Androgen disrupting chemicals (ADCs) are endocrine disrupting chemicals (EDCs) that mimic or antagonize the effect of physiological androgens. Microarray-based detection of altered gene expression can be used as a biomarker of EDC exposure. Therefore, the purpose of this study was to identify and compare gene expression profiles of the androgen 11-ketotestosterone (11-KT), the antiandrogen flutamide (FLU), and the antiandrogenic fungicide vinclozolin (VIN), on Qurt medaka (Oryzias latipes). Biologically effective concentrations for 11-KT (100 microg/L), VIN (100 microg/L), and FLU (1000 microg/L) determined in range-finding studies were used for exposures. The oligonucleotide microarray included 9379 probes for EDC-affected genes, medaka cDNAs, sequences from the medaka genome project, and the UniGene database. We found that treatment with FLU, VIN, and 11-KT caused significant (false discovery rate = 0.01) differential expression of at least 87, 82, and 578 genes, respectively. Two sets of responsive genes are associated to vertebrate sex differentiation and growth, and 50 genes were useful in discriminating between ADC classes. The discriminating capacity was confirmed by a remarkable similarity of the antiandrogenic expression profiles of VIN and FLU, which were distinct from the androgenic profile of 11-KT. Interest in the environmental fate of fluorotelomer alcohols (FTOHs) has spurred efforts to understand their equilibrium partitioning behavior. Experimentally determined partition coefficients for FTOHs between soil/water and air/water have been reported, but direct measurements of partition coefficients for dissolved organic carbon (DOC)/water (K(doc)) and octanol/ water(K(ow)) have been lacking. Here we measured the partitioning of 8:2 and 6:2 FTOH between one or more types of DOC and water using enhanced solubility or dialysis bag techniques, and also quantified K(ow) values for 4:2 to 8:2 FTOH using a batch equilibration method. The range in measured log K(doc) values for 8:2 FTOH using the enhanced solubility technique with DOC derived from two soils, two biosolids, and three reference humic acids is 2.00-3.97 with the lowest values obtained for the biosolids and an average across all other DOC sources (biosolid DOC excluded) of 3.54 +/- 0.29. For 6:2 FTOH and Aldrich humic acid, a log K(doc) value of 1.96 +/- 0.45 was measured using the dialysis technique. These average values are approximately 1 to 2 log units lower than previously indirectly estimated K(doc) values. Overall, the affinity for DOC tends to be slightly lower than that for particulate soil organic carbon. Measured log K(ow) values for 4:2 (3.30 +/- 0.04), 6:2 (4.54 +/- 0.01), and 8:2 FTOH (5.58 +/- 0.06) were in good agreement with previously reported estimates. Using relationships between experimentally measured partition coefficients and C-atom chain length, we estimated K(doc) and K(ow) values for shorter and longer chain FTOHs, respectively, that we were unable to measure experimentally. 相似文献

Impact of drip and overhead sprinkler irrigation on the persistence of attenuated Escherichia coli O157:H7 in the lettuce phyllosphere was investigated using a split-plot design in four field trials conducted in the Salinas Valley, California, between summer 2007 and fall 2009. Rifampicin-resistant attenuated E. coli O157:H7 ATCC 700728 (BLS1) was inoculated onto the soil beds after seeding with a backpack sprayer or onto 2- or 4-week-old lettuce plant foliage with a spray bottle at a level of 7 log CFU ml−1. When E. coli O157:H7 was inoculated onto 2-week-old plants, the organism was recovered by enrichment in 1 of 120 or 0 of 240 plants at 21 or 28 days post-inoculation, respectively. For the four trials where inoculum was applied to 4-week-old plants, the population size of E. coli O157:H7 declined rapidly and by day 7, counts were near or below the limit of detection (10 cells per plant) for 82% or more of the samples. However, in 3 out 4 field trials E. coli O157:H7 was still detected in lettuce plants by enrichment 4-weeks post-inoculation. The efficacy levels of practices used by consumers to wash smooth-surface fruits and vegetables were compared. Golden Delicious apples were spot inoculated near the blossom end with 50 microl of a cocktail of six serotypes of Salmonella enterica (with a total inoculum level of approximately 10(9) CFU per apple). The inoculum was dried for 1.5 h, and apples were either treated immediately or held for 24 h prior to treatment. Treatments included wetting with approximately 5 ml of water, vinegar (5% acidity), or a 200-ppm chlorine solution, rubbing for 5 or 30 s, rinsing with 200 to 600 ml of 24 or 43 degrees C water, and drying with a sterile paper towel. Residual populations of Salmonella were determined by rubbing the treated apple for 30 s in 20 ml of Dey-Engley neutralizing broth and plating on tryptic soy agar and bismuth sulfite agar. Rubbing treatments carried out for 5 and 30 s both resulted in a significant reduction in Salmonella populations (1 log10 CFU per apple) relative to populations on samples held for 30 s. A 5-s rub followed by a 200-ml flowing-water rinse reduced populations by 3 log10 CFU per apple. No further decrease in population was obtained by rinsing with 400 or 600 ml of water. Increasing the rinse water temperature to 43 degrees C did not significantly improve microbial removal. Drying the apple with a sterile paper towel resulted in an additional decrease of approximately 0.4 log10 CFU per apple. A reduction of 3.2 log10 CFU was achieved with a combination of wetting with water, rubbing for 5 s, rinsing with 200 ml of water, and drying with a paper towel for apples inoculated just prior to or 24 h before treatment. The potential of mid-infrared (MIR) and near-infrared (NIR) spectroscopy for their ability to differentiate between apple juice samples on the basis of apple variety and applied heat-treatment was evaluated. The heat-treatment involved exposure of juice samples (15 ml) for 30 s in a 900 W microwave oven and the apple varieties used to produce the juice samples were Bramley, Elstar, Golden Delicious and Jonagold. The chemometric procedures applied to the MIR and NIR data were partial least squares regression (PLS1 for differentiation on the basis of heat-treatment, PLS2 for varietal differentiation) and linear discriminant analysis (LDA) applied to principal component (PC) scores. PLS1 and PLS2 gave the highest level of correct classification of the apple juice samples according to heat-treatment (77.2% for both MIR and NIR data) and variety (78.3–100% for MIR data; 82.4–100% for NIR data), respectively. 相似文献

This study investigated the potential of Northern shrimp (Pandelus borealis Kreyer) by-products as a source of omega-3 polyunsaturated fatty acids (ω-3 PUFAs). The by-products (heads, shell and tail) of processing accounted for approximately 50–60% of the catch. Supercritical CO2 extraction (SFE) of the by-products at 35 MPa and 40 °C generated a deep red oil, rich in ω-3 PUFAs, specifically 7.8 ± 0.06% eicosapentaenoic acid (EPA) and 8.0 ± 0.07 % docosahexaenoic acid (DHA). 相似文献

Beef longissimus dorsi muscle samples matured over a 21 day period were analysed using three different analytical techniques; 1H NMR, GC–MS and HPLC. The data from the three experimental techniques were correlated with each other to determine if the results were statistically similar to each other. From our analysis we determined that the metabolites measured using 1H NMR were statistically similar to the compounds quantified using the chromatography techniques (p < 0.001). In addition, using PCA, we were able to show that different metabolites, measured using the various analytical techniques produced very similar scores and loadings plots for all the analysis and extraction techniques undertaken across the 21 day time domain. Using a combination of these three different techniques provides a unique and holistic insight into the biochemistry behind the conversion of muscle to meat which would not be possible using any single technique alone. 相似文献
