation. VAN and CAB exert comparable effects on TT and MZ-CRC-1 viability inhibition and cell cycle perturbation, and stimulated apoptosis with a prominent effect by VAN in MZ-CRC-1 and CAB in TT cells. Regarding zebrafish, both drugs inhibit angiogenesis in a dose-dependent manner, in particular CAB shows a more potent anti-angiogenic activity than VAN. To conclude, although VAN and CAB show comparable antiproliferative effects in MTC, the anti-angiogenic activity of CAB appears to be more relevant.  相似文献

Ability of laboratory determination of a sand behaviour in static and dynamic loading conditions are influenced by (among other things): sample preparation, number of tests, size of strains, speed of loading and averaging of the errors during examination. Dynamic load per se causes accumulation of the pore water pressure and the phenomenon of stiffening-threshold-degradation which makes the understanding of the sand behaviour more difficult. The complex behaviour of granular material, i.e. sand, is caused by chemical and physical properties of individual particles and their mutual interaction. Obviously, these interrelationships could not be analysed on the basis of laboratory testing. One way to analyse it is numerically, with algorithm that takes into consideration the characteristics of individual particles as well as their interaction in the sand matrix. Discrete element method (DEM) is a numerical method which takes into consideration the discrete nature of sand and shape of particles and is used as an elementary research tool for sand behaviour. Program package PFC3D is based on DEM and allows the modelling of the laboratory equipment, materials and calibration of its micro-characteristics, based on experimental results. The research of cyclic threshold shear strain for degradation in sand includes observation and visualization of the sample preparation (creation of the material skeleton), pouring of the material (transition from liquid to meta-stable state), influence of the particle shape (interlocking, arching), consolidation (deformation of the skeleton) and development and braking of force chains through the sample. This paper explores the suitability of the selected numerical method for modelling of NGI-DSS device, calibration of the tested granular material (Nevada sand) and preparation of the sample for testing and presentation of the stiffening-threshold-degradation phenomenon.  相似文献

Direct probing of the conductivity distribution in organic materials is motivated by the need to obtain a deeper understanding of carrier behavior in organic thin-film transistors (OTFT), organic electro-luminescent devices, organic photoconductors, and organic biosensors. Here we used a near-field scanning microwave microscope to visualize conductivity profiles in OTFT channel. Applying this technique to pentacene field-effect transistors has revealed changes of the conductivity distribution in the channel arising from the development and exhaustion of an accumulated charge region. The electric field profiles, the complementary image of conductivity profiles, which are visualized by using an optical second harmonic generation method, support the results. We anticipate that direct observation by this microwave method will find wide application in measurement of carrier conductivity in organic and nonorganic materials.  相似文献
